プロタンパク質変換酵素
発明の分野
本発明は、タンパク質加工酵素又はプロホルモンコンバターゼ(PCs)、特に
PC5に、とりわけヒトPC5に関する。
発明の背景
プロホルモンコンバターゼ(PCs)は、活性なタンパク質又は酵素へのタンパ
ク質前駆体の成熟の原因である酵素のファミリーに属する。これまで、そのファ
ミリーのヒトの酵素は、すなわちフリン、PC1、PC2、PC4及びPC7が
同定されている。各酵素は、制限され得る(例えば、PC4はオスの生殖細胞に
制限される)又は偏在する(フリンはそのような実例である)組織の分布を有する
。たとえこれら全ての酵素が塩基又は二塩基残基で前駆体タンパク質の分割の特
性を共有しても、それらは、それにも係わらず異なる分割特性を有する。特有な
プロホルモンコンバターゼの作用は、それ故に与えられたタンパク質基質の配列
の分割によって、及び/又は与えられたタンパク質の基質成長因子又はホルモン
に発現又は対応する組織中のその酵素の配置によって支配される。
レニンは、血管作用性のオクタペプチドアンギオテンシンII(AII)の合成
においてその鍵となる役割を通して心臓血管生理学及び病態生理学に重要な貢献
をなすアスパルチルプロテアーゼである。腎臓は、環状活性レニンの最初のソー
スである一方、脳下垂体及び副腎、胎盤、子宮、卵巣、精巣、心臓、血管系及び
脳を含む、幾つかの追加された組織はそのレニン遺伝子を発現する(1-4中参照)
。これらの組織においてRAS(レニン-アンギオテンシン系)の補足の成分の存
在は、アンキオテンシン変換酵素(ACE)とアンギオテンシンIIレセプターを
含め、たとえ各種のtRAsの実際の機能が依然として大部分が推測の事である
としても、ある種の組織が局部的に活性な組織レニン-アンギオテンシン系(tR
AS)を含み得るとの提案に至っている。
レニンは、アミノ末端基プロセグメントの43アミノ酸の蛋白分解性除去によっ
て活性レニンに変換される、酵素的に不活性な前駆体、プロレニンとして最初に
合成された。いずれかの与えられる組織内のRASの活性は、それ故に、プロレ
ニンの活性レニンへの変換の可能な蛋白分解性酵素の存在及びプロレニンを発現
する同じ細胞中のプロレニン加工酵素(PPEs)の発現とに基づくであろう。イ
ンビボでヒトのプロレニンを蛋白分解的に活性化するために応答可能な酵素の同
定は、依然として確定していない。さらに、多数のPPEsがヒトにおいて存在
することが可能であり、またレニン-生産組織の中で異なってよい。腎臓におけ
るレニンの生化学的及び顕微鏡的研究は、PPEs候補がプロセグメント5のLys42
、Arg43でのヒトブロレニンの分割のために選択するべきであり、且つ傍糸球
体細胞6の分泌顆粒において活性であろうことを示唆する。リソソーム酵素カテ
プシンBは、JG細胞の分泌顆粒中のヒトのレニン/プロレニン及びヒトの下垂
体ラクトトロフ7,8と共-配置されており、且つ固有分割サイトのために高い親和
性及び選択性によってインビトロでヒトのプロレニンを分割することを示してい
る9。プロホルモンコンバターゼPC1はまた、移入した細胞10において訂正サ
イト-及び小器官特異性を持ったヒトのプロレニンを分割すること及び副腎と誘
導した腫瘍11における、がJG細胞でない、レニンと共-配置することも示して
いる12。
新規なPPEsを同定するための努力において、我々は、オンコジーン-誘導
マウス腫瘍(As4.1細胞13)から誘導した確立されたレニン-発現組織培養細胞中の
加工酵素の分布を最近決定した。そのような酵素の一つ、マウスプロホルモンコ
ンバターゼPC5が見出された。マウスPC5はヒトのプロレニンの特異的な分
割が可能である。
ミランダ(miranda)ら(38)は、CD4+Tリンパ球からのPCRによって得られ
るヒトPC6酵素のためのcDNAとタンパク質の配列を記載する。PC5とP
C6は、同じ酵素がどのように出現するかによって異なる名称が与えられる。し
