法
発明の背景発明の分野
この発明は遺伝子の特異的突然変異を作成する方法とベクターに関する。さら
に具体的に言うと、発明は真核生物ゲノム、特に胚性幹細胞の相同組み換えの手
法による正確かつあらかじめ決められた遺伝子座における、ゲノムの修飾に有効
なベクターシステムの使用に関係する。関連技術の説明
染色体変質とそれによる遺伝子の構造及び/あるいは発現の修飾を導くことが
多くの異なった科学技術において記載されてきた。標的突然変異誘発に対するあ
る技術は相同組み替えに基づいて作られている。細胞のゲノム内での標的突然変
異の一般的な方法論と特異的な遺伝的障害をもつ遺伝的に変えられた胚性幹(E
S)細胞由来のマウス系の発生の過程はよく知られている(Bradly,1991,Cur . Opin.Biotech.,
2:823−829)。
標的染色体遺伝子座の遺伝情報を含む合成組み換えベクターは細胞に導入され
た後、ゲノムDNAと組み換えをする。ベクターは通常非組み換え細胞のプール
に対しベクターが染色体DNAと問題なく組み換えられる細胞を豊富にする標的
遺伝子座の遺伝情報に隣接した正の選択カセットを持つ。安
定した組み込みはある薬理学上の毒に対して長期間の耐性を導きだす。例はG41
8すなわちゲネチシンあるいはヒグロマイシンに対する、それぞれネオマイシン
あるいはヒグロマイシン耐性遺伝子の作用による耐性である。染色体ベクターD
NAでの正の選択カッセトの配置は古典的なノックアウト実験すなわち遺伝子機
能の不活性化に関する遺伝子の突然変異をさらに導きだすことができる。さらに
、標的遺伝子の調節要素の不活性化あるいは修飾は転写/翻訳遺伝子産物のドメ
インのそれと同様に将来の標的遺伝子機能に対し正、負あるいは調節する効果を
持つことができる。
相同組み換えは正の選択カセットに隣接する標的DNAにより実行され、ベク
ターの末端を越えて起こると考えられる非要望の非相同組み換えをバックグラン
ドとして選択されなければならない。ベクターの末端に位置している負の選択カ
ッセトは非相同組み換え結果により頻繁に組み込まれる可能性がある。負の選択
標識の安定的発現は別の非細胞毒性薬剤の細胞毒性を導きだす。例は単純ヘルペ
スウイルスチミジンキナーゼ遺伝子産物(HSK−TK)の作用によるGancyclo
virの細胞毒性の活性化である。相同組み換えの機会が得られる可能性は染色体
ベクターDNAの大きさともにを増加しさらにベクターのゲノムDNAと標的細
胞の間の同質遺伝子度(isogenecity)に依存する。
標的遺伝子の巨大な染色体断片(5,000−15,000塩基対)のクローニング、バ
クテリアプラスミドベクター内のこのD
NAのサブクローニング、遺伝子構造のマッピング、ベクターへの正の選択カッ
セトの組み込み、そして最後に、負の選択標識による相同ベクターアームの一つ
あるいは両方の隣接化を行う作業はは技術的に厳しい作業で一般的に長い構築時
間(3−6カ月)が要求される。それゆえ、組み換えベクターそれ自身の構築が
最も頻繁に標的突然変異誘発実験の時間的な制限段階となる。
以前の技術では突然変異誘発を指示する相同組み換えのためのベクターの構築
の効果的な手法が欠如した不完全なのもである。本発明はその技術においてこの
長年の必要性と要望を満たすものである。
発明の要約
本発明の目的は哺乳類細胞における相同組み換えによる遺伝子の標的突然変異
のためのベクターの構築の平易化である。これらのベクターは特異的な突然変異
細胞、個別の突然変異細胞から誘導された細胞系、及びトランスジェッニク非ヒ
ト動物の生産のために使用される細胞の構築を容易にする。
本発明の別の目的は従来のベクター構築における欠点を除くベクター系を供給
することである。
本発明は正/負の選択カセットの構築を平易化するベクター系と新しい手順を
供給する。この新しい方法はこれらのベクターの構築に要求される時間を3−6
カ月から約14日に減少させる。
本発明で意図される特に有用なベクター類はスタッファー
断片、複製のE.coli原点、抗生物質耐性遺伝子、複製のイースト原点、イース
トでの使用に適する選択できる標識、哺乳類細胞での使用に適する負の選抜でき
る標識、線状ラムダファージベクターのE.coli/イーストシャトルプラスミドへ
の転換を指示するCre組み換え酵素のためのLoxP配列を構成するゲノムDNAラ
イブラリーの構築ための線状ラムダベクター(ラムダKOS、すなわちノックア
ウトシャトル)を含む。
本発明により意図される追加的ベクターはクローンゲノムDNAに正の選択カ
セットを特異的に挿入することをデザインされたベクターである。そのベクター
は複製のE.coli原点、抗生物質耐性遺伝子、イーストでの使用に適する選抜で
きる標識、哺乳類細胞での使用に適した正の選抜できる標識、標識が別の正の選
択できる標識と交換するための正の選択できる標識に隣接する特有の制限エンド
ヌグレアーゼ、ベクターの正の選択カセットの削除のための特有の制限エンドヌ
クレアーゼ及びシャトルプラスミドのゲノム標的部位へのイースト媒介の組み換
えの後にベクターからこれらの配列の除去を容易にする細菌及びイースト配列に
隣接する制限エンドヌクレアーゼ配列を構成する。
本発明の付加的な具体化例はクローンゲノムDNAの特異的な部位に突然変異
を発生させる方法で、線状ラムダベクターでのゲノムDNAクローニング、関心
対象クローンの分離、関心対象ゲノムDNAを含む線状ラムタベクターの環状E
.
