カルシウム溶解性化合物
発明の分野
本発明は新規なアリールアルキルアミンのカルシウム溶解性化合物(calcilyt
ic compound)、該化合物含有の医薬組成物およびそのカルシウムレセプターア
ンタゴニストとしての使用に関する。
哺乳動物において、細胞外Ca2+は厳正な恒常的調節下にあり、凝血塊形成、
神経および筋肉興奮性および適度の骨形成などの種々のプロセスを制御する。細
胞外Ca2+は上皮小体細胞から由来の上皮小体ホルモン(PTH)の分泌を阻害
し、破骨細胞による骨吸収を阻害し、カルシトニンのC−細胞からの分泌を刺激
する。カルシウムレセプタータンパク質はある種の分化細胞を細胞外Ca2+濃度
の変化に応答させることができる。
PTHは血液および細胞外流体中のCa2+恒常性を制御する主たる内分泌性因
子である。PTHは骨および腎細胞に作用することで血中のCa2+レベルを増大
させる。この細胞外Ca2+の増加は、PTH分泌を抑制する、負のフィードバッ
クシグナルとして作用する。細胞外Ca2+とPTH分泌との間の相互関係はCa2+
恒常性を具体的に維持する重要な機構を形成する。
細胞外Ca2+は上皮小体細胞に直接作用してPTH分泌を調節する。細胞外C
a2+の変化を検出する上皮小体細胞の表面タンパク質の存在が確認されている。
Brownら,Nature 366:574,1993を参照のこと。上皮小体細胞にて、カルシウム
レセプターであるこのタンパク質は、細胞外Ca2+のレセプターとして作用し、
細胞外Ca2+のイオン濃度の変化を検出し、機能的な細胞応答、PTH分泌を開
始させる。
細胞外Ca2+は、Nemethら,Cell Calcium 11:319,1990に報告されているよ
うに、種々の細胞機能に影響を及ぼす。例えば、細胞外Ca2+は傍濾胞細胞(C
−細胞)および上皮小体細胞にて役割を果たしている。Nemeth,Cell Calcium 1
1:323,1990を参照のこと。破骨細胞における細胞外Ca2+の役割も
研究されている。Zaidi,Bioscience Reports 10:493,1990を参照のこと。
カルシウムレセプター分子に対する細胞外Ca2+の作用を真似る種々の化合物
が知られている。カルシウム溶解性物質(calcilytics)はカルシウムレセプタ
ー活性を阻害し、それにより細胞外Ca2+によって喚起される1またはそれ以上
のカルシウムレセプター活性を減少させることのできる化合物である。カルシウ
ム溶解性物質は、Ca2+レセプターで活性である有用なカルシウムモジュレータ
ーを発見、開発、設計、修飾および/または構築する場合のリード分子として有
用である。このようなカルシウム溶解性物質は、発現および/または分泌が1ま
たはそれ以上のCa2+レプターでの活性により制御または影響される1またはそ
れ以上の要素、例えば、ホルモン、酵素または成長因子などのポリペプチドの異
常なレベルにより特徴付けられる種々の病状の治療にて有用である。カルシウム
溶解性化合物の標的となる疾患または障害として、骨およびミネラルの異常恒常
性に関連する疾患が挙げられる。
カルシウムの異常恒常性は、1またはそれ以上の以下の活性:血清中カルシウ
ムの異常な増加または減少;尿排泄物中のカルシウムの異常な増加または減少;
骨中カルシウム濃度の異常な増加または減少(例えば、骨ミネラル密度測定によ
り評価される);食物カルシウムの異常な吸収;PTHおよびカルシトニンなど
の血清中カルシウム濃度に影響を及ぼすメッセンジャーの産生および/または放
出の異常な増加または減少;および血清中カルシウムレベルに影響を及ぼすメッ
センジャーにより惹起される応答の異常な変化により特徴付けられる。
すなわち、カルシウムレセプターアンタゴニストは、骨またはミネラルの異常
恒常性に伴う疾患、例えば、上皮小体機能低下症、骨肉腫、歯周疾患、骨折治癒
機転、変形性関節症、慢性関節リウマチ、パジェット病、悪性疾患および骨折治
癒機転に伴う体液性高カルシウム血症、ならびに、骨粗髭症の薬物療法に対する
唯一の方法を提供するものである。
発明の概要
本発明は、下記式(I)で示されるアリールアルキルアミン誘導体、ならびに
、限定するものではないが、上皮小体機能低下症、骨肉腫、歯周疾患、骨折治癒
機
転、変形性関節症、慢性関節リウマチ、パジェット病、悪性疾患および骨折治癒
