【発明の属する技術分野】
本発明は、病原性システインプロテアーゼ、中でもアルギニン特異的システインプロアテーゼ(Rgp)やリジン特異的システインプロアテーゼ(Kgp)の活性を有効に阻害する病原性システインプロテアーゼ活性阻害剤に関する。

【発明の背景】
プロテアーゼとは、タンパク質(protein)を分解する酵素の総称であり、タンパク質のペプチド結合を加水分解反応によって切断し、より小さいペプチド分画やアミノ酸に分解する。そしてこのプロテアーゼは、活性原基によって、1)セリンプロテアーゼ、2)システインプロテアーゼ、3)アスパラギン酸プロテアーゼ、4)メタロプロテアーゼなどに分類される。

システインプロテアーゼは、その活性亢進が様々な疾患に関与していることが知られ、これら疾患において亢進したシステインプロテアーゼを阻害することができれば、疾患の治療・予防に有効である可能性がある(現代化学増刊 プロテアーゼの生体機能、東京化学同人 1993年; プロテアーゼとそのインヒビターなど)。

中でも、アルギニン特異的システインプロアテーゼ(以下「Rgp」という。)及びリジン特異的システインプロアテーゼ(以下「Kgp」という。)は、どちらの酵素も、歯周病原性細菌であるグラム陰性嫌気性細菌Porphyromonas gingivalis(ジンジバリス菌)が産生する主要な酵素として知られているが、歯周病のビルレンス因子としての機能を備えているのみならず、様々な病原性機能を備えている。例えば、Kgp及びRgpは、互いに共同して歯周組織の直接破壊や生体防御系の破壊する病原性機能を有するほか、Kgpは独自に線溶系の亢進などの病原性機能を発揮し、Rgpは独自に血液凝固系の亢進などの病原性機能を発揮することが知られている(化学と生物、vol.40、No.2、72−74、2002)。よって、これらの病原性機能を阻害することができれば、様々な疾患の治療・予防を期待することができる。

なお、Rgpは、アルギニン残基を特異的に認識してそのC末端側で切断するArg−ジンジパインであり、Kgpは、リジン残基を特異的に認識してそのC末端側で切断するLys−ジンジパインである。

【従来の技術】
病原性システインプロテアーゼ、中でもKgpやRgpの活性を阻害する物質としては、KYT−1がCbz−Lys−Arg−Co−Lys−NMe2(Cbz:カルボベンジルオキシ,Me:メチル)の構造を有し、Rgpを特異的に阻害し、また、KYT−36はCbz−Glu(NHNMePh)− Lys−CO−NHCH2Phの構造を有し、Kgpを特異的に阻害することが知られている(非特許文献1参照)。

また、特許文献1記載の発明は、緑茶抽出物を含む各種天然物の抽出物についてプロテアーゼ阻害効果を検討しており、その実施例1では、各種天然物の抽出物の水溶液にプロテアーゼを産生する嫌気性細菌であるPorphyromonas gingivalis菌体懸濁液を加えて攪拌し、35℃、5分間インキュベートし、これにカゼイン溶液を加え、35℃、10分間インキュベートし、その後、トリクロロ酢酸溶液を加えて反応を停止し、フォリン試薬を加え発色させた後660nmの吸光度を測定し、予め作製した検量線よりプロテアーゼの力価を求めており、その結果、緑茶抽出物のプロテアーゼ阻害率100%という結果を示す一方、その実施例2では、カテキンのプロテアーゼ阻害率0%という結果を示している。

【発明が解決しようとする課題】
本発明は、天然物である茶の抽出成分の病原性システインプロテアーゼ阻害作用、特にKgpやRgpに対する活性阻害作用について鋭意研究し、茶の抽出成分に由来し、特にKgpやRgpの活性を阻害する作用に優れた病原性システインプロテアーゼ活性阻害剤を提供せんとするものである。

