本発明は、タンパク質チロシンキナーゼ2(PYK2)をコードする核酸、マ
ウスPYK2、前記核酸を用いる前記タンパク質の製造方法、及びPYK2阻害
剤に対するアッセイに関する。発明の背景
細胞接着キナーゼβ(CAKβ)及び関連アドヒージョンフォーカルチロシン
キナーゼ(RAFTK)としても公知のタンパク質チロシンキナーゼ2(PYK
2)は、フォーカルアドヒージョンキナーゼファミリーの最近発表されたメンバ
ーである(Avrahamら,J.Biol.Chem.,270:27742
−27751(1995);Levら,Nature,376:737−745
(1995);及びSasakiら,J.Biol.Chem.,270:21
206−21219(1995))。PYK2はまずヒト脳からGrb−2結合
タンパク質としてクローン化され、ラット及びヒト脳ライブラリ
ーからもクローン化された。その細胞発現に関して矛盾する報告もある。1つの
研究では、大量のPYK2転写物が脳で発見され、少量が腎臓で検出された。別
の研究では、PYK2発現がラット脳において最も多量に存在することが認めら
れたが、その転写物は腎臓、脾臓、肺、小腸及び副睾丸でも検出された。PYK
2転写物はラット線維芽細胞3T1及びWFB細胞系、及びヒトT細胞白血病J
urkat細胞でも検出された。ヒト巨核球CMK細胞系及びマウス脳からクロ
ーン化すると、脳を越えて広い組織に、特に脾臓、肺、胸腺及び末梢血白血球に

本発明の第1面は、マウスタンパク質チロシンキナーゼ2(PYK2)をコー
ドする、関連核酸を実質的に含まない核酸である。1つの実施態様では、PYK
2をコードする核酸はDNAである。
本発明の別の面は、マウスPYK2 cDNAである。マウスPYK2 DN
Aは図1に記載されている(配列番号5)。
本発明の別の面は、関連マウスタンパク質を含まないマウス
PYK2である。1つのマウスPYK2は図1に記載されている(配列番号6)
。
本発明の別の面は、細胞にPYK2をコードする核酸からなる核酸をPYK2
の転写及び翻訳が起こる条件下で導入することによるPYK2の製造方法である
。好ましくは、核酸をプラスミドまたはバクロウィルスベクターのようなベクタ
ーに存在させる。また、核酸がプロモーター及び任意にエンハンサーのような転
写調節因子の制御下にあることが好ましい。前記調節因子は当業界で公知である
。
PYK2を発現する宿主細胞も本発明の一部をなす。好ましい宿主細胞には、
哺乳動物細胞、昆虫細胞、酵母、及び大腸菌のような細菌細胞が含まれる。(一
時的よりむしろ)永久的にマウスPYK2を発現する細胞系も本発明の一部をな
す。
本発明のクローニング方法を用いて製造される組換えPYK2は、PYK2の
生物学的機能を更にキャラクタライゼーションし、その活性を調節するアゴニス
トやアンタゴニストのような化合物を同定するためのアッセイにおいて使用され
得る。本発明の更なる面は、PYK2の活性を調節する化合物及び特にPYK2
活性の阻害剤を同定するためのアッセイである。この
アッセイは、組換えPYK2を放射能標識ATP及び推定される活性調節化合物
の存在下でチロシン基質と接触させるステップ、形成された放射能標識チロシン
の量を測定するスステップ、及び任意に推定される活性調節化合物の存在下で形
成された放射能標識チロシンの量を推定される活性調節化合物の非存在下で形成
された放射能標識チロシンの量と比較するステップを含む。
インテグリンは、隣接細胞または細胞外マトリックスのいずれかに対する接着
相互作用を媒介する細胞表面受容体の主要ファミリーである。インテグリンのシ
グナル化は、タンパク質のフォーカルアドヒージョンキナーゼ(FAK)ファミ
リーを介して媒介される。PYK2はFAKファミリーのメンバーであり、巨核
球、脳組織及び造血細胞におけるインテグリン媒介シグナル変換経路に関与して
いる。従って、PYK2のモジュレータは血小板レベルを調節するための有力な

明細書及び請求の範囲で使用した用語は以下のように定義される。
「PYK2」は、タンパク質チロシンキナーゼ2、タンパク質チロシンキナー
ゼ2のアレリック変異体、及び天然PYK2の生物学的活性の少なくとも約85
%、好ましくは約90%が保持されているその突然変異体または断片を指す。
「天然PYK2」は、生物中にもともと存在するタンパク質チロシンキナーゼ
を指す。
「関連核酸を実質的に含まない」とは、サンプル中にPYK2をコードする核
酸以外の核酸が約5重量%未満しか存在しないことを意味する。
「関連マウスタンパク質を実質的に含まない」とは、サンプル中にマウスPY
K2以外のタンパク質が約5重量%未満しか存在しないことを意味する
「FAK」は、フォーカルアドヒージョンキナーゼを指す。
「異種」PYK2核酸は、核酸が細胞と同じ種に由来するかに関係なく核酸が