機転に伴う体液性高カルシウム血症、ならびに、骨粗鬆症を含む、骨またはミネ
ラルの異常恒常性に伴う種々の疾患の治療に有用な、そのカルシウムレセプター
アンタゴニストとしての使用を提供する。
本発明の化合物は、他のカルシウムレセプターアンタゴニストと比較して、カ
ルシウムレセプター活性を維持する一方、安定性の増大を示す。
本発明は、さらには、ヒトを含む、動物のカルシウムレセプターを拮抗する方
法であって、その拮抗作用を必要とする動物に、有効量の下記式(I)で示され
る化合物を投与することからなる方法を提供する。
発明の詳細な説明
本発明の化合物は、下記式(I):

アリールまたはC1-20アルキルであり、Yは、CONH、COO、CONHN
HCOおよびCOからなる群から選択され;

およびSR3からなる群から選択され、ここで、R3は、H、C1-15アリールま
たはC1-20アルキルである]
で示される化合物から選択される。
好ましくは、nは、1であり、R2は、Nヘテロ原子に対してベータ位にある
。nが2である場合、R2は、Nヘテロ原子に対してアルファ位またはガンマ位
にある。
好ましくは、R1は、CH2YR3である。
好ましくは、Yは、CONHまたはCOOである。
好ましくは、R2は、CONHR3である。
本明細書で用いる場合、「アルキル」なる語は、炭素−炭素単結合により結合
され、一緒に結合された炭素原子1−20個を有する、所望により置換されてい
てもよい炭化水素基を表す。アルキル炭化水素基は、直鎖状、分枝状または環状
で、飽和または不飽和であってもよい。置換基は、F、Cl、Br、I、N、S
およびOから選択される。好ましくは、3個以下の置換基が存在する。さらに好
ましくは、アルキルは、炭素原子1−12個を有しており、非置換である。好ま
しくは、アルキル基は、直鎖状である。好ましくは、アルキル基は、飽和してい
る。
本明細書で用いる場合、「アリール」なる語は、共役パイ電子系を有する少な
くとも1つの環を有し、2個までの結合または縮合した環系を含有する、所望に
より置換されていてもよい芳香族基を表す。アリールとしては、炭素環式アリー
ル、複素環式アリールおよびビアリール基が挙げられ、これらの全ては、所望に
より置換されていてもよい。置換基は、F、Cl、Br、I、N、SおよびOか
ら選択される。好ましくは、アリールは、所望により置換されていてもよいフェ
ニル、または、所望により置換されていてもよい2−ナフチルである。好ましく
は、アリール基は、非置換である。最も好ましいアリール基は、非置換フェニル
である。
本発明において有用な好ましい化合物としては、以下の化合物が挙げられる:

本発明において有用な最も好ましい化合物は、3−カルバモイル−1−((ドデ
シルカルバモイル)メチル)ピリジニウムクロリドである。
本発明の化合物は、医薬上許容される塩およびその複合体として処方すること
ができる。医薬上許容される塩はその投与される量および濃度で非毒性である。
医薬上許容される塩としては、硫酸塩、塩酸塩、フマル酸塩、マレィン酸塩、
リン酸塩、スルファミン酸塩、酢酸塩、クエン酸塩、乳酸塩、酒石酸塩、メタン
スルホン酸塩、エタンスルホン酸塩、ベンゼンスルホン酸塩、p−トルエンスル
ホン酸塩、シクロヘキシルスルファミン酸塩およびキナ酸塩を含有するような酸
付加塩が挙げられる。医薬上許容される塩は、塩酸、マレイン酸、硫酸、リン酸
、スルファミン酸、酢酸、クエン酸、乳酸、酒石酸、マロン酸、メタンスルホン
酸、エタンスルホン酸、ベンゼンスルホン酸、p−トルエンスルホン酸、シクロ
ヘキシルスルファミン酸、フマル酸およびキナ酸などの酸から得ることができる
。
医薬上許容される塩としてはまた、カルボン酸またはフェノールなどの酸性官
能基が存在する場合、ベンザチン、クロロプロカイン、コリン、ジエタノールア
ミン、エチレンジアミン、メグルミン、プロカイン、アルミニウム、カルシウム
、リチウム、マグネシウム、カリウム、ナトリウム、アンモニウム、アルキルア
ミンおよび亜鉛を含有するような塩基付加塩が挙げられる。
本発明は、標準的技法を用いて製造することのできる上記式(I)で示される
化合物を提供する。本明細書に記載の好ましい化合物を製造する全体的な方法は
、このセクションに記載されているように行うことができる。下記実施例は、特
