本発明は、新規なユニットを含むポリヒドロキシアルカノエート共重合体と、微生物を利用するその製造方法に関し、より具体的には、構成ユニットとして、末端にエステル構造を有する3−ヒドロキシ−アルカン酸ユニット、及びフェニル構造、チエニル構造、シクロヘキシル構造のいずれかの構造を有する残基を含む3−ヒドロキシアルカン酸ユニットとを同時に分子中に含むポリヒドロキシアルカノエート共重合体と、ジカルボン酸モノエステル化合物と、フェニル構造、チエニル構造、シクロヘキシル構造のいずれかの構造を有する残基を含むアルカン酸とを原料として、当該ポリヒドロキシアルカノエート共重合体の産生能を有する微生物を利用し、構成ユニットとして、末端にエステル構造を有する3−ヒドロキシーアルカン酸ユニット、及びフェニル構造、チエニル構造、シクロヘキシル構造のいずれかの構造を有する残基を含む3−ヒドロキシアルカン酸ユニットとを同時に分子中に含むポリヒドロキシアルカノエート共重合体の製造方法に関する。

【従来の技術】
これまで、多くの微生物が、ポリ−3−ヒドロキシ酪酸(PHB)あるいはその他のポリ−3−ヒドロキシアルカノエート(PHA)を生産し、その菌体内に蓄積することが報告されている(「生分解性プラスチックハンドブック」,生分解性プラスチック研究会編,(株)エヌ・ティー・エス,P178−197(1995))。これらPHAなどの微生物が産生するポリマーは、従来のプラスチックと同様に、溶融加工等により各種製品の生産に利用することができる。さらに、微生物が産生するポリマー、例えば、PHAなどは、生分解性を有しており、自然界の微生物により完全分解されるという利点を有している。従って、例えば、微生物が産生するPHAは、廃棄した際、従来の多くの合成高分子化合物のように自然環境にそのまま残留し、汚染を引き起こす要因となることがない。また、微生物が産生するPHAは、一般に生体適合性にも優れており、医療用軟質部材等としての応用も期待されている。

この微生物産生PHAは、その生産に用いる微生物の種類、ならびに、培地組成、培養条件等により、様々な組成や構造のものとなり得ることも知られている。これまで、主にPHAの物性の改良という観点から、微生物産生PHAの組成や構造の制御を試みる研究がなされてきた。

微生物産生PHAの組成や構造の制御を目的とする研究の一つとして、近年、ユニット中に芳香環を有するPHAを微生物に生産させる研究が盛んになされている。

Makromol.Chem.,191,1957−1965(1990)及びMacromolecules,24,5256−5260(1991)には、5−フェニル吉草酸を基質として、シュードモナス オレオボランス(Pseudomonas oleovorans)が3−ヒドロキシ−5−フェニル吉草酸をユニットとして含むPHAを生産することが報告されている。

Macromolecules,29,1762−1766(1996)には、5−(p−トリル)吉草酸を基質として、シュードモナス オレオボランス(Pseudomonas oleovorans)が3−ヒドロキシ−5−(p−トリル)吉草酸ユニットを含むPHAを生産することが報告されている。

Macromolecules,32,2889−2895(1999)には、5−(2,4−ジニトロフェニル)吉草酸を基質として、シュードモナス オレオボランス(Pseudomonas oleovorans)が3−ヒドロキシ−5−(2,4−ジニトロフェニル)吉草酸ユニット、及び3−ヒドロキシ−5−(p−ニトロフェニル)吉草酸ユニットを含むPHAを生産することが報告されている。

Macromol.Chem.Phys.,195,1665−1672(1994)には、11−フェノキシウンデカン酸を基質として、シュードモナスオレオボランス(Pseudomonas oleovorans)が3−ヒドロキシ−5−フェノキシ吉草酸ユニットと3−ヒドロキシ−9−フェノキシノナン酸ユニットを含むPHAコポリマーを生産することが報告されている。

特許掲載公報第2989175号には、3−ヒドロキシ−5−(モノフルオロフェノキシ)ペンタノエート(3H5(MFP)P)ユニット、あるいは3−ヒドロキシ−5−(ジフルオロフェノキシ)ペンタノエート(3H5(DFP)P)ユニットからなるホモポリマー;少なくとも、3H5(MFP)Pユニットあるいは3H5(DFP)Pユニットを含有するコポリマー;これらのポリマーの産生能を有するシュードモナス・プチダ;シュードモナス属を用いた、前記のポリマーの製造法に関する発明が開示されている。加えて、その発明の効果として、置換基を有する長鎖脂肪酸を資化して、側鎖末端に、1から2個のフッ素原子が置換したフェノキシ基をもつポリマーを合成することができ、また、かかるポリマーは、融点が高い上、良い加工性を保持しつつ、加えて、立体規則性、撥水性を与えることができる点を記載している。