coli/イーストシャトルベクターへの転換、正の選択カセットの標的化を意図す
るゲノムDNA配列の同定、環状ゲノムシャトルベクターに対する正の選択カセ
ットの標的化を意図する部位に隣接する配列に相補的なデオキシオリゴヌクレオ
チドの合成、連結反応あるいはPCRによるデオキシオリゴヌクレオチドの正の
選択カセットへの付着、イースト宿主細胞中で関心対象ゲノムDNAを含むE.c
oli/イーストシャトルベクター及び修飾された正の選択カセットとの同時形質転
換の段階を構成し、そこにおいて完全な組み換えプラスミドはE.coli/イースト
シャトルベクター及び正の選択カセットの2つの独立したイーストの選択できる
標識による遺伝子産物供給方法により適切な培地でのイーストの培養により選抜
され、そしてゲノムDNAを含むベクターとその結果ゲノムDNA配列に挿入さ
れた正の選択カセットを持つ新しい組み換えベクターを発生する正の選択カセッ
トの終末に結合されている合成された配列の相同領域との間での相同DNA組み
換えを行うことにより得られる。イーストプラスミドの分離、E.coliの形質転
換及びE.coliプラスミドの分離の後、正の選択カセットベクターのE.coli/イ
ースト配列は除去され、新しい組み換えベクターをベクター中でE.coli/イース
ト配列と隣接する正の選択カセットベクターと一体となった特有の制限エンドヌ
クレアーゼ配列に対し特異的な制限エンドヌグレアーゼによる切断、前記切断ベ
クターの連結、及び除去されることが要望されるE.coli/イースト配
列を欠く連結産物の同定の段階で構成される。
クローンゲノムDNAの配列の外側の特有の制限エンドヌクレアーゼ配列での
新たな組み換えベクターの線状化、線状DNAの動物細胞への導入、正の選択で
きる標識を発現する導入動物細胞の選択の段階で構成される突然変異動物細胞の
生産の方法もまた供給される。なるべくなら、動物細胞は胚性幹細胞あるいは個
体発生の潜在性を持ついずれかの他の細胞タイプがよい。
本発明のさらに別に具体化例は、動物胚への突然変異細胞の導入、母体の子宮
への突然変異細胞を含有する胚の移植、及び変換ゲノムDNAを含有する核と受
精卵の核との置換の段階から構成される非ヒトトランスジェニック動物の生産の
ための方法の供給である。
さらに線状ラムダベクターで構築されるゲノム及びサブゲノムライブラリーが
供給され、そこにおいてライブラリーは動物細胞、哺乳類細胞、齧歯類細胞、ネ
ズミ細胞、及び他の高等真核生物細胞で構成される群から分離されたゲノムDN
Aから派生する。

ットの挿入に対する3個の特有のエンドヌクレアーゼ制限部位(BamHI,EcoRI及
びXhoI)に隣接する3個のエンドヌクレアーゼ制限部位(SfiI,SwaI及びAscI)
の逆方向繰り返しを構成している。中央の3個の部位のいずれもが選択カセット
の挿入に使用することができる。
発明の詳細な説明
ここにおいて「挿入突然変異誘発システム」とは挿入部位においてゲノム配列
の置換を伴うかあるいは伴わない遺伝標識の挿入により遺伝子座に突然変異を起
こすことを許す遺伝
システムに言及したものとして使われる。
ここにおいて「標的突然変異誘発」とは選択カセットによる遺伝子座の挿入あ
るいは置換部分による遺伝子座の突然変異に言及したものとして使われる。突然
変異の部位は一般には相同組み換えにより選択される。
ここにおいて「線状ラムダベクター」とはラムダファージに感染され、線状フ
ァージDNAが環状プラスミドDNAに結合しているラムダファージCre発現
バクテリアから調整されたDNAと対比して、線状であるラムダファージから調
整されたDNAに言及したものとして使われる。
ここにおいて「E.coli/イーストシャトルベクター」とはE.coli及びイース
ト細胞の中で生き残る、すはわち増殖及び選択されるプラスミドの複製原点を含
むあらゆるプラスミドに言及したものとして使われる。
ここにおいて「Cre組み換え酵素」とは2つのLoxP要素を構成するDNAの短