定化合物の合成を示す。モデルとして本明細書に記載のプロトコルを用いて、当
業者であれば本発明の別の化合物も容易に製造することができる。
全ての試薬および溶媒は、販売元から入手した。出発物質(例えば、アミンお
よびエポキシド)は、標準的技法および手順を用いて合成した。
本発明の化合物の多くを合成するために用いる一般的な手順は、以下のとおり
である:
本明細書に記載する化合物の製造は、従前に公表された方法に従うか、または
、以下に記載する方法に従って行った。本明細書に記載の好ましい化合物を製造
するための全体的な方法は、このセクションに記載されているように行うことが
できる。以下の実施例は、特定の化合物の合成を示す。モデルとして本明細書に
記載したプロトコルを用いて、当業者であれば式(I)で示される別の化合物を
容
易に製造することができる。
全ての試薬および溶媒は、販売元から入手した。出発物質は、標準的技法およ
び手順を用いて合成する。
当該化合物の多くを合成するために用いた一般的な手順は、下記スキーム1に
記載されているように行った。置換ピリジン(例えば、ニコチンアミド、1ミリ
モル)および過剰のアルキル化剤(例えば、N−ドデシル−2−クロロアセトア
ミド、2ミリモル)のニトロメタンまたは類似溶媒(2mL)中溶液を90−1
00℃で24時間攪拌する。典型的には、冷却して溶液からピリジニウム塩を沈
殿させ、対応する塩を回収する。
スキーム1 a.BrR1、NO2CH3、90−100℃、24時間
全ての化学的官能基の適当な操作および保護と共に、上記方法および実験セク
ションに記載する方法と同様の方法により、式(I)で示される残りの化合物の
合成を行う。
ヒトおよび他の哺乳動物の治療に式(I)で示される化合物またはその医薬上
許容される塩を用いるために、通常、標準的な製薬プラクティスに従って医薬組
成物として製剤化する。
当該カルシウム溶解性化合物は、静脈内、腹腔内、皮下、筋肉内、経口、局所
(経皮)または経粘膜投与を含む種々の経路により投与できる。全身投与につい
ては、経目投与が好ましい。経口投与については、例えば、化合物は、カプセル
剤、錠剤ならびにシロップ剤、エリキシル剤および濃縮滴剤のような液体調製物
などの慣用的な経口投与形態に製剤化できる。
別法としては、注射(非経口投与)、例えば、筋肉注射、静脈注射、腹腔内注
射および皮下注射を用いてもよい。注射については、本発明の化合物は、溶液、
好ましくは、生理食塩水、ハンクス溶液またはリンゲル溶液などの生理学上適合
性の緩衝液または溶液中で製剤化する。さらに、当該化合物は、固体形態で製剤
化され、使用直前に再溶解または懸濁してもよい。凍結乾燥形態もまた調製でき
る。
全身投与は、また、経粘膜または経皮手段により行うことができる。経粘膜ま
たは経皮投与のためには、浸透させようとする障壁に適した浸透剤を処方におい
て用いる。かかる浸透剤は、当該技術分野で一般に知られており、例えば、経粘
膜投与については、胆汁酸塩およびフシジン酸誘導体が挙げられる。さらに、洗
浄剤を用いて浸透を促進してもよい。経粘膜投与は、例えば、鼻スプレー剤、直
腸坐剤または膣坐剤により行われる。
局所投与については、本発明の化合物は、当該技術分野で一般に知られている
ように、軟膏剤、膏薬(salve)、ゲル剤またはクリーム剤に製剤化できる。
種々のカルシウム溶解性化合物の投与量は、化合物IC50、EC50、化合物の
生物学的半減期、患者の年齢、大きさおよび体重、ならびに、患者が罹患してい
る疾患または障害などの因子を考慮して標準的な手段により決定できる。これら
の因子および考えられる他の因子の重要性は、当業者に知られている。
投与量は、また、投与経路および経口生物学的利用能の程度にもよる。例えば
、経口生物学的利用能が低い化合物については、比較的高い用量を投与しなけれ
ばならない。
好ましくは、当該組成物は、単位投与形態である。経口用途については、例え
ば、錠剤またはカプセル剤を投与してもよく、鼻用途については、定量型エアゾ
ール剤を投与してもよく、経皮用途については、局所製剤またはパッチ剤を投与
してもよく、経粘膜デリバリー用途については、バッカルパッチ剤を投与しても
よい。各々の場合、投与量は、患者が単回投与で投与してもよいような量である
。
経口投与についての各単位用量は、式(I)で示される化合物またはその医薬