【課題を解決するための手段】
本発明は、カテキン又はカテキン混合物を有効成分とする病原性システインプロテアーゼ活性阻害剤を提案する。

カテキンとしては、(−)エピカテキン(EC)、(−)エピガロカテキン(EGC)、(−)エピカテキンガレート(ECg)、(−)エピガロカテキンガレート(EGCg)、(−)カテキン(C)、(−)ガロカテキン(GC)、(−)カテキンガレート(Cg)及び(−)ガロカテキンガレート(GCg)のいずれかを用いることができ、カテキン混合物としては、前記カテキンのうちの二種類以上の組合わせからなる混合物を用いることができる。

カテキン又はカテキン混合物を有効成分とする病原性システインプロテアーゼ活性阻害剤は、病原性システインプロテアーゼの中でもRgp及びKgpの活性を有効に阻害する。中でも特に、GC及びGCgのいずれか或いはこれらの混合物は、他のカテキンに比べてRgpに対する活性阻害効果がより一層優れており、EGC、EGCg、GC、GCgのいずれか、或いはこれらのうちの二種類以上の組合わせからなる混合物は、他のカテキンに比べてKgpに対する活性阻害効果がより一層優れている。
よって、本発明はまた、GC及びGCgのいずれか、或いはこれらの混合物を有効成分とするRgp活性阻害剤、並びに、EGC、EGCg、GC、GCgのいずれか、或いはこれらのうちの二種類以上の組合わせからなる混合物を有効成分とするKgp活性阻害剤を提案するものである。

なお、本発明において「特定のカテキン(例えば(−)エピカテキン(EC)或いは(−)エピガロカテキン(EGC)、或いはこれらの混合物など、限定されたカテキン)を有効成分とする」とは、当該特定のカテキンの病原性システインプロテアーゼ活性阻害効果を阻害しなければ、その他の成分、例えば当該特定のカテキン以外のカテキンなどの成分を含んでいてもよいという意を包含する。
また、EC、EGC、ECg、EGCg、C、GC、Cg及びGCgそれぞれのカテキンの重合体、並びに、当該それぞれのカテキンと他のカテキンとの共重合体、或いはそれらの二種類以上の組合わせからなる混合物は、「カテキン又はカテキン混合物」「EC、EGC、ECg、EGCg、C、GC、Cg及びGCgのいずれか、或いはこれらのうちの二種類以上の組合わせからなる混合物」の均等物であると考えることができる。

【発明の実施の形態】
本発明の病原性システインプロテアーゼ活性阻害剤は、カテキン又はカテキン混合物を適宜濃度で配合することによって製造することができる。但し、カテキン又はカテキン混合物の薬理活性を阻害しない範囲でその他の成分を配合したり、その他の処理を施すことは適宜可能である。

上記のカテキンとしては、EC、EGC、ECg及びEGCg、これらのエピマーであるC、GC、Cg及び GCgの8種類のカテキンのいずれかを用いることができる。
また、上記のカテキン混合物としては、上記8種類のカテキンのうちの二種類以上の組合わせからなる混合物を用いることができる。
8種類全てのカテキンに病原性システインプロテアーゼ、中でもRgp及びKgpに対する活性阻害効果が認められているが、各カテキンの活性阻害能には大きな差が見られる。
例えば、Rgpに対する活性阻害効果は、効果の大きい順に
GC、GCg>EGCg、EGC>Cg、ECg>EC、C
であり、
Kgpに対する活性阻害効果は、効果の大きい順に
GC、GCg、EGCg、EGC>Cg、ECg>EC、C
であることが確かめられている。
(ちなみに、上記の「GC、GCg>EGCg、EGC」という表示は、GCとGCgの効果は同程度であり、かつ、GCやGCgの効果は、EGCgやEGCの効果に比べて大きいという意を表しており、本明細書における同様の表示は同様の意を有している。)

