前立腺ガンの治療において有用な共役体 発明の背景
1994年に、米国で200,000人の男性において前立腺ガンであると診
断されることが予想され、この病により38,000人のアメリカ人男性が死ぬ
であろう(Garnick,M.B.著(1994年),前立腺ガンのジレンマ(
The Dilemmas of Prostate Cancer),Sci
entific American,4月:72〜81)。すなわち、前立腺ガ
ンは米国の男性において最も頻繁に診断される悪性疾患(皮膚の悪性疾患以外で
)であり、この群においてガンにより死ぬ第2番目の原因(肺ガンに次ぐ)であ
る。
前立腺特異的抗原(PSA)は、殆どもっぱらヒト前立腺上皮により生成され
る一本鎖33kDaグリコタンパクであり、ヒトの精液において0.5〜2.0

PSAエピトープが完全に失われるので、この複合体形成

較して、血清中の未反応α1−抗キモトリプシンおよびα2−マクログロブリン

第57巻:50〜55頁)ことから、前立腺転移が免疫学的反応性PSAを分泌
することも知られている。従って、PSAのタンパク分解活性により活性化され
得る細胞毒性化合物は、前立腺細胞特異的であると共にPSA分泌前立腺転移に
特異的であるはずである。
本発明の目的は、溶解性を増加させるオリゴペプチド封鎖基を有するオリゴペ
プチドを、細胞毒性剤と連結されて有する、前立腺ガンの治療に有用な新規抗ガ
ン組成物を提供することである。
本発明のもう一つの目的は、新規抗ガン組成物を投与することを含んでなる前
立腺ガンの治療方法を提供することである。発明の概要
遊離前立腺特異性抗原(PSA)により選択的にタンパク分解されるアミノ酸
配列を有するオリゴペプチド、親水性オリゴペプチド封鎖基および既知の細胞毒
性剤を含む化学的共役体を開示する。そのような共役体は、前立腺ガンおよび良
性前立腺肥大(BPH)の治療に有用である。発明の詳細な説明
本発明は、前立腺ガンの治療に有用な新規抗ガン組成物に関
する。そのような組成物は、細胞毒性剤に直接または化学的リンカーを介して共
有結合しているオリゴペプチドを含む。オリゴペプチドは、遊離前立腺特異的抗
原(PSA)により選択的に認識されるオリゴマーから選択され、遊離前立腺特
異的抗原の酵素活性によりタンパク分解されるようなものである。そのようなオ
リゴペプチドと細胞毒性剤の組み合わせを共役体と呼ぶ。
本発明の共役体は、共役体の水溶性に寄与する親水性封鎖基をオリゴペプチド
のN−末端において有することによりさらに特徴付けられる。そのような親水性
封鎖基の例は、ヒドロキシル化およびポリヒドロキシル化アルカノイル部、およ
びエーテル官能基を組み込むアルカノイル部を含むが、これらに限定されるわけ
ではない。
理想的には、細胞毒性剤の細胞毒性活性は、PSAタンパク分解部を含むオリ
ゴペプチドが細胞毒性剤に直接または化学的リンカーを介して結合したとき、大
きく低下するか存在しなく、かつ無傷のままである。また、理想的には、開裂位
置において付着オリゴペプチドがタンパク分解されると、細胞毒性剤の細胞毒性
活性が大きく増加するかまたは非修飾細胞毒性剤の活性
に戻る。
さらに、オリゴペプチドは、遊離酵素的活性PSAの存在下におけるオリゴペ
プチドの開裂と比較して、非PSAタンパク分解酵素の存在下においてかなり遅
い速度で開裂されるまたは開裂されないオリゴペプチドから選択されることが好
ましい。
前記理由から、オリゴペプチドは、好ましくは10アミノ酸朱満である、短い
ペプチド配列を含むことが望ましい。最も好ましくは、オリゴペプチドは7アミ
ノ酸以下を含む。共役体は好ましくは短いアミノ酸配列を含むので、共役体の溶
解性は、細胞毒性剤成分の通常の疎水性により大きく影響されよう。従って、本
発明の共役体の親水性封鎖基は、細胞毒性剤によるそのような疎水性の寄与を除
くかまたは減衰させるように選択される。
本発明のこの観点を実施するのに必ずしも必要はないが、本発明の好ましい態
様は、オリゴペプチドおよび存在する場合は化学的リンカーが遊離PSAおよび
近くの組織に存在する他の天然タンパク分解酵素のタンパク分解活性により細胞
毒性剤から分離され、それにより非修飾細胞毒性剤がタンパク分解部位において
生理学的環境中に放出されるような、共役体である。
共役体の薬学的に許容される塩も含まれる。
細胞毒性剤に連結しているオリゴペプチドは、直接共有結合であろうとまたは
化学的リンカーを介してであろうと、遊離PSAにより最も良く認識され遊離P
SAにより最も容易にタンパク分解されるオリゴペプチドである必要はないこと
がわかる。すなわち、このような抗ガン組成物中に組み込むために選択されたオ
リゴペプチドは、遊離PSAによる選択的タンパク分解と、そのような分解から
生じる細胞毒性剤−タンパク分解残基共役体(または、理想と思われる状況にあ
っては、非修飾細胞毒性剤)の細胞毒性活性との両方について選択される。タン
パク分解PSA開裂に関連して用いられる「選択的」という用語は、アミノ酸の
ランダム配列を含むオリゴペプチドの開裂と比べて、遊離PSAによる本発明の
オリゴペプチド成分の大きな開裂速度を意味する。従って、本発明のオリゴペプ
チド成分は、遊離PSAの好ましい基質である。「選択的」という用語は、オリ
ゴペプチド中の二つの特異的アミノ酸の間においてオリゴペプチドが遊離PSA
によりタンパク分解されることも意味する。
本発明のオリゴペプチド成分は、遊離前立腺特異的抗原(P
SA)により選択的に認識され、遊離前立腺特異的抗原の酵素的活性によりタン
パク分解され得る。そのようなオリゴペプチドは、以下のものから選択されるオ
リゴマーを含む:

ペプチド化学分野の当業者は、生物学的活性オリゴペプチド
中の特定のアミノ酸を、原オリゴペプチドの生物学的活性が修飾オリゴペプチド
中に保存されるようにして、他の同族、等立体構造及び/又は等電的アミノ酸に
より置換し得ることを容易に理解しよう。本発明のオリゴペプチド中の対応する
天然アミノ酸を置換するために特定の非天然および修飾天然アミノ酸も利用する

本発明の共役体は、N−末端アミノ酸が親水性封鎖基により修飾されているオ
リゴマーを含む。そのような封鎖基は、親水性官能基の存在により選択される。
親水性官能基の存在は、本発明の共役体を、N−末端封鎖基をすでに有する先に
開示された共役体から区別される。そのような末端アミノ基の封鎖は、温血動物
の血漿中に存在するエキソアミノペプチダーゼの作用によるペプチド治療剤の酵
素的分解を低減または除去すること
もできる。共役体の親水性を増加させ、それにより共役体の水溶性を増加させる

本発明の共役体は、非対称中心を有することができ、ラセミ体、ラセミ混合物
、および全ての可能な異性体を有する個々のジアステレオマーとして得られ、本
発明に含まれる光学異性体を含む。いずれかの変化(例えばアリール、ヘテロ環
、R3等)が任意の要素において一回を超えて起こる場合、各々の場合の定義は
他のものから独立している。例えば、HO(CR1R2)2−は、HOCH2CH2

つまでの環員を有する安定な単環式または二環式炭素環を意味し、ここで少なく
とも一つの環は芳香族である。そのようなアリール要素の例は、フェニル、ナフ
チル、テトラヒドロナフチル、インダニル、ビフェニル、フェナンスリル、アン

ここで用いられるヘテロサイクルまたはヘテロ環式という用語は、飽和または
不飽和で、炭素原子およびN、OおよびSからなる群より選択される1〜4のヘ
テロ原子からなる安定な5〜7員単環式または安定な8〜11員二環式ヘテロ環
式環を意味し、任意の前記ヘテロ環式環がベンゼン環に融合している二環式基を
含む。ヘテロ環式環は、任意のヘテロ原子または炭素原子に結合し、それにより

細胞毒性剤は所望の生物学的活性を有するタンパクまたはポリペプチドであって
よい。そのようなタンパクは、例えば、アブリン、リシンA、シュードモナス外
毒素またはジフテリア毒素のような毒素;腫瘍壊死因子、α−インターフェロン
、β−インターフェロン、神経成長因子、血小板誘導成長因子、組織プラスミノ
ーゲンアクチベーター、のようなタンパク;または例えば、リンフォカイン、イ
ンターロイキン−1(「IL−1」)、インターロイキン−2(「IL−2」)