上許容される塩を、遊離塩基として計算して、0.01〜500mg/kg含有
するのが適当であり、好ましくは、0.1〜50mg/kg含有する。非経口、
鼻、経口吸入、経粘膜または経皮経路についての日用量は、式(I)で示される
化合物を0.01mg〜100mg/kg含有するのが適当である。局所製剤は
、式(I)で示される化合物を0.01〜5.0%含有するのが適当である。有
効成分は、当業者に容易に明白であるように所望の活性を示すのに充分な用量を
1日あたり1〜6回、好ましくは、1回投与してもよい。
本明細書で用いる場合、「モジュレーター」とは、アンタゴニストを意味する
。
本明細書で用いる場合、疾患の「治療」としては、限定するものではないが、
疾患の予防(prevention)、遅延および予防(prophylaxis)が挙げられる。
疾患細胞をベースとする治療または予防する疾患および障害としては、骨およ
びミネラル関連疾患または障害;上皮小体機能低下症;中枢神経系のもの、例え
ば、発作(seizure)、発作(stroke)、頭部外傷、脊髄損傷、心停止または新
生児ジストレスにおいて生じるような低酸素誘発神経細胞損傷、癲癇、アルツハ
イマー病、ハンチントン病およびパーキンソン病のような神経変性疾患、痴呆症
、筋肉緊張、鬱病、不安、パニック障害、強迫障害、心的外傷後ストレス障害、
精神分裂病、神経弛緩性悪性症候群およびツレット症候群;ADH分泌異常症候
群(SIADH)のような腎臓による過剰の水再吸収に伴う疾患、肝硬変、うっ
血性心不全およびネフローゼ;高血圧症;カチオン性抗生物質(例えば、アミノ
グリコシド抗生物質)からの腎毒性の予防および/または軽減;下痢および痙性
結腸のような腸運動障害;GI潰瘍疾患;サルコイドーシスのような過剰のカル
シウム吸収を伴うGI疾患;自己免疫疾患および臓器移植拒絶反応;扁平上皮癌
;ならびに膵炎が挙げられる。
本発明の好ましい具体例においては、本発明の化合物は、血清中上皮小体ホル
モン(「PTH)レベルを増加させるために用いる。血清中PTHレベルの増加
は、上皮小体機能低下症、骨肉腫、歯周疾患、骨折、変形性関節症、慢性関節リ
ウマチ、パジェット病、体液性高カルシウム血症悪性疾患および骨粗鬆症などの
疾患の治療に役立ち得る。
経口投与の場合に有効な式(I)で示される化合物およびそれらの医薬上許容
される塩は、シロップ剤、錠剤、カプセル剤およびロゼンジ剤として製剤化でき
る。シロップ製剤は、一般に、当該化合物または塩の、フレーバーリング剤また
は着色剤を有する、エタノール、落花生油、オリーブ油、グリセリンまたは水な
どの液体担体中懸濁液または溶液からなる。該組成物が錠剤の形態である場合、
固体製剤を調製するために慣用的に用いられるいずれの医薬担体を用いてもよい
。かかる担体の例としては、ステアリン酸マグネシウム、白土、タルク、ゼラチ
ン、アラビアガム、ステアリン酸、デンプン、ラクトースおよびスクロースが挙
げられる。該組成物がカプセル剤の形態である場合、例えば、ハードゼラチンカ
プセルシェル中で上記担体を用いるなど、いずれの慣用的なカプセル化も適して
いる。該組成物がソフトゼラチンシェルカプセル剤の形態である場合、例えば、
水性ガム、セルロース、シリケートまたは油などの分散液または懸濁液の調製に
慣用的に用いられるいずれの医薬担体であってもよく、ソフトゼラチンカプセル
シェルに一体化される。
典型的な非経口組成物は、所望により、例えば、ポリエチレングリコール、ポ
リビニルピロリドン、レシチン、落花生油またはゴマ油などの非経口上許容され
る油を含有していてもよい無菌水性または非水性担体中の化合物または塩の溶液
または懸濁液からなる。
典型的な吸入用組成物は、乾燥粉末として投与してもよい溶液剤、懸濁剤もし
くは乳剤の形態またはジクロロジフルオロメタンまたはトリクロロフルオロメタ
ンなどの慣用的な噴霧剤を用いるエアゾール剤の形態である。
典型的な坐剤製剤は、結合剤および/または滑沢剤、例えば、高分子グリコー
ル、ゼラチン、カカオ脂または他の低融点植物ワックスまたは脂肪またはそれら
の合成類似物と一緒にこのような投与の場合に有効な式(I)で示される化合物
またはその医薬上許容される塩からなる。
典型的な皮膚用および経皮用製剤は、慣用的な水性または非水性ビヒクル、例
えば、クリーム剤、軟膏剤、ローション剤またはペースト剤からなるか、または