このユニット中の芳香環上にフッ素置換を有するフッ素置換PHA以外に、ユニット中の芳香環上にシアノ基やニトロ基が置換したPHAの研究もなされている。

Can.J.Microbiol.,41,32−43(1995)及びPolymer International,39,205−213(1996)には、シュードモナス オレオボランス(Pseudomonas oleovorans)ATCC29347株及びシュードモナス プチダ(Pseudomonas putida)KT2442株を用いて、オクタン酸と6−(p−シアノフェノキシ)ヘキサン酸あるいは6−(p−ニトロフェノキシ)ヘキサン酸を基質として、3−ヒドロキシ−6−(p−シアノフェノキシ)ヘキサン酸あるいは3−ヒドロキシ−6−(p−ニトロフェノキシ)ヘキサン酸をモノマーユニットとして含むPHAの生産が報告されている。

これら環状に置換基を持つ芳香環を有するユニットを含むPHAは、融点が高く、加工性も良いという、芳香環に由来するポリマー性状を維持しつつ、芳香環上に存在している置換基に由来する新たな機能も付与された、多機能のPHAとなる。

また、その一方で、ユニット中にエステル基を有するPHAを基に、多機能のPHAを得ることを目的とした研究も盛んに行われている。

Macromol.Chem.Phys.,195,1405−1421(1994)には、ポリヒドロキシアルカノエート生産微生物としてシュードモナス オレオボランス(Pseudomonas oleovorans)を用いて、アルカン酸エステルを原料として側鎖にエステル基を有するユニットを含むポリヒドロキシアルカノエートの生産が生産されたと報告されている。

また、University of Massachusetts Ph.D.Dissertation Order Number 9132875(1991)には、同じくシュードモナス オレオボランスを生産微生物として、側鎖にベンジルエステル構造を有するユニットを含むポリヒドロキシアルカノエートが生産されたと報告されている。

その他、側鎖にエステル基を有するユニットを含むポリヒドロキシアルカノエートの製造方法としては、ポリリンゴ酸誘導体の報告が多数ある。

米国特許第4,265,247号には、エステル基を有するラクトン化合物を開環重合による、側鎖にエステル基を有するポリヒドロキシアルカノエートの製造方法が開示されている。

また、Int.J.Biol.Macromol.,25,255−264(1999)には、側鎖にベンジルエステル構造及びアンモニウム塩構造を有するポリヒドロキシアルカノエートを、やはりラクトン化合物の開環重合によりの製造する方法が報告されている。

【発明が解決しようとする課題】
この様な側鎖のエステル構造は、水素添加反応によりカルボキシル基に変換することが可能であり、その後の化学修飾等で様々な物理化学的性質を有するポリヒドロキシアルカノエートを創製することが可能であるが、ガラス転移温度が低い等の問題点があり、また、ポリマー自身の疎水性等を制御することも困難であり、この様な原因がこの種のポリマーの応用範囲を制限していた。この様なことから、ユニット中にエステル基を有するPHAであって、ポリマーとしての応用範囲を広げ得るような、熱的に安定な、更には物性を任意に制御し得るようなPHAそのもの、及びその製造方法が求められていた。

【発明の実施の形態】
本発明のポリヒドロキシアルカノエート共重合体の製造方法における微生物の培養条件の詳細は、以下のとおりである。

リン酸緩衝液及びアンモニウム塩或いは硝酸塩を基本とした無機塩培地に、以下に示すように種々の必要基質及び栄養素を加える。

目的とするポリヒドロキシアルカノエートを生産するための基質として前記化学式(15)で示すジカルボン酸モノエステル化合物、及び前記化学式(16)で示す化合物を培地あたりそれぞれ0.01%から1%(w/v)、更に好ましくは0.02%から0.2%の割合で含有していることが望ましい。

微生物増殖のための炭素源及び窒素源、ポリヒドロキシアルカノエート生産のためのエネルギー供給源として加える上記の共存基質濃度は、通常培地あたり0.1%から5%(w/v)、更に好ましくは0.2%から2%の割合で含有していることが望ましい。

本発明で用いる培地としては、リン酸塩及びアンモニウム塩或いは硝酸塩等の窒素源を含む無機塩培地ならいかなる培地でも良いが、窒素源の濃度を調節することでPHAの生産性を向上せしめることが可能である。

培養温度としては上記の菌株が良好に増殖可能な温度であれば良く、15℃から37℃、更に好ましくは20℃から30℃程度が適当である。

培養は液体培養、固体培養等該微生物が増殖し、PHAを生産する培養方法ならいかなる培養方法でも用いることができる。さらに、バッチ培養、フェドバッチ培養、半連続培養、連続培養等の種類も問わない。液体バッチ培養の形態としては、振とうフラスコによって振とうさせて酸素を供給する方法、ジャーファーメンターによる攪拌通気方式の酸素供給方法がある。

微生物にPHAを生産・蓄積せしめる方法としては、上に示した方法の他に、一旦十分に増殖させて後に、塩化アンモニウムのような窒素源を制限した培地へ菌体を移し、目的ユニットの基質となる化合物を加えた状態で更に培養すると生産性が向上する場合がある。