かしながら、ミランダらの配列は、現存するPC5配列に比べた場合に複数の置
換が含まれる。その上、PC6とPC5をコード化するメッセンジャーRNAの
サイズは類似するがしかし同一ではない。現存するPC5配列がヒトの副腎から
得られることから、両方の酵素は、イソ型とされてよく、組織中で特異に発現し
且つそれらは異なる活性を有してよい。
プロレニンとHIV gp160はたぶん、PC5によって認識され及び分割
されるための単なるタンパク質前駆体ではないであろう。細胞増殖に応答が可能
な多くの成長因子は、1又はそれ以上のPCsによって分割される。それらは、
血小板由来成長因子AとB(PDGF’s)、上皮増殖因子(EGF)、インシュ
リン様成長因子IとII(IGF’s)、トランスホーミング増殖因子αとβ(T
GF's)を含む。これら命名された成長因子の各々は、典型的な分割サイトモチ
ーフK/R-(X)n-R↓(式中n=0,2,4,6)を有する。完全な生物学的潜
在力は、これらのサイトにて、1又はそれ以上のPC酵素ファミリーによって分
割後、これらの成長因子に与えられる。それ故に、PCsの発現の操作が不完全
な成長因子活性を経て細胞増殖に影響を及ぼすであろう可能性がある。
経皮的経管的冠状動脈形成(PTCA)を受ける米国とカナダにおける>450
,000人の患者からの、30-50%の患者は、3−6ヶ月以内にそれの冠状
動脈が再狭窄するであろう。この内皮の及び平滑筋細胞増殖の再燃は非常にたく
さんの調節性成長因子によるものである。それ故に、酵素がこれの活性に応答可
能であるとの知見、及びこれ又はこれら酵素の発現のレベルを操作することは、
再狭窄を防止するために特に有用であろう。
発明の供述:
本発明は、ヒトのPC5(hPC5)に関する。我々は、ヒトの副腎から分離し
たhPC5が予想したサイト-及び小器官-特異性を持ったヒトのプロレニンを蛋
白分解的に活性化することを、及びそれが副腎皮質の球状体中のプリレニンと共
-発現されることを証明する。それ故に、PC5はプロレニン-加工酵素(PPE)
である。PC5の発現の沈黙化は、レニンの生産を抑制することにおいて特異的
な適用が見出されるであろうし、またレニンの生産を抑制するための方法は、本
発明の目的である。レニンの生産がRASの標的の一つであることから高血圧症
も包含される。その上、我々は、hPC5がアテローム硬化症の冠動脈炎におい
て過剰発現されることを証明する。アンチセンスオリゴヌクレオチドは、培養に
おける平滑筋細胞中のhPC5の発現を首尾よく沈黙化することが示されている
ものの中から設計されている。このアンチセンスはインビボでのウサギ頸動脈損
傷モデルにおいて頸動脈狭窄症を抑制した。これらの結果は、PC5の発現の鎮
静化方法が再狭窄の防止における特異的な適用を見出すであろうことを示す。
PC5は、フリン及びPC7と一緒にCD4+T細胞中に発現することが知ら
れる。3つの酵素がHIV gp160をその融合原性形態に変換することがで
きる。それ故に、アンチセンス構造、特に再狭窄を首尾よく抑制するオリゴヌク
レオチドは、HIV gp160に向けたPC5の活性の発現を抑制することの
使用が見出されるであろう。
hPC5の完全なアミノ酸及び拡散配列がこれより後に記載され、且つ本発明
の別な目的である。hPC5の全体又は部分を新たに挿入するように含んだ組換
えベクターと宿主もまた、本発明の目的である。
hPC5の蛋白配列から誘導されるオリゴペプチドもまた、本発明の目的であ
る。
hPC5全体のタンパク質又はそれの一部に対し指示した抗体もまた、本発明
の目的である。
オリゴヌクレオチド又は抗体結合P5拡散又はタンパク質又はペプチドを備え
る診断方法及びキットもまた、本発明の目的である。
本発明は、本発明の予期される態様の説明を目的とし、且つ本発明の範囲を限
定するものでない特有な実施態様、実施例及び図面によってこれより後に記載さ
れるであろう。

発明の説明
