抗癌剤としての2,4−ジアミノピリミジン化合物
本発明はras突然変異に関連するある種の癌の治療における、2,4−ジア
ミノピリミジン化合物の使用に関する。
葉酸拮抗剤、例えばメトトレキセートは、長年にわたって抗腫瘍剤として使用
されており、1954年にはメトプリン(式Aの化合物、ここにおいて、R1=C
l、R2=Cl、R3=Me)が臨床試験にはいった。
この薬剤には重度の毒性が伴うためさらなる評価には至らなかったが、ピリメ
タミン(式A、ここにおいて、R1=Cl、R2=H、R3=Et)の使用が探査され
、抗菌剤及び抗マラリア剤として固有の種選択性を持つ、ジアミノピリミジン化
合物が開発された。
WO88/04293(US4992444)はメトプリンと同等若しくはそ
れ以上にジヒドロ葉酸レダクターゼ(DHFR)を阻害し、比較的毒性の低いこ
の種の化合物について開示している。これらの化合物は、腫瘍、乾癬、並びに、
細菌性、マラリア性、及びトリパノソーマ性感染の治療に役立つ抗増殖剤として
の関心から記載され、DHFR阻害により作用することが言及されている。
これらの化合物のうち若干は、また、アジド置換ピリミジン誘導体に関係す
るWO84/04746において、中間体として開示されている。WO94/0
2469は他の関連化合物を開示しており、ここにおいて、R2が−N=N−N
R8R9のものは、特にAIDS付随感染の治療における使用が目的である。
本発明者らは、今回、これらジアミノピリミジン化合物群が、腫瘍のDHFR
阻害活性とは独立に、変異型−rasに関連した腫瘍に対する活性を有するとい
う点に関して、以前には確認されていなかった特性を持つことを明らかにした。
Ras、Rho、Rac、及びRabタンパク質はGTPアーゼモノマーの大
きな一群を形成している。Rasタンパク質は原形質膜の細胞質側に埋め込まれ
ており、受容体であるチロシンキナーゼから細胞核に信号を伝え、細胞分裂若し
くは分化を刺激する。
これらRasタンパク質はras遺伝子によって作られ、その変異体は細胞分
裂及び分化のこのコントロールを破壊することにより癌を促進する。人間の癌の
約30%が変異型−ras遺伝子を含むと考えられている。F. McCorm

系に対する活性が実証された。
本発明者らは今回、これらの化合物が、DHFR阻害活性の活性とは独立に、
変異型−ras癌細胞、特に非小細胞性肺癌(NSCL)、大腸癌、及び膵臓癌
の細胞に対して特異的な活性を持つことを決定した。このような活性は変異型r
as癌が存在することが推測されるか、若しくは確認された時に特別に使用され
る。
このように本発明の第一の側面は、一般式(I);

本発明の使用にあたって特に好ましい化合物は、式(VI):