好ましい細胞毒性剤は、一般的に、アルキル化剤類、抗増殖剤類、チューブリ
ン結合剤類等を含む。好ましい種類の細胞毒性剤は、例えば、アントラサイクリ
ンファミリーの薬剤類、ビンカ薬剤類、マイトマイシン類、ブレオマイシ類、細
胞毒性ヌクレオシド類、タキサン類、プテリジンファミリーの薬剤類、
ジーネン(diynenes)類、およびポドフィロトキシン類を含む。これら
のうち特に有用なものは、例えば、ドキソルビシン、カルミノマイシン、ダウノ
ルビシン、アミノプテリン、メトトレキサート、メトプテリン、ジクロロ−メト
トレキサート、マイトマイシンC、ポルフィロマイシン、5−フルオロウラニル
、6−メルカプトプリン、シトシンアラビノシド、ポドフィロトキシン、または
ポドフィロトキシン誘導体、例えば、エトポシドまたはエトポシドホスフェート
、メルファラン、ビンブラスチン、ビンクリスチン、ロイロシジン、ビンデシン

本発明のオリゴペプチド、ペプチドサブユニットおよびペプチド誘導体(「ペ
プチド類」とも呼ぶ)は、成分であるアミノ酸から、従来のペプチド合成技術に

PSA開裂部位および細胞毒性剤を含むオリゴペプチドを含む本発明の共役体
は、同様に、医科学分野において良く知られている技術により合成することがで
きる。例えば、細胞毒性剤上の遊離アミン部を、カルボキシル末端においてオリ
ゴペプチドに共有結合させてアミド結合を形成することができる。同様に、オリ
ゴペプチドのアミン部と細胞毒性剤のカルボキシル部を共有結合することにより
アミド結合を形成することができる。これらの目的のために、2−(1H−ベン

等のような試薬を組み合わせて、または単独で使用することができる。
さらに、PSA開裂部位と細胞毒性剤との間の非ペプチド結合により本発明の
共役体を形成することができる。例えば、細胞毒性剤は、細胞毒性剤上のヒドロ
キシル部を介してオリゴペプチドのカルボキシル末端に共有結合させて、それに
よりエステル結合を形成することができる。この目的のために、HBTUとHO
BTの組み合わせ、BOPとイミダゾールとの組み合わせ、DCCとDMAPと
の組み合わせ等のような試薬を利用することができる。カルボン酸を、ニトロ−
フェニルエステル等を形成することにより活性化し、DBU(1,8−ジアザビ
シクロ[5,4,0]ウンデク−7−エンの存在下に反応させることもできる。
本発明の共役体は、オリゴペプチドをリンカー単位を介して細胞毒性剤に結合
させることにより形成することもできる。そのようなリンカー単位は、例えば、
ビスカルボニルアルキルニ価基を含み、それにより細胞毒性剤上のアミン部がリ
ンカー単位と結びつけられてアミド結合が形成され、オリゴペプチドのアミノ末
端がリンカー単位の他の端部と結び付けられて同様に
アミド結合が形成される。逆に、ジアミノアルキル二価リンカー単位も有用であ
り得、それにより細胞毒性剤上のカルボニル部がリンカー部のアミンの一つに共
有結合し、結合部の他のアミンはオリゴペプチドのC末端に共有結合する。遊離
PSAが存在しない場合に生理学的環境に安定であるが、PSAタンパク分解部
の開裂時に分解する他のそのようなリンカー単位も考えられる。さらに、PSA
タンパク分解部の開裂時に細胞毒性剤に結合したままであるがそのような開裂後
細胞毒性剤誘導体の細胞毒性活性が非修飾細胞毒性剤と比較して大きく低下しな
いリンカー単位を利用することができる。
当業者は、本発明の化合物の合成時に、分子の他の部分において所望の反応を
行うときに出発化合物および中間体の種々の反応性官能基を保護する必要がある
ことを理解する。所望の反応の完了後、または任意の所望の時間に、通常、その

