【発明の属する技術分野】本発明は、特定のペプチドに
より、う蝕誘発細菌の1つであるミュータンス連鎖球菌
の口腔内への定着を特異的に抑制し、う蝕を予防あるい
は治療することができるう蝕原因菌付着防止剤に関す
る。
【0002】
【従来の技術及び発明が解決しようとする課題】う蝕
は、ミュータンス連鎖球菌を中心とした病原性細菌が口
腔内に付着、定着し、これらの細菌の産生する酸がエナ
メル質を脱灰して引き起こされる。そのため、う蝕予防
には、殺菌剤による原因菌の除去が有効的である。しか
し、殺菌剤は非特異的に細菌に作用し、口腔内の細菌叢
をむやみに破壊することへの抵抗感から、原因菌である
ミュータンス連鎖球菌のみを特異的に排除する方法が望
まれ、例えば、ミュータンス連鎖球菌を動物又は家禽類
に免疫して得られた抗体などが提案されている。しか
し、抗体は高分子化合物であり、歯垢内部に生息する病
原性細菌に対しては作用し難く、より効果の高い特異的
付着抑制剤の開発が望まれる。

【課題を解決するための手段及び発明の実施の形態】本
発明は、上記要望に応えるためになされたもので、ミュ
ータンス連鎖球菌に対し特異的な細菌付着防止剤につい
て種々検討したところ、N末端にLYP残基、C末端に
YTF残基を有するアミノ酸残基数15以下のペプチド
が、ミュータンス連鎖球菌の口腔内への付着を特異的に
防止することを見いだし、本発明を完成するに至った。
【0004】以下、本発明につき詳しく説明する。
【0005】本発明のう蝕原因菌付着防止剤は、N末端
にLYP残基及び/又はC末端にYTF残基を有するア
ミノ酸残基数15以下、好ましくは5〜9のペプチド及
びその誘導体の少なくとも1つを有効成分とするもので
ある。
【0006】なお、本明細書で用いるペプチドの構成ア
ミノ酸およびその保護基等についての略号は、ペプチド
の分野で通常用いられるものである。例えば、Lはロイ
シン、Yはチロシン、Pはプロリン、Qはグルタミン、
Tはトレオニン、Fはフェニルアラニンを表す。
【0007】ミュータンス菌は菌表層に特異的な付着因
子(PAc、P1、SAI/IIなどと呼ばれる)をも
ち、歯面上に形成されるペリクルを構成する唾液成分と
特異的に結合することが知られる。そこで、この特異的
付着因子の活性部位ペプチドにより、ミュータンス菌特
異的付着を拮抗的に抑制する方法が提案されている(N
ature Biotechnology 17,42
−47,1999)。一方、スタセリンは、43残基の
アミノ酸からなる唾液タンパクで、唾液中のリン酸カル
シウムの析出抑制効果などが報告されているが、ミュー
タンス菌の付着因子としては機能しないと考えられてい
る。しかし、意外なことにこのスタセリンのC末端(2
9−43残基)側にミュータンス菌の特異的付着抑制効
果を見出したものである。このスタセリンC末端(29
−43残基)ペプチドの両端、すなわち、N末端のLY
P残基、C末端のYTF残基を含むペプチドにも効果が
認められるが、本発明においては、特にLYPQPYQ
PQ、PQYQQYTFが望ましく、さらには、この両
者(LYPとYTF)を結合したLYPQYTFが望ま
しい。
【0008】該ペプチドは、ペプチド合成に常用される
固相法などで容易に合成することが可能である。例え
ば、ジャーナル・オブ・ケミカル・ソサイェティー、第
85巻、2149−2154ぺージ(1963年)記載
のメリフィードらの方法に準じて合成することが可能で
ある。市販のペプチドシンセサイザーなどによっても合
成することができる。また、本発明のペプチドのアミノ
酸配列をコードする遺伝子を利用することで遺伝子工学
的手法を用いて、微生物細胞、植物細胞、動物細胞にお
いて大量に生産することが可能である。得られた粗合成
ペプチドは、ゲル濾過、逆相HPLC、イオン交換カラ
ム精製など、通常の蛋白質・ペプチドの精製に用いられ
る手段により、さらに高純度化することが可能である。
【0009】本発明においては、これらペプチドは単独
であるいは2種以上組合せて用いることができる。
【0010】本発明のう蝕原因菌付着防止剤は、各種口
腔ケア剤(口腔用組成物)に配合して使用することがで
き、この場合、う蝕原因菌付着防止剤(上記ペプチド)
の配合量は、組成物全体の0.0001〜5%(質量百
分率、以下同じ)、特に0.001〜1.0%が好まし
い。
【0011】本発明の口腔用組成物においては、これら
ペプチドは単独で、あるいは2種以上組合せて用いるこ
とができ、組成物中の該ペプチド配合量は0.0001
〜5%、特に0.001〜1.0%が好ましい。
【0012】本発明に係わるう蝕原因菌付着防止剤は、
このように各種の口腔ケア剤、例えば口腔液剤、口腔用
パスタ、歯磨、洗口剤、口腔用トローチ、タブレット、
チュウーイングガムなどに適用できる。それぞれの組成
物は、その特徴に応じ、その他の成分を本発明の効果を
損ねない範囲で使用し、通常の方法で調製することがで
きる。任意成分としては、例えばリン酸水素カルシウム
二水和物、リン酸水素カルシウム無水物、炭酸カルシウ
ム、水酸化アルミニウム、酸化アルミニウム、無水ケイ
酸等の研磨剤、グリセリン、ソルビット、プロピレング
リコール等の粘稠剤、カルボキシメチルセルロースナト
リウム、カラギナン、キサンタンガム等の粘結剤、ラウ
リル硫酸ナトリウム等の界面活性剤、サッカリン等の甘
味剤、その他防腐剤、香料、着色剤、pH調整剤、賦形
剤、各種薬効成分等を配合することができる。

【発明の効果】本発明の細菌付着防止剤は、唾液蛋白由
来ペプチドにより、ミュータンス連鎖球菌の口腔内定着
を特異的に抑制し、う蝕の予防に非常に有効である。

【実施例】次に、実施例を示して本発明の効果を具体的
に説明するが、本発明は下記実施例に制限されるもので
はない。
【0015】[実施例]細菌付着阻止実験
歯のモデルとしてハイドロキシアパタイトビーズ(旭光
学工業社製)をヒト濾過唾液と60分反応させ、さら
に、非特異的部分をコートするためアルブミン溶液(5
mg/ml)で30分インキュベートした。洗浄後、K
Cl緩衝液(50mM KCl、1mM KH2PO4、
1mM CaCl2、0.1mM MgCl2、pH7)
に懸濁した、[H3]ラベルした細菌溶液(約107ce
lls)とサンプル溶液を加えて60分間反応した。菌
はストレプトコッカス・ミュータンス(Strepto
coccus mutans)、口腔内常在菌としてス
トレプトコッカス・ゴルドニィ(Streptococ
cus gordonii)、アクチノマイセス・ナエ
スルンディ(Actinomyces naeslun
dii)を用いた。洗浄後、アパタイトビーズをバイア
ルに移し、液体シンチレーションカウンターでその放射
活性を測定し、付着した菌量を求めた。付着阻止率は、
下記の式に従って算出した。結果を表1に示す。

作用せず、ミュータンス菌にのみ、唾液コートハイドロ
キシアパタイトビーズヘの付着抑制効果を示し、ミュー