、医薬用プラスター、パッチまたは膜の形態である。
好ましくは、該組成物は、患者が単回投与で投与してもよいような、単位投与
形態、例えば、錠剤、カプセル剤または定量型エアゾール剤である。
本発明に従って本発明の化合物を投与した場合、許容されない毒性作用は考え
られない。
式(I)で示される化合物の生物活性を以下の試験により示す:
(I)カルシウムレセプター阻害薬アッセイ
カルシウム溶解活性は、ヒトカルシウムレセプターを安定に発現しているHE
K293 4.0−7細胞中の細胞外Ca2+により誘起される細胞内Ca2+の増
加を遮断することについて試験化合物のIC50を測定することにより測定した。
HEK293 4.0−7細胞は、Rogersら,J.Bone Miner.Res.10Suppl.1:
S483,1995による記載(出典明示により本明細書の記載とする)に従って構築し
た。細胞内Ca2+増加は、細胞外Ca2+を1mMから1.75mMに増加させる
ことにより誘起した。細胞内Ca2+は、蛍光カルシウム指示薬であるfluo−
3を用いて測定した。
方法は、以下のとおりである:
1.T−150フラスコ中、37℃で5%CO2:95%空気の下、選択培地
(10%ウシ胎児血清および200ug/mLのハイグロマイシンBで補足した
DMEM)に細胞を維持し、集密度90%まで増殖した。
2.培地をデカントし、細胞単層を、37℃に維持したリン酸緩衝生理食塩水
(PBS)で2回洗浄した。2回目の洗浄後、PBS中0.02%EDTA 6
mLを添加し、37℃で4分間インキュベートした。インキュベーション後、穏
やかに攪拌することにより細胞を分散させた。
3.フラスコ2個または3個から得た細胞をプールし、ペレット化した(10
0×g)。細胞ペレットをSPF−PCB+10−15mLに再懸濁し、遠心分
離により再度ペレット化した。この洗浄を2回行った。
無硫酸無リン酸上皮小体細胞緩衝液(SPF−PCB)は、20mM Na−
Hepes(pH7.4)、126mM NaCl、5mM KClおよび1mM
MgCl2を含有する。SPF−PCBを調製し、4℃で保存した。使用日に、
SPF−PCBを1mg/mLのD−グルコースおよび1mM CaCl2で補足
し、2つのフラクションに分けた。一方のフラクションにウシ血清アルブミン(
BSA;フラクションV、ICN)を5mg/mLで添加した(SPF−PCB
+)。この緩衝液を細胞の洗浄、添加および維持のために用いた。無BSAフラ
クションは、蛍光測定用キュベット中での細胞の希釈のために用いた。
4.ぺレットを、2.2uM fluo−3(モレキュラー・プローブズ
(Molecular Probes))を含有するSPF−PCB+10mLに再懸濁し、室温
で35分間インキュベートした。
5.インキュベーション期間後、細胞を遠心分離によりペレット化した。得ら
れたペレットをSPF−PCB+で洗浄した。この洗浄後、細胞をSPF−PC
B+に細胞1−2×106個/mLの濃度で再懸濁した。
6.蛍光シグナルを記録するために、細胞懸濁液300uLを、1mMCaC
l2および1mg/mLのD−グルコースを含有するSPF緩衝液1.2mLで
希釈した。分光蛍光計を用いて、一定に攪拌しつつ、37℃で蛍光を測定した。
励起および発光波長を、各々、485nmおよび535nmで測定した。蛍光シ
グナルを検量するために、ジギトニン(エタノール中5mg/mL)を添力旺て
、Fmaxを得、Tris−EGTA(2.5MTris−Base、0.3M E
GTA)を添加することにより見掛のFminを測定した。細胞内カルシウムの濃
度を下記方程式:
細胞内カルシウム=(F−Fmin/Fmax)×Kd
(ここで、Kd=400nM)
を用いて計算した。
7.試験化合物の潜在的なカルシウム溶解活性を測定するために、細胞外Ca2+
の濃度を1mMから2mMに増加させる前に90秒間、細胞を試験化合物(ま
たは対照の場合はビヒクル)と一緒にインキュベートした。カルシウム溶解性化
合物は、濃度依存方法で、細胞外Ca2+により誘起される細胞内Ca2+の濃度の
増加を遮断するその能力により検出した。
一般に、カルシウムレセプター阻害薬アッセイにおけるIC50値が低いこれら
の化合物がより好ましい化合物である。IC50が50uMを超える化合物は、不