材料と方法
cDNAライブラリー構造とスクリーニング: トータルのヒト副腎RNAに
由来するcDNAライブラリーは、ファージベクターUni-Zap XRの中からStrata
gene(La Jolla,CA)により構築された。60万のファージプラークが放射能プロ
ーブと標準方法論14を用い、最初にスクリーニングした。最初のハイブリダイゼ
ーションプローブは、予めクローン化したマウスPC515との類似性の高い度合
を持ったジンバンク(Genbank)(Accession # M85522)中の朱同定ヒトcDNA配
列タグから誘導した情報を用いてヒト脳RNAの逆転写酵素PCRから誘導した
320塩基対DNAフラグメントである。フラグメントの標識化は、32P dC
TPとランダムプライマー標識キット(Boehringer-Mannheim Canada,Lavel,Qu
ebec,Canada)とを用い、説明書の指示に従い実行した。一つの陽性のハイブリ
ッド形成ファージ(hPC5A)が同定された。それの挿入は、ジデオキシ-鎖終
結法を用いそれの全体において配列決定され、且つマウスPC5に高度に類似し
た配列を持った1150塩基対cDNAをコード化することを見出した(データ
は示さず)。1070塩基対フラグメント(ポリAのテールを排除)がhPC5A
から削られ、cDNAライブラリーから追加した600,000ファージを再スクリー
ンするために標識され且つ使用される。第2のファージクローン(hPC5B)が
分離され且つ1807塩基対cDNA挿入オーバーラッピングhPC5AとcD
NAの5’末端に向かう拡張を含むことが見出される(図1)。
逆転写酵素PCR: ポリA+トータルのヒトの副腎からのRNA(Clontech
Laboratories,Palo Alto,CA)は、公知の方法16と以下のオリゴヌクレオチドを
用い、逆転写酵素ポリメラーゼ連鎖反応(RT-PCR)を行う:
前記のオリゴヌクレオチド;マウスPC5のシグナルペプチドに一致する領域か
ら誘導される15。人工のHindIII制限酵素分割サイトを、下線を付したクローニ
ングの目的のための増幅したフラグメントの5’-末端に加える:
5'-CCAAGCTTGGCTGCTGTGCGTGCTGGC-3'
逆オリゴヌクレオチド;ファージhPC5Bの5'-末端から誘導。内部のBgIII
制限酵素サイトは下線を付した:5'-CTGCCTCAGATCTGTAGTG-3'
全体のRT-PCR反応は、4回繰り返され且つ増幅したフラグメントの4つ
の独立して誘導したクローンが配列決定され且つその配列を比較した。ジンバン
ク(Genbank)に提出した配列(Accession # U49114)は、3/4クローンにおいて発現
するいずれかのヌクレオチドとして定義されるコンセンサス配列を表す。
ノーザンブロット分析: PC5 mRNAの組織分布が各種のヒトの組織(Cl
ontech Laboratories,Palo Alto,CA)からのポリ-A RNAのアリコート(2μ
g)を含む市販品を購入したニトロセルロースフィルターのハイブリッド形成に
よって決定される。用いたプローブは、完全長hPC5 cDNAから誘導した
相補RNAとした。プローブ標識化とハイブリッド形成は前述した通り実施した17
。
発現ベクター構造: KpnIサイト(図1)から停止コドン直後までのcDNAフ
ラグメントは、ファージhPC5Bから削られ、KpnIと2つの独立したRT-P
CRクローンの部分から誘導したHindIII(上記参照)フラグメントとを結合した(
Tagポリメラーゼから生じるエラーを消去するため)。mPC5から誘導した
シグナルペプチドの最初の16アミノ酸に一致する領域は、オーバーラップ拡張
PCRによって5'-末端に付着される18。かくして、cDNA全体、mPC5シ
グナルペプチドから誘導したコード化アミノ酸1−16及びhPC5からの残余