EC、EGC、ECg及びEGCgは、天然物、特に茶葉中に多く含有されているから、従来公知の方法或いは今後公知となる方法によって抽出及び精製などによって得ることができる。

他方、これらのエピマーであるC、GC、Cg及び GCgは、茶葉を含めて天然植物中にはほとんど存在しないが、EC、EGC、ECg及びEGCgのそれぞれ或いはこれら二種類以上の組合わせからなる混合物を加熱処理することにより熱異性化(エピマー化)させて得ることができる。例えば、精製したEC、EGC、ECg及びEGCgのそれぞれ或いはこれら二種類以上の組合わせからなる混合物、又は、茶の抽出液或いは浸出液を、好ましくはpH5〜6に調整した上で、80℃以上の加熱処理することによりカテキンの熱異性化を促してC、GC、Cg及び GCgの含有濃度を高め、このような加熱処理後物からC、GC、Cg及び GCgのそれぞれ或いはこれら二種類以上の組合わせからなる混合物を分離・精製して得ることができる。
カテキンを熱異性化(エピマー化)させるための加熱処理としては、カテキンを含有する溶液(カテキン溶液)を少なくとも80℃以上に加熱処理すればよい。例えば、100℃×15分加熱、115℃×20分加熱、120℃×3〜30分加熱、123℃×10分加熱、131℃×30秒加熱、133℃×45秒のいずれにおいても、カテキン類の熱異性化が認められている。
カテキン溶液をpH5〜6に調製した上で加熱処理するのが好ましいとするのは、pH4.5以下ではカテキン類はほとんど熱異性化しないことが報告されているからである(末松伸一ら、:日食工誌、39,178(1992))。

なお、カテキンの分離・精製方法は、従来公知の方法或いは今後公知となる方法によって行えばよい。例えば、茶の抽出液を、例えば水−アセトニトリル−リン酸の混合液を移動相とした逆相HPLCにかけ、アセトニトリル濃度でグラジエントをかけることによってそれぞれ分離できることが知られている。

本発明の「カテキン混合物」として、カテキン濃度20重量%以上、好ましくは25%以上、中でも特に30%以上の茶抽出物を用いることができる。
茶抽出物は、茶を水、温水または熱水、好ましくは40℃〜100℃の温熱水、中でも90〜100℃の熱水、或いは人体に無害なエタノール水溶液またはエタノールなどの有機溶媒で抽出して得ることができ、好ましくは、この茶抽出物を溶媒抽出法、樹脂吸着法、限外濾過・逆浸透濾過等の濾過などの精製手段によってカテキンの含有量を高める方向に精製して得ることができる。抽出の際、カテキン含有量を高めるべく塩酸等を添加した酸性条件下で抽出を行ってもよい。また、市販の茶抽出物を用いることがもできる。例えば、テアフラン35及び30A(商品名;伊藤園社製)のいずれも、緑茶を熱水抽出処理し、この抽出物を乾燥させてカテキン濃度を約30%とした緑茶抽出物(組成(重量%):EGC7.62、EGCg13.70、EC2.13、ECg2.61、GC2.61、GCg0.49、(+)C0.43、Cg0.28)であり、テアフラン90S(商品名;伊藤園社製)は、緑茶を熱水抽出処理して得た抽出物を、水と低・高濃度アルコールを使って吸着カラムにて分離し乾燥させ、茶ポリフェノール濃度を約85〜99.5%とした緑茶抽出物(組成(重量%):EGCg50.18、EC0.57、ECg17.11、GCg3.96、Cg0.91)である。

本発明の病原性システインプロテアーゼ活性阻害剤において、カテキン又はカテキン混合物は、それぞれ単独で本発明の有効成分として配合することも可能であるが、既に或いは将来的に病原性システインプロテアーゼ、中でもRgp及びKgpに対する活性阻害効果が認められた物質と混合し、これらの有効成分と併用することも可能である。
なお、単独で本発明の有効成分として配合する場合、例えばカテキン又はカテキン混合物を、精製水や生理食塩水などに溶解して病原性システインプロテアーゼ活性阻害剤(例えば経口投与剤、腹腔内投与剤等)とすることも可能である。