細胞に導入されたことを意味する。或いは、
その先祖が細胞に導入されたPYK2を有する細胞においてPYK2をコードす
る核酸を指す。
「異種」PYK2タンパク質は、PYK2が異種核酸によりコードされること
を意味する。
細胞接着過程に関与するFAKタンパク質は、多数のマクロファージ細胞系に
おいて検出されなかった。従って、FAKに相同の別の細胞接着依存性キナーゼ
はこれらの細胞においてその機能を有し得ると仮定された。PYK2は最近、F
AKファミリーの別のメンバーとして同定され、その発現が脾臓、胸腺、肺及び
末梢血白血球において検出された(上掲のAvraham(1995);上掲の
Sasaki(1995))。PYK2をマクロファージにおいて可能性ある接
着依存性キナーゼとして評価するために、PYK2及びFAKに対する特異的プ
ローブを作製し、これをマウス組織においてPYK2及びFAKの発現を試験す
るために用いた。他の種については既に報告されているように、PYK2は脳及
び脾臓において高度に発現し、腎臓、肺及び肝臓における発現はより低レベルで
あり、FAKに比してより狭い組織に分布している。
PYK2プローブを用いて、完全長cDNAをマウス脾臓cDNA
ライブラリーからクローン化した。完全長クローンの推定アミノ酸配列は、最近
公表されたマウスRAFTKのアミノ酸配列と同一であることが判明した(上掲
のAvrahamら(1995))。
更に、完全長FAKをマウス骨芽細胞MB1.8細胞系(Wesolowsk
iら,Exp.Cell Res.,219:679−686(1995))か
らクローン化した。
PYK2 cDNA及びFAK cDNAをその後ヒト胚腎臓(HEK)29
3細胞にトランスフェクトした。永久的にPYK2またはFAKを発現する細胞
系が樹立され、外来発現したマウスキナーゼの発現レベルをノーザン分析により
評価した。
本発明を更に例示するために下記実施例を提示する。
実施例1細胞培養
チオグリコレートを成体BALB/cマウスの腹膜腔に注入してマクロファー
ジを誘導した。4日後、細胞を集め、洗浄し、10%FBS含有RPMI 16
40培地において培養した。37℃で3時間後、培養物を十分に洗浄して非接着
細胞を除去し、一晩培養後、免疫沈降用サンプルを作成した。骨髄由来マ
クロファージを、援用により本明細書に含まれるとするLi及びChen,J.
Leuk.Biol.,57:484−490(1995)に記載されているよ
うに作成した。非接着細胞を、ヒトマクロファージコロニー刺激因子(MCS−
F,250単位/ml,マサチューセッツ州ケンブリッジに所在のGeneti
cs Institute)の存在下でRPMI補充培地において培養した。5
日間培養後、免疫沈降用の分化マクロファージを作製した。
骨髄由来骨芽細胞様細胞を、援用により本明細書に含まれるとするWesol
owskiら,Exp.Cell Res.,219:679−686(199
5)に記載されているように作製した。コラゲナーゼーディスパーゼ処理後、組
織培養プレートから30nMエチスタチン(ペンシルバニア州ウェストポイント
に所在のMerck Res.Labs.)を用いて単核のタータレート耐性ホ
スファターゼ陽性細胞を遊離させた。新しく単離した骨芽細胞様細胞を直ちに免
疫沈降緩衝液で溶解した。
実施例2マウスPYK2のcDNAクローニング及び発現
マウスPYK2及びFAKに対する特異的プローブをまず、キナーゼドメイン
のC−末端に隣接するタンパク質間の非相同領域に基づいて作製した。ポリメラ
ーゼ連鎖反応(PCR)により、PYK2(570bp)に対する特異的プロー
ブを5’−プライマ−AGTGA CATTT ATCAG ATGGA G(
配列番号1)及び3’−プライマ−GAATG GACTG TGCAC CG
AGC C(配列番号2)を用い、マウス骨髄由来骨芽細胞様細胞のcDNAを
鋳型として作製した(上掲のWesolowskiら(1995))。
同様に、FAK(700bp)に対する特異的プローブを、プライマ−5’−
CAGCA CACAA TCCTG GAGGA G(配列番号3)及び3’
−GCTGA AGCTT GACAC CCTCA T(配列番号4)を用い
、マウス骨芽細胞MB1.8細胞のcDNAを鋳型として作製した(上掲のWe
solowskiら(1995))。これらのプローブを配列決定分析により確
認した。PYK2 cDNA断片を、特異的PYK2プローブを用いてマウス脾
臓λ−ZAP II cDNAライブラリー
(カリフォルニア州ラ・ホーヤに所在のStratagene)からクローン化
した。完全長PYK2 cDNAをVspIサイトで2つの重複クローンを連結
して構築した。単離したPYK2 cDNAクローンのアミノ酸配列は既報(上