更に本発明の製造方法においては、上記のような条件下で前記微生物を培養し、前記微生物が産生した前記化学式(1)で示す3−ヒドロキシ−ジカルボン酸モノエステル化合物ユニット、及び前記化学式(2)で示すユニットを同時に分子中に含むポリヒドロキシアルカノエート共重合体を微生物細胞から回収する工程を有することを特徴とするポリヒドロキシアルカノエート共重合体の製造方法とすることができる。

微生物細胞から目的のPHAを回収する方法としては、通常行なわれている方法を適用することができる。例えば、クロロホルム、ジクロロメタン、アセトンなどの有機溶媒による抽出が最も簡便ではあるが、それ以外にジオキサン、テトラヒドロフラン、アセトニトリルが用いられる場合もある。また、有機溶媒が使用しにくい環境中においては、SDS等の界面活性剤による処理、リゾチーム等の酵素による処理、次亜塩素酸塩、アンモニア、EDTA等の薬剤による処理、或いは超音波破砕法、ホモジナイザー法、圧力破砕法、ビーズ衝撃法、摩砕法、擂潰法、凍結融解法のいずれかの方法を用いて微生物細胞を物理的に破砕することによって、PHA以外の菌体成分を除去して、PHAを回収する方法を用いることもできる。

本発明の製造方法で用いる前記微生物としては、前記条件を満たす能力を有する微生物であれば如何なる微生物でも良いが、その中でも特にシュードモナス(Pseudomonas)属に属する微生物が望ましく、更に詳しくはシュードモナス チコリアイ(Pseudomonas cichorii)、シュードモナス プチダ(Pseudomonas putida)、シュードモナス フルオレセンス(Pseudomonas fluorecense)、シュードモナス オレオボランス(Pseudomonas oleovorans)、シュードモナス アルギノーサ(Pseudomonas aeruginosa)、シュードモナス スツッツェリ(Pseudomonas stutzeri)、シュードモナス ジェッセニイ(Pseudomonas jessenii)等が望ましい。更に詳しくは、さらに詳しくは、シュードモナス チコリアイ YN2株(Pseudomonas cichorii YN2;FERM BP−7375)、シュードモナス チコリアイ H45株(Pseudomonas cichorii H45、FERM BP−7374)、シュードモナス ジェッセニイ P161株(Pseudomonas jessenii P161、FERM BP−7376)、シュードモナス プチダ P91株(Pseudomonas putida P91、FERM BP−7373)が挙げられる。これら4種の微生物は独立行政法人 産業技術総合研究所 特許生物寄託センターに寄託されており、特開2001−288256号公報にされている微生物である。

【実施例】
[実施例1]
前記M9培地1000mLに、ポリペプトン(和光純薬)5.0g、及び5−フェニル吉草酸、セバシン酸モノメチルエステルをそれぞれ最終濃度4mm、1mmになるよう添加し、2000mL容振とうフラスコに入れてオートクレーブにより滅菌した後室温まで冷却した。調製した培地中に、予めポリペプトン0.5%を含むM9培地で30℃、8時間振とう培養したシュードモナス チコリアイ YN2株の培養液を5mL加え、30℃、40時間培養した。培養後、遠心分離により菌体を収穫し、メタノールで洗浄した後凍結乾燥した。乾燥菌体を秤量後、クロロホルムを加え、50℃で48時間攪拌することによりポリマーを抽出した。ポリマーが抽出されたクロロホルムをろ過し、エバポレーターにより濃縮した後、冷メタノールで沈殿固化した部分を集め、減圧乾燥して、目的とするポリマーを得た。

得られたポリマーの構造決定は、以下のメチル化分解−GC/MS法を用いて行った。即ち、ポリマー5mgを2mLのクロロホルムに溶解し、更に2mLのメタノール−3%硫酸溶液を加え、100℃で3時間半還流反応を行った。反応終了後室温まで冷却し、脱イオン水を10mL加えて攪拌分液し、有機層を硫酸マグネシウム(無水)で脱水した後、反応液をガスクロマトグラフィー−質量分析装置(GC/MS;ShimadzuQP−5050A:カラム:DB−WAXETR 0.32mm×30m)で測定を行った。そのトータルイオンクロマトグラム(TIC)を図1に示す。メインピークとして35.6分、38.0分、45.8分の3本が見られた。35.6分のピークのマススペクトル(MS)を図2に、38.0分のピークのMSを図3に、45.8分のピークのMSを図4にそれぞれ示す。

The structure of the obtained polymer was determined by the following methylation-decomposition-GC / MS method. That is, 5 mg of the polymer was dissolved in 2 mL of chloroform, 2 mL of a methanol-3% sulfuric acid solution was further added, and a reflux reaction was performed at 100 ° C. for 3.5 hours. According to the method of the present invention, a novel polyhydroxyalkanoate copolymer, a unit having an ester structure in a side chain and a unit having a residue having a phenyl structure, a thiophene structure, and a cyclohexyl structure in a side chain are included in a molecule. A polyhydroxyalkanoate copolymer containing the same and a method for producing the same are provided.