本発明の病原性システインプロテアーゼ活性阻害剤は、経口投与剤または非経口投与剤(筋肉注射、静脈注射、皮下投与、直腸投与、経皮投与、経鼻投与など)とすることができ、それぞれの投与に適した配合及び剤型とするのが好ましい。
剤型について言えば、例えば経口投与剤用としては液剤、錠剤、散剤、顆粒、糖衣錠、カプセル、懸濁液、乳剤、丸剤などの形態に調製することができ、非経口投与剤用としては注射剤、アンプル剤、直腸投与剤、油脂性坐剤、水溶性坐剤などの形態に調製することができる。
配合(製剤)について言えば、通常用いられている賦形剤、増量剤、結合剤、湿潤化剤、崩壊剤、表面活性剤、潤滑剤、分散剤、緩衝剤、保存剤、溶解補助剤、防腐剤、矯味矯臭剤、無痛化剤、安定化剤などを用いて常法により製造することができる。また、例えば乳糖、果糖、ブドウ糖、でん粉、ゼラチン、炭酸マグネシウム、合成ケイ酸マグネシウム、タルク、ステアリン酸マグネシウム、メチルセルロース、カルボキシメチルセルロースまたはその塩、アラビアゴム、ポリエチレングリコール、シロップ、ワセリン、グリセリン、エタノール、プロピレングリコール、クエン酸、塩化ナトリウム、亜硫酸ソーダ、リン酸ナトリウムなどの無毒性の添加剤を配合することも可能である。

また、本発明の病原性システインプロテアーゼ活性阻害剤は、医薬品のほか、医薬部外品、薬理効果を備えた健康食品・健康飲料・特定保健用食品・機能性食品、食品添加剤、その他ヒト以外の動物に対する薬剤や餌、餌用添加剤などとして提供することが可能である。
例えば、医薬部外品として調製し、これを瓶ドリンク飲料等の飲用形態、或いはタブレット、カプセル、顆粒等の形態とすることにより、より一層摂取し易くすることができる。

病原性システインプロテアーゼ活性阻害効果を備えた健康食品・健康飲料・特定保健用食品・機能性食品としては、例えば、カテキン又はカテキン混合物、或いはこの混合物を含むカテキン溶液(例えばカテキン抽出液や浸出液)などを、炭酸、賦形剤(造粒剤含む)、希釈剤、或いは更に甘味剤、フレーバー、小麦粉、でんぷん、糖、油脂類等の各種タンパク質、糖質原料やビタミン、ミネラルなどの飲食品材料群から選ばれた一種或いは二種以上と混合したり、或いは、現在公知の飲食品、例えばスポーツ飲料、果実飲料、乳飲料、茶飲料、野菜ジュース、乳性飲料、アルコール飲料、ゼリー、ゼリー飲料、炭酸飲料、チューインガム、チョコレート、キャンディ、ビスケット、スナック、パン、乳製品、魚肉練り製品、畜肉製品、冷菓、乾燥食品、サプリメントなどに添加して製造することができる。

本発明における有効成分の含有量は、使用方法によっても異なるが、医薬品であれば、カテキン或いはこれらの混合物を、乾燥重量換算で0.001〜1重量%、特に0.01〜0.5重量%配合するのが好ましく、 飲食品であれば、カテキン或いはこれらの混合物を0.001〜1重量%、特に0.01〜0.5重量%配合するのが好ましい。
なお、摂取量としては、カテキン或いはこれらの混合物を乾燥重量換算で大人一日に10〜5000mg、好ましくは100〜1500mg程度摂取するのが好ましい。