実施例 実施例1 PSA媒介間裂部位を含むオリゴペプチドの調製
封鎖ドオリゴペプチドを、アプライド・バイオシステムズ(Applied
Biosystems)モデル430A自動ペプチド合成器においてアミノ酸導
入のためのダブルカップリングプロトコールを用いて、固相合成により調製した
。オリゴペプチドの脱保護および樹脂支持体からの除去は、液状フッ化水素酸で
の処理により達成した。オリゴペプチドを、逆相C18シリカカラム上の分取高
圧液体クロマトグラフィーにより水性0.1%トリフルオロ酢酸/アセトニトリ
ル勾配を用いて精製した。オリゴペプチドの同一性および同質性は、アミノ酸組
成物分析、高圧液体クロマトグラフィー、および高速原子衝突質量分光分析によ
り確認した。この方法により調製したオリゴペプチドを表2に示す。
実施例2 遊離PSAによるオリゴペプチドの認識の評価
実施例1に記載のように調製したオリゴペプチドを個々にPSA消化緩衝液(
12mMトリス(ヒドロキシメチル)アミノメタン、pH8.0、25mM N
aCl、0.5mM CaCl2)に溶解し、溶液をPSAにモル比100:1
で添加した。反応を、種々の反応時間後に、トルフルオロ酢酸(TFA)を最終
的に1%(体積/体積)となるように添加することによ
り停止する。反応停止反応液を、逆相C18カラム上のHPLCで水性0.1%
TFA/アセトニトリル勾配を用いて分析した。評定の結果を表2に示す。表2
は、示したオリゴペプチドを酵素的活性遊離PSAで50%開裂するのに必要な
時間(分)を示している。遊離アミン部を含む(すなわち、hArg、Orn、
Lys及び/又は3PALを含む)オリゴペプチドを、TFA塩として試験した
。全ての他のオリゴペプチドは中性化合物として試験した。
実施例3 N−(2−ヒドロキシアセチル)−Ser−Ser−Ser−Chg−Gln− Ser−Leu−Dox(3−3)の調製 工程A
: 2−HO−Ac−Ser(Bzl)−Ser(Bzl)−Ser(B zl)−Chg−Gln−Ser−Leu−PAM樹脂(3−1)

Iペプチド合成器上で保護ペプチドを合成した。プロトコールは、次の保護アミ

Cl)に溶解し、溶液をPSAにモル比100:1で添加した。反応液を、種々
の反応時間後に、トルフルオロ酢酸(TFA)を最終的に1%(体積/体積)と
なるように添加することにより停止する。反応停止反応液を、逆相C18カラム
上のHPLCで水性0.1%TFA/アセトニトリル勾配を用いて分析した。評
定の結果を表3に示す。表3は、示したオリゴペプチド−細胞毒性剤共役体を酵
素的活性遊離PSAで50%開裂するのに必要な時間(分)を示している。共役

共役体の評価に利用した。実施例8 ドキソルビシンのペプチジル誘導体の細胞毒性の生体外アッセイ
非修飾ドキソルビシンにより殺されることが知られている細胞系に対する、実
施例3〜6に記載のように調製した開裂性オリゴペプチド−ドキソルビシン共役
体の細胞毒性をアラマー・ブルー(Alamar Blue)アッセイを用いて評
価した。特に、96ウエルプレート中のLNCap前立腺腫瘍細胞またはDuP
RO細胞の細胞培地を、種々の濃度の所定の共役体を含む媒体で希釈した(最終
的プレートウエル体積:200μl)。細胞を37℃で3日間インキュベートし
、アラマー・ブルー20μlをアッセイウエルに添加する。細胞をさらにインキ
ュベートし、アッセイプレートをEL−310ELISAリーダー上でアラマー
ブルーの添加後4時間および7時間において570nmおよび600nmの二重
波長で読み取った。次に、試験した共役体の種々の濃度における相対的生存率を
、対照(非共役体)培地と比較して計算した。このアッセイの結果を表4に示す

LNCaP.FGCまたはDuPRO−1細胞をトリプシン処理し、成長培地
中に再懸濁させ、200×gで6分間遠心分離する。細胞を血清−遊離α−ME
M中に再懸濁させ、計数する。次ぎに、所望数の細胞を含むこの溶液の適当な体
積を、円錐遠心分離管に移し、前述のように遠心分離し、α−MEM−マトリク