物は、RSVプロモーターのコントロールの下にcDNAを配置し且つウサギベ
ータグロブリン遺伝子からの3'イントロンとポリアデニル化シグナルを提供する
発現ベクターRSV-グロブリン19内にサブクローンされる。サブクローンされ
たフラグメント全体は、DNAシークェンシングによって続いて証明される。
細胞培養と移入:GH4Cl細胞はウエル当たり5x105細胞の濃度で6-ウ
エル培養皿内にプレートした。24時間の後、培地は交換され、その細胞は市販
キット(CellPect Transfection kit,Pharmacia Biotech,Bair D'urfe,Quebec
,Canada)を用い説明書の指導に従って、DEAE-デキストラン法により移入さ
れる。各ウエルは、hPC5発現ベクター又は中立ブラスミドベクター(pUCl8)
の0.18μgと、ヒトプロレニンのための発現ベクター又はプロレニンプロセ
グメントの42と43のアミノ酸がアラニンに変異したそれの同等物(それぞれ
、K/A-2とR/A-1)の0.18gのいずれかを与える。上清が移入の30時間後に
捕集され、上述した通りにプロレニンとレニンを分析した20。
分泌顆粒中に発生したプロレニンの変換を実証するため、ヒトのプリレニンと
hPC5発現ベクターとともに移入したGH4Clは、既に公知の技法21を用い
る脱分極化によって分泌顆粒を放出することにより刺激される。共-移入の40
時間後、移入した細胞の並行ウエル中の培地が、NaCl(コントロール)又はK
Cl(分泌促進物)のいずれかの50mモルの最終濃度に補充した予熱培地で置換
した。その培地は20分後に捕集し、レニン/プロレニンを分析した。細胞上清
中に含まれる活性レニンパーセンテージが増大するカリウム-従属は移入細胞の
分泌顆粒から放出される活性レニンの指標として扱われる。図5中に示す結果は
、3つの独立した移入実験の平均を表す。
ヒト組織におけるhPC5の免疫局在: ヒトの組織は、Bouin's溶液中で融
合し且つパラフィンで包理した、ポスト-モーテム(post-mortem)(腎臓と副腎)又
はポスト-パーツム(Post-partum)(胎盤分葉)を得た。免疫局在のために、5μm
切片がゼラチン被覆スライド上に搭載され、脱パラフィンされ、且つラットPC
5のN末端16アミノ酸に一致するペプチドに対して生起したポリクローナルウ
サキ抗血清(PC5.MAP抗体)の1:50希釈液又は組換えヒトプロレニンに対するポリ
クローナルウサギ抗血清の1:200希釈液と共にインキュベートした。腎臓と胎盤
標本のために、免疫複合体は、GhitescuとBendayanの方法22に従い、テトラクロ
ロ金酸(BDH)から合成したプロテインA-コロイダル金(15nm粒子)とのインキュベ
ーションによって現した。金粒子は、銀(IntenSETMM silver Enhancement Kit,
Amersham Life Science,Oakville,Ontario,Canada)とのインキュベートによ
って顕微鏡光中での視感を増大させ、且つ切片はヘマトキシリンとメチルグリー
ンによってカウンター着色した。ヒト副腎切片上の免疫複合体は、ビオチン-標
識化ロバライト-ウサギIgGの1:200希釈液と、ストレプトアビジン-ホースラ
デイッシュペルオキシダーゼ複合体(Amersham Life Science,Oakville,Ontari
o,Canada)の1:300希釈液によって検出し且つクロモゲンとして、ジアミノベン
ジジンと過酸化水素と共にインキュベートした。全ての陽性着色パターンは、最
初の抗体の省略によって特異性を実質的に証明した。
結果
ヒトPC5の最初の配列は図2に示される。我々は、RACEプロトコル16ま
たはマウスPC515,23の公知の配列に基づくオリゴヌクレオチドを用いること
のいずれかによってcDNAの先端の5'-末端をクローン化することができず、
この領域中のcDNAの公知の配列の高いG/C含有量のためかもしれない。し