<試験1>
(細胞の培養)
黒色コロニー形成嫌気性菌は、5%緬羊脱繊維素血液(日本バイオテスト研究所)添加ブルセラHK寒天培地(極東)を用い、嫌気性菌培養装置MIP−1025(Forma Scientific)にて37℃、N280%、CO210%、H210%の条件で培養し菌株を維持した。実験に当たり、ブレイン−ハートインフージョンブロスに0.5%イーストエキス、0.05%システイン塩酸塩、5μg/mLヘミン1μg/mL、メナジオン添加培地で上記の培養条件で培養した。本研究ではPrevotella intermedia ATCC25611T, Prevotella nigrescens ATCC33563T,ATCC25261およびPorphyromonas gingivalis ATCC33277Tの菌株を使用した。

(細胞抽出液の調整)
培養された細菌は遠心分離後、リン酸緩衝食塩液(pH7.2)で2度洗浄し、1%Triton−Xを含むTris−HCL緩衝液(pH7.4)に懸濁し、ガラスビーズ(粒子径0.15mm)を加え、激しく攪拌して破砕した。破砕液を12,000回転、20分の遠心後、上清を酵素活性測定に使用した。

(アルカリホスファターゼのザイモグラフ)
細胞抽出液は7.5%ポリアクリルアミドゲルと通常用いるトリス・グリシン泳動用緩衝液を用いて電気泳動し、泳動したゲルを1mg/mLナフト−ルAS−BIホスフェート、0.1mg/mLファーストブルーBBをトリス塩酸緩衝液(0.1M Tris−HCL buffer(pH10.0) 、5μmMgCL2含む)で溶解した液中で反応させ、酵素活性を検出した。

(アルカリホスファターゼ活性の測定)
アルカリホスファターゼ活性は、パラニトロホスフェート(pNP)を基質とし、96穴マルチウェル中で反応させ、波長405nmの吸光度を測定した。すなわち、アルカリホスファターゼ活性の測定は、0.1mM MgCL2、8mM pNPを含む0.1Mトリス緩衝液(0.1M Tris−HCL buffer(pH10.0) 、5μmMgCL2含む)80μL、種々の濃度のEGCg10μL、及び粗酵素抽出液10μLを攪拌し、37℃、20分反応させた。

(Rgp活性の測定)
Rgp活性は、Benzoyl arginine p−nitroanilide (BapNA)を基質とし、96穴マルチウェル中で反応させ、波長405nmの吸光度を測定した。すなわち、10mM Benzoyl arginine p−nitroanilide (HCL)をDMSOに溶解して基質ストック溶液とし、0.1Mトリス塩酸緩衝液(pH7.4)中に1mMシステイン塩酸塩を溶解する一方、上記基質ストック溶液を1/10 buffer2で10倍に希釈、溶解した後、基質ストック溶液80μL(最終基質濃度0.8μM)、種々濃度のEGCg10μL、及び粗酵素抽出液10μLとなるように攪拌し、37℃、10分反応させた。

(Kgp活性の測定)
Kgp活性は、N−p−tosyl−Gly−Lys−p−nitroanilideを基質とし、96穴マルチウェル中で反応させ、波長405nmの吸光度を測定した。すなわち、10mMN−p−tosyl−Gly−Lys−p−nitroanilide (HCL)をDMSOに溶解して基質ストック溶液とし、0.1Mトリス塩酸緩衝液(pH7.4)中に1mMシステイン塩酸塩を溶解する一方、上記基質ストック溶液を1/10 buffer2で50倍に希釈、溶解した後、基質ストック溶液80μL(最終基質濃度0.16μM)、種々濃度のEGCg10μL、及び粗酵素抽出液10μLとなるように攪拌し、37℃、10分反応させた。