掲のAvrahamら(1995))のマウスRAFTK配列と同一であった。
完全長FAK cDNAを、Hanksら,Proc.Natl.Acad.S
ci.USA,89:8487−8491(1992)に記載されている公知の
配列に従ってPCRにより作製した。
PYK2 cDNA及びFAK cDNAの両方をpCDNA3プラスミド(
カリフォルニア州サンジェゴに所在のInVitrogen)にサブクローン化
し、GenePulser(カリフォルニア州リッチモンドに所在のBiora
d Labs)を用いて200V、960μFでのエレクトロポレーションによ
りヒト胚腎臓(HEK)293細胞(メリーランド州ロックランドに所在のAT
CC)にトランスフェクトした。その後、HEK293細胞をG418選択(8
00μg/ml,Gibco BRL)にかけ、選択培地で3週間後クローンを
選んだ。
PYK2及びFAKのHEK293細胞における発現をそれそれのプローブを
用いてノーザン分析により確認し、ウェスタ
ンブロットを抗−PYK2抗体を用いて実施した。マウス多組織ノーザンブロッ
トはClonetech(カリフォルニア州パロアルト)から購入し、PYK2
、FAK及びグリセルアルデヒド3−ホスフェートデヒドロゲナーゼ(GAPD
H)に対して特異的なプローブを用いるノーザンブロットのハイブリダイゼーシ
ョンは上記(上掲のWesolowskiら(1995))のように実施した。
実施例3マウスPYK2に対するポリクローナル抗体の産生及びアフィニティー精製
PYK2のC−末端ドメイン(メチオニン残基685から末端まで)をマウス
PYK2を鋳型として用いてPCRにより増幅した。増幅産物をプラスミドpG
EX−4T(ニュージャージー州ピスカタウェーに所在のPharmacia
Biotech)にクローン化し、大腸菌XL1−Blue(Stratage
)に形質転換した。本質的に製造業者(Pharmacia)の指示に従って、
GST−PYK2 C−末端断片の発現を0.5mM IPTGを用いて誘導し
、精製し、GSTからトロンビンを用いて開裂した。マウスPYK2の精製C−
末端断片を、
2羽の家兎(アラバマ州ハンツビルに所在のResearch Genetic
s)を免疫化するために使用し、両方の抗血清の力価をまずPYK2の組換えC
−末端断片を用いるELISAにより測定した。その後、免疫血清の特異性を、
前免疫血清と比較することによりウェスタンブロットにより測定した。次いで、
ポリクローナル抗体を両方の抗血清の合わせた画分を、製造業者(Pharma
cia)の指示に従ってCNBr−活性化セファロース4Bに交差結合した同じ
精製抗原を用いて構築したアフィニティーカラムに通すことによりアフィニティ
ー精製した。
抗体を0.2Mグリシン(pH2.5)及び1mM EGTAを用いてカラム
から溶離させた後、溶離画分を0.02%アジド含有PBSに対して透析した。
抗−PYK2抗体を0.5mg/mlの濃度で−70℃で保存した。
実施例4インビトロキナーゼ活性
ECMに細胞接着後、IC−21細胞を50mMトリス−HCl(pH7.4
)、150mM NaCl、1% NP−40、1mM EDTA、10%グリ
セロール、50mM NaF、
1mMバナジン酸ナトリウム及び上記したプロテアーゼ阻害剤を含有するTNE
溶菌緩衝液に溶解した。PYK2を清澄化ライゼートから免疫沈降させ、サンプ
ルの半分を上記した抗PYK2抗体を用いる免疫ブロッティングにかけ、他の半
分を同じ溶菌緩衝液及び20mMトリス−HCl(pH7.4)、10mM N
aCl、10mM MnCl2及び1mMジチオトレイトールを含むキナーゼア
ッセイ緩衝液(1×)を用いて2回洗浄した。洗浄緩衝液を除去後、5μCi[
γ−32P]ATP(3000Ci/mmol,Amersham)、10mM
ATP、0.1% BSA及び100μgポリ(Glu,Tyr)(モル比4:
1,Sigma)を含有するキナーゼアッセイ緩衝液50μlをビーズに添加し
、30℃で10分間インキュベートした(Howell及びCooper,Mo
l.Cell.Biol.,14:5402−5411(1995))。反応混
合物(25μl)を30%トリクロロ酢酸(TCA)及び0.1Mピロリン酸ナ
トリウム25μlに添加後、4℃で15分間インキュベートした。沈降したタン
パク質をMultiscreen−FCフィルタープレート(マサチューセッツ
州Marlboroughに所在のMillipore)に移し、氷冷15%
TCA(3×)
で洗浄し、乾燥し、基質への32P取込みをPackardトップカウントマイク
ロプレートシンチレーションカウンター(コネチカット州メリデンに所在のPa
ckard)で計数した。各アッセイを3回実施した。特異的活性を、放射活性