い範囲の間での組み換えるCre組み換え酵素タンパク質の活性に言及したものと

ここにおいて「Cre/LoxP組み換えシステム」とはCre組み換え酵素タンパク質
の発現と活性が二つの同族のLoxP配列の間の組み換え効果を導くシステムについ
て言及したものとして使われる。
本発明は哺乳類細胞において相同組み換えにより遺伝子の標的突然変異の作業
を十分に改良するDNAベクター構築の方法を記載する。一つを具体化すると、
本発明は相同組み換えによる哺乳類細胞の標的複製突然変異誘発のためのベクタ
ー構築に有効な挿入突然変異誘発システムを供給する。そこの突然変異誘発シス
テムは(a)負の選択標識により隣接されているゲノムDNAのクローニングの
ための線状ラムダベクター、及び(b)負の選択カセットをクローンゲノムDN
Aに挿入するためのベクターを構成する。
この方法論の非常に特異な特徴は、クローニング部位及びE.coli及びイース
ト細胞に対して、正の選択カセットをクローンゲノムDNA配列へイーストにお
いてイースト複製欠如の正の選択カセットの末端に結合し、標的ゲノムDNA配
列と同一な合成DNA配列により指示される相同組み換えによりの挿入する複製
原点と隣接する負の選択できる標識を持つベクターにおけるゲノムDNA構築を
含むことである。この方法論は正/負の選択カセットの構築を簡易化し、さらに
これらのベクターの構築に必要とされる時間を3−6カ月から約14日に減少する
。
線状ラムダベクター(ラムダKOS)システムは本質的に
すべての真核生物ゲノムの典型的ゲノムライブラリー構築を許すラムダファージ
クローニングを根拠に発明された。本発明のこのベクターはゲノムDNA、複製
のE.coli原点、抗生物質耐性遺伝子、複製のイースト原点、イーストでの使用
に適する選択できる標識、哺乳類細胞での使用に適した負の選択できる標識、線
状ラムダファージベクターからE.coli/イーストシャトルプラスミドへの転換を
指示する組み換え酵素に対する組み換え酵素配列の直線的繰り返しの置換および
さらにはクローニングを容易にする制限エンドヌグレアーゼ配列に隣接するDN
Aのスタッファー(stuffer)断片を構成する。なるべくなら、このベクターあ
るいはそのかわりとなる変異体は別の負の選別できる標識の使用により作られる
ほうがよい。この挿入突然変異誘発システムでは組み換え酵素配列及び一致する
組み換え酵素はLoxP配列−Cre組み換え酵素及びFrt配列−Flp組み換え酵素ある
いはこの技術により通常の技能を持つことが容易に理解できる同様な代用物質か
ら構成されるグループから選抜される。
本発明により意図される付加的なベクター、すなわち正の選択カッセトベクタ
ーは正の選択カセットをクローンゲノムDNAに特異的に挿入することを予定さ
れたベクターである。このベクターは複製のプラスミド原点、抗生物質耐性遺伝
子、イーストでの使用に適する選択できる標識、哺乳類細胞での使用に適する正
の選択できる標識、標識が別の正の選択標識に交換されることができるために正
の選択できる標識に隣接
する特有の制限エンドヌクレアーゼ配列、ベクターから配列の除去を容易にする
細菌及びイースト配列に隣接する制限エンドヌクレアーゼ配列及びベクターの直
線化のための特有の制限エンドヌクレアーゼ配列で構成される。なるべくなら、
正の選択カセットベクターあるいはそれに代わる変異体は別の正の選択標識を使
用することにより作成されるほうがよい。
発明の付加的な具体例は、クローンゲノムDNAの特定の部位に対する突然変
異を発生させる方法の供給であり、線状ラムダベクター中でのクローニング、関
心対象クローンの分離、関心対象のゲノムDNAを含有する線状ラムダベクター
をCre組み換え酵素発現細菌株を感染により環状E.coli/イーストシャトルベク
ターへの変換、正の選択カセットの標的化を意図するゲノムDNA配列の同定、
環状E.coli/イーストシャトルベクターへの正の選択カセットの標的化を意図す