(溶血活性および血球凝集活性の測定)
溶血活性の測定は、10%ウサギ赤血球および培養菌体を96穴マルチウェル中で混合し、37℃で適宜反応させ、反応液25μLを10倍量のNCN緩衝液(3mMsodium citrate,150mMsodium chloride,pH6.8)に懸濁させ、遠心後上清の波長414nmの吸光度を測定し、遊離するヘモグロビン量とした。
溶血の程度は、[(EGCgとP.intermediaを加えたサンプル)−(EGCgのみ加えたサンプル)]/[(100%溶血させたサンプル)−(blank)]×100(%)として表した。
血球凝集活性の測定は、等量の2%ウサギ赤血球と培養菌体および各濃度のEGCgをV底マルチウェルに加え、30分間攪拌した後、冷蔵庫にて一夜保存し、凝集を起す最高希釈倍率から凝集価を求めた。

(結果および考察)
1)アルカリホスファターゼのザイモグラフ
黒色コロニー形成嫌気性菌の産生するアルカリホスファターゼのザイモグラフを図1に示す。
P.intermediaの移動度は最も少なく、次いでP.nigrescensが、P.gingivalisは最も移動度が大きく、細菌の種により異なる種類のアルカリホスファターゼを産生することが考えられた。
表1は各酵素に対するバナジン酸化合物の効果を示した。バンジン酸化合物はPrevotellaのアルカリホスファターゼ活性は強く阻害するが、P.gingivalisのアルカリホスファターゼ活性は阻害しなかった。

2)EGCgのアルカリホスファターゼに及ぼす影響
EGCgはP.intermediaの産生するアルカリホスファターゼを強く阻害し、酵素活性を50%阻害する濃度は約5μg/mLであった(図2)。
P.intermediaの培養上清にも本酵素は検出され、EGCgの50%阻害する濃度はP.nigrescensのアルカリホスファターゼとほぼ同程度で約50μg/mLであった。
しかし、P.gingivalisの産生するアルカリホスファターゼ活性に対しては阻害効果を示さなかった。

3)EGCgのP.intermediaの溶血活性および血球凝集活性に及ぼす影響
図3は、最終濃度0−100μg/mLのEGCgを反応系に加え、2、4および6時間後の溶血活性を示したものである。
EGCgの濃度が増加するにつれP.intermediaの溶血活性は阻害され、100μg/mL、6時間の反応では対照の溶血活性の23%に減少した。

表2は、EGCgの黒色コロニー形成嫌気性菌の血球凝集活性に及ぼす影響を調べたものである。
3.6μg/mLのEGCgを反応系に加え、細菌と血球のみ加えた対照と比較した。その結果、P.intermediaは対照の2分の1、P.nigrescensでは対照の8分の1に凝集活性を減少した。一方、EGCgはP.gingivalisの血球凝集活性には影響を及ぼさなかった。
黒色コロニー形成嫌気性菌は溶血活性、血球凝集活性を持ち、これらの細菌の育成に必要な鉄イオンを血球凝集、溶血という形で補給していることが示唆されている。EGCgがこれらの活性を阻害することは、これらの細菌の育成を抑制し、種々の有害な代謝産物産生を抑制する可能性が示唆される。

4)EGCgのP.gingivalisの産生するRgp活性に及ぼす影響
EGCgはP.gingivalisの産生するRgp活性を強く阻害し、酵素活性を50%阻害する濃度は約10μg/mLであった(図4)。
生体蛋白質の分解や、多形核白血球の殺菌活性の抑制、さらには菌の生長などにP.gingivalisのRgpやリジンジンジパイン(Kgp)らの酵素が重要な役割を演じていることから、EGCgはこれらの病原性効果を阻害できることが示唆される。

また、EGCg以外のカテキンすなわちEC、EGC、ECg、C、GC、Cg 、GCg及びGA(表ではg)、さらにカテキン混合物としてのテアフラン35(商品名;伊藤園社製)及びテアフラン90S(商品名;伊藤園社製)についても、EGCgと同様にRgp活性に及ぼす影響について試験し、各カテキンのRgp活性阻害率に関する結果を図5〜図7に示し、各カテキンの50%阻害濃度を下記表3に示した。