かしながら、ラットとマウスPC515のための公知のcDNA配列に基づいて我
々は、シグナルペプチドの最初の12アミノ酸に一致するcDNAの先端5'-部
分を全てではないが分離していると確信した。マウスPC5の公知の配列との比
較によって、我々は分離したcDNAが、シグナルペプチドとそれぞれ32と8
4アミノ酸のプロセグメントを含む、915アミノ酸のプレプロPC5をコード
化するであろうと予測する。その推定されるhPC5の配列は、従来公知のマウ
スPC5cDNAと88%の相同であり、またマウスPC5タンパク質と96%
の相同である。
各種のヒトの組織からのポリARNAのノーザン分析は、ほぼ6.6Kbの主
要バンドとほぼ3.8Kbのマイナーバンドを示す(図3)。PC5RNAは脳、
心臓、胎盤、肺、甲状腺及び精巣中で、及び低レベルで骨格筋、腎臓及び膵臓、
小腸及び胃において検出される。副腎において、PC5は皮質中に得に豊富であ
る(図3)。
PC5 RNAが活性レニンを含むことが従来報告された多くの組織中に発現
することが明らかになったことから、我々は、細胞共-移入アッセイにおいてヒ
トのプロレニンを分割するためのhPC5能力を試験している(図4A)。従来報
告されているように10、培養したラットソマトトロフィックGH4Cl細胞がヒ
トプロレニンコード化発現ベクターと中立プラスミドベクターと共に共-移入し
た場合、加工していないプロレニンのみが培養上清内に分泌される。これに対し
、もしヒトプロレニン発現ベクターがヒトPC5コード化発現ベクターと共に共
-移入されるならば、発現したプロレニンの一部は活性レニンとして分泌される
。固有な分割サイト(リジン42またはアルギニン43)形成塩基残基のいずれか
で変異したプロレニンとヒトhPC5の共-発現は活性化を防止する。これらの
結果は、ヒトPC5が、ヒトにおけるレニンの活性化のための従来報告された5
サイトで蛋白分
解的な分割によってヒトのプロレニンを活性化することを示唆する。ヒトPC5
がGH4Cl細胞内でヒトのプロレニンを分割する一方、共-移入体がチャイニー
ズハムスター卵巣(CHO)細胞中に運ばれる際、活性レニン分泌が増加すること
は見出されていない(図4B)。GH4Cl細胞との比較としてCHO細胞ライン
中の明らかな相違は、ヒトPC5またはヒトプロレニンまたは両者のいずれかが
蛋白分解工程の間分布顆粒環境を要求することを示唆する、それの分泌顆粒の欠
落である。この結論は、塩化カリウム(図5)、分泌顆粒の放出の原因となる脱分
極剤21と20分間処理した共-移入したGH4Cl細胞の上清中に検出される活性
レニンの急激な増加によって裏付けられる(図5)。
マウスPC5から誘導したペプチドに対抗して生起したポリクローナル抗体を
用い、我々は幾つかのヒトの組織中でのヒトのPC5の分布を実験している(図
6)。今日までに、我々は、切片がレニンにより陽性に着色したとしても、ヒト
の腎臓においてPC5による着色を検出することはできていない。胎盤分葉にお
いて、PC5は絨毛膜絨毛の合胞体栄養細胞層中に局在し、抗-レニン抗体は絨
毛膜中胚葉を最初に着色する。副腎において、レニンとPC5の両方に対する抗
体は、カプセル(c)と束状帯(f)の非常に僅かな着色と共に、副腎皮質(g)中の球
状帯の優先的な着色を示した。着色なしは、第1の抗体の削除から明らかである
(データは示さず)。かくして、我々の免疫組織化学的研究は、3つの次式実験で
、それがプロレニンとPC5がヒトの副腎皮質の球状帯中で明確に共-局在され
るのみであると思われること示唆するであろう。
解説
本研究において、我々はヒトプロホルモンコンバターゼPC5のクローン化と
発現、及びヒトPPEとしてのそれの活性化を記載する。培養した細胞における
共-移入アッセイは、hPC5が予期したサイト-特異性を持ったヒトプロレニン
を活性化すること及びこの分割は濃密なコア分泌顆粒中で最も起こり得ることを