る部位に隣接する配列に相補的なデオキシオリゴヌクレオチドの合成、連結反応
あるいはPCRによる合成デオキシオリゴヌクレオチドの正の選択カセットへの
付着(実施例2参照)、イースト宿主細胞中の関心対象のゲノムDNAを含有す
る前記E.coli/イーストシャトルベクター及び修飾した正の選択カセットの同時
形質転換、そこにおいて完全な組み換えプラスミドはE.coli/イーストシャトル
ベクター及び正の選択カセットのイーストの選択できる標識により供給される遺
伝子産物の手法により適切な培地でイーストの培養により選択され、ゲノムDN
A含有ベクターの相同領域及び
そこにおいてゲノムDNA配列に挿入された正の選択カセットを持つ新たな組み
換えベクターを発生させる正の選択カセットの末端に連結されている合成的に派
生した配列の間での相同DNA組み換えの実行により獲得される段階で構成され
る。なるべくなら、正の選択カセットのE.coli/イースト配列は除去されること
がよく、これはベクター内でE.coli/イースト配列と隣接する正の選択カセット
ベクターを組み込んだ特有の制限エンドヌグレアーゼ配列により特異的な制限エ
ンドヌクレアーゼによる新たな組み換えベクターの切断、切断ベクターの連結、
除去されることが望まれるE.coli/イースト配列を欠く連結産物の確認の段階で
構成される。
本発明はまた突然変異動物の生産の方法を指示し、これはクローンゲノムDN

発明を他の相同組み換えベクター構築の技術と比較してみると以下の利点が挙
げられる。第1に、組み換えファージライブラリーは関心対象の個々のゲノムに
対し一度だけで準備でき、その後バクテリア内のファージライブラリーとしてを
1000倍に拡大することができる。第2に、それぞれの遺伝子座は関心対象の遺伝
子プローブにより自動的に分離の準備がなされる。第3に、ファージシステムは
相同組み換えの成功を助ける巨大な染色体DNAを増殖させることができる。第
四に、Cre組み換え酵素発現細菌株の単純なトランスフェクションはゲノムDN
A自動的に負の選択カセットと隣接するシャトルプラスミドを作り出す。第5に
、正の選択カセットの導入は公表されている方法に従って行った(Nuclec Acid Res ,
24:4594−4596)、しかし、ここで記載され新たに開発した正の選択カセ
ット及び変異体の構築を簡易にするpMCS−1によりさらに改良される。
本発明どおりに、これらは技術の技能の範囲内で従来の分子生物学、微生物学
、免疫学及び組み換えDNA技法に使用が可能と推測される。これらの技法は文

以下の用語は以下に掲げるような定義もつように定められ。「DNA分子」と
は一本鎖型あるいは二重鎖らせんのいずれかの状態のデオキシリボヌクレオチド
(アデニン、グアニン、チミン、あるいはシトシン)の重合体について使われる
。この用語は分子の1次及び2次構造に体してのみ使われるが、特殊な3次構造
に対してこれを制限するものではない。したがって、この用語はとりわけ線状D
NA分子(例えば、制限断片)、ウイルス、プラスミド及び染色体で見いだされ
る二重鎖DNAを含む。ここでの議論では、一般の慣例に従いDNAの未転写鎖
5'から3'方向に沿った方向で配列についての構造について行われる(すなわち
、鎖はmRNAと相同な配列をもつ)。
DNA「解読配列」とはin vivoにおいて適切な調節配列の支配下で転写され
ポリペプチドに翻訳される二重鎖DNAである。解読配列の境界は5'(アミノ
)末端の出発コドン及び3'(カルボキシル)末端の翻訳停止コドンにより決定
される。解読配列は、真核生物の配列、真核生物mRNAからのcDNA、真核
生物(例えば、哺乳類)DNAからのゲ
ノムDNA、及び合成DNA配列さえも含まれるが、それに限定するものではな
い。ポリアデニレーション信号及び転写終止配列は通常解読配列に対して3'に
所在するであろう。
ここにおいて使用される「オリゴヌクレオチド」あるいは「デオキシオリゴヌ