上記の式(I)から(VI)のすべてにおいてアミノという単語は、一級、二
級、三級、及び四級アミノ基を含むが、好ましい基は無置換、一または二置換の
アミノ基であり、これらの中には複素環の一部を形成するアミノ基も含まれてお
り、特に3から7の炭素、窒素、硫黄、及び/又は酸素原子からなる環、あるい
は上記−NR6R7で定義されたように、より好ましくは炭素及び窒素原子のみか
らなる環である。アミノ基は一般に1つ若しくは2つのアルキルまたはアルアル
キル基により置換されており、ここで、アルキル基若しくは部は好ましくは1−
6炭素原子を含む。
R1及びR2は好ましくは無置換の一級アミノ基あるいはC1-4アルキル基で置
換された二級若しくは三級のアミノ基である。
ここにおいてR4,R5、R10,若しくはR11のうちの一つ又はそれ以上がアミ
ノ基であり、下記の基が特に興味深い:C1-4アルキルアミノ、ジ−C1-4アルキ
ルアミノ、C6-10アルアルキルアミノ、C1-4アルキルC6-10アルアルキルアミ
ノ、ジ−C6-10アルアルキルアミノ。
すべての上記基は置換されてもよく;例として、ハロゲン、若しくはC1-4ア
ルキル、特に塩素によるものである。基がベンゼン環を含むとき、例えばアルア
ルキルアミノ基であるが、この環は1回以上、好ましくは1又は2回、置換され
てよく、もっとも好ましくはハロゲン、例えばクロロで置換されてもよい。
R4,R5,R10,及びR11における特に好ましいアミノ基は、−NHCH3,
−NHC2H5、−NH(nC4H9)、−NHCH2CH2フェニル、−NHCH2
フェニル、−N(CH3)CH2フェニル、−N(CH2フェニル)2,N(C2H5
)CH2フェニル、及び−NHCH(CH3)フェニルから選択される。
アリール環上の置換基は適切にはハロゲン、C1-6アルキル若しくはハロアル
キル、C2-6アルケニル若しくはハロアルケニル、C1-6アルコキシ若しくはハロ
アルコキシ、ニトロ、若しくは−CO2R12であり、ここにおいてR12は水素、
C1-6アルキル、若しくはC1-6アルコキシアルキルである。これらのフェニル基
の4位を−CONHCH3置換すると、抗ras−活性の損失を招くと考えられ
るので好ましくないことが明らかとなった。しかし、ハロ、例えばクロ
ロ、特に3,4−ジクロロ置換は最高の活性を示し、例として、実施例の化合物
1を参照されたい。
このように、基がアルアルキルとして明示されているとき、それらは好ましく
はベンゼン環に結合したC1-4アルキレン基、例えば、フェニルメチル、2−フ
ェニルエチル、3−フェニルプロピル、若しくは4−フェニルブチルである。基
がハロアルキルと明示されているとき、それは好ましくはトリフルオロメチルで
ある。
R4がアルコキシ基であるとき、好ましくはC1-6アルコキシ基、OCH3、O
C2H5及び特に好ましくはO−nC4 H9である。
基R3は好ましくはC1-6アルキルで、好ましくはメチル若しくはエチルである
。
もっとも好ましい基R10は−NR12R13のような1若しくは2置換アミノ基で
あり、ここで、R12は水素原子若しくはC1-4アルキルであり、R13はアルアル
キル、もっとも好ましくは無置換若しくはハロ置換アルアルキルであり、さらに
もっとも好ましくはR10は−N(CH3)CH2フェニル、−N(CH3)CH2−
(3,4−ジクロロフェニル),−NH−CH2フェニル若しくは−NH−CH2
−(3,4−ジクロロフエニル)である。
R5及びR11はアジドもよい活性を導くが、もっとも好ましくはニトロである
。R10及びR11の一方がニトロ、もう一方がハロゲン、特にクロロである時、活
性は位置に関わらず良好である。同様な位置に独立な活性はアジドにも期待でき
る。
本発明に記述されているように、製造に使用される化合物はすべて、既存の方
法で得ることができ、ここに記述されている実施例はすべて既知化合物である。
WO88/04293(US4992444)に記載されている化合物は、本製
造で使用するための特別な化合物としてここでは引用されており、WO94/0
2469及びWO84/0446においてと同様、それらは上で開示された式に

本発明における第二の側面は、上記式(I)若しくは式(VI)に記載の化合
物の、効果的な投与量を患者に投与することを含む、好ましくはNSCL,大腸
、若しくは膵臓の変異型−rasに関連する癌患者の治療方法を提供する。
本発明の製造若しくは方法において使用されるこれらの化合物の量は、もちろ
ん、治療されるべき癌の種類及び個々の症例によって異なるであろう。投薬の形
態及び投与の経路は、上記の国際出願公開とその関連文献においてより深く考察
されており、例えば塩のような形態が投与に適している(例えばWO94/02
4696−9ページ参照)。対応するUS出願であるUS4992444及び継
続出願第08/374508号は、その目的及び本発明の方法において使用する
ための化合物調製を教示するために、参考文献として本明細書中に援用される。
このような化合物の投薬は簡便には、下記の実施例で示される、ras及び野生
型癌細胞に対して明示される相対的な効能に照らし合わせて評価しても良い。