クレオチド」の用語は2個かそれ以上、一般には3個以上のオリゴヌクレオチド
あるいはデオキシオリゴヌクレオチドで構成される分子として定義される。その
正確な大きさはオリゴヌクレオチドの究極機能と使用に依存する多くの要因に順
に依存するであろう。
ここにおいて使用される「プライマー」とは精製された制限分断物としての天
然性の発生物か合成的な製造物であるかにかかわらず、核酸鎖に対し相補的であ
るプライマー伸展合成が起きこる条件下、すはわちヌクレオチド及びDNAポリ
メラーゼのような誘導試薬が存在し適切な温度とpHの条件におかれた場合に合成
の開始の地点として作用することができるオリゴヌクレオチドとして言及される
。ブライマーは一本鎖あるいは二重鎖のいずれかであり、誘導試薬の存在下で要
望する伸展産物の合成を十分長期間始動しなければならない。プライマーの正確
な長さは、温度、プライマーの原料、および使用する方法を含む様々な要因によ
り決定される。例えば、診断学上の応用の場合、標的配列の複雑性に依存し、オ
リゴヌクレオチドプライマーは少数のヌクレオチドしか含まれない場合もあるか
もしれないが、典型的なものは15−25かそれ以上のヌクレオチドを含有している
。
ここにおいて使用される「PCR」の用語は技術上知られている他の改良点を
加えた、Millisに対しとU.S.Patent Nos.4,689,195及び4,683,202の主題であ
るポリメラーゼ鎖反応について言及したものである。
ここにおいて使用される「制限エンドヌクレアーゼ」及び「制限酵素」の用語
は、いずれも特異的なヌクレオチドのその地点あるはその近辺で二重鎖DNAを
切断する細菌の酵素につて言及したものである。
これらの研究で最も一般的に用いられる標識には放射性元素、酵素、紫外線を
照射した場合に蛍光を発する化学物質、その他のものが使用される。数多くの蛍
光物質が知られており標識として利用することができる。これらには、例えば、
フルオレセイン、ローダミン、オーラミン、テキサツレッド、AMCAブルー、
およびルシフェラーイエローが含まれる。特殊な検出物質はヤギの体内で調整さ
れイソチオシアネートによってフルオレセインと結合した抗ウサギ抗体である。
以下の実施例は発明の様々な具体例を示す目的で掲げられており、いかなるや
り方においても本発明を制限することを意味するものではない。
実施例1 線状ラムダベクター(ラムダKOS)の創作
ベクターシステムは本質的にすべての真核生物ゲノムの典型的ゲノムライブラ
リーの構築ができるラムダファージクローニングを根拠として創作された。線状
ラムダベクター(ラ
ムダKOS)は左から右にかけて次の特質部を持つ(図1参照)。cos末端及び
ファージの左腕、合成LoxP断片、HSV−tk遺伝子、エンドヌクレアーゼ認知部
位BamHI及びSalI、スタッファー断片、エンドヌクレアーゼ認知部位SalI及びBam
HI,HSV−tk遺伝子、プラスミドベクター(複製の細菌原点、アンピシリン耐
性のためのβ−ラクタマーゼ遺伝子)、エンドヌクレアーゼ認知部位NotI、2ミ
クロン複製のイースト原点、イースト栄養要求変異TRP1遺伝子、合成LoxP断
片(前のものと相対的に同じ方向性)及びファージの右腕及びcos末端。溶菌成
長したファージから調整したDNAはエンドヌグレアーゼSalI及びBamHI、及び
エンドヌクレアーゼSau3AIにより部分切断したゲノムDNAにより置換され
たスタッファー断片により切断することができる。真核生物ゲノムの数倍の典型
的ゲノムライブラリーがそれによって確立できる。それぞれのゲノム断片(10,0
00−15,000塩基対)はいずれかの部位で負の選択カセットに自動的に隣接する。
ゲノムライブラリーは数倍過剰であるので、実質的にそれぞれすべてのゲノム遺
伝子座がラムダKOSファージにより表される。関心対象のゲノムグローンはラ
ムダファージライブラリーの古典的なフィルターハイブリット形成によりふるい
