誘導体の(S)−エナンチオマーの使用
本発明は、1,4-ジピドロピリジンの(S)-エナンチオマーの調製、心不全の処置
におけるそれらの使用およびそのような処置のための医薬製造におけるそれらの
使用に関する。
カルシウム拮抗活性を有する1,4-ジヒドロピリジンは、幾つかの心臓血管疾患
、例えば、高血圧およびアンギナの処置に広く使用されている。これらの化合物

本発明によると、化合物Iの(S)-エナンチオマーは、患者にそのままで、ある
いは薬学的に許容されるその塩、水和物または溶媒和物のいずれかの形態で投与

本発明の方法では、化合物IIの(R)-エナンチオマーは化合物IIIを用いてエス
テル化される。反応は、周知の合成方法:アルバートソン(Albertson)、Org.Re
act.、12、205-218(1962);ドハティー(Doherty)ら、J.Med.Chem.、35、2(199
2);スタアブ(Staab)ら、Newer Methods Prep.Org.Chem.、5、61(1968);イ
シハラ(Ishihara)、Chem.Pharm.Bull.、39、3236(1991)に従い、カップリング
剤(例えば、ジシクロヘキシルカルボジイミド、N,N’-カルボニルジイミダゾー
ルまたはジエチルシアノホスホナート)の存在下に、非プロトン性溶媒または塩
素化溶媒(例えば、ジメチルホルムアミドまたはクロロホルム)中、必要に応じ
で促進剤(例えば、N-ヒドロキシスクシンイミドまたは4-ジメチルアミノピリジ
ン)の存在下に、-10〜140℃の範囲の温度で、行われ得る。
或いは、化合物IIの(R)-エナンチオマーは、まず第三級アミン(例えば、トリ

これらの方法のどれが使用されても、得られる化合物Iのホモ-キラルな(S)-エ

それらの薬理学的活性を評価するために、(S)-エナンチオマーを、イヌでイン

レート・ハイドロクロリド.0.5H2O.
0.13mlの塩化チオニルを、-10℃で、2.9mlの無水ジクロロメタンおよび0.75ml
の無水ジメチルホルムアミド中0.54gのR-(-)-1,4-ジヒドロ-2,6-ジメチル-4-(3-
ニトロフェニル)-5-メトキシカルボニル-ピリジン-3-カルボン酸[エー.アシモ
リ(A.Ashimori)ら、Chem.Pharm.BuII.、39、108-111、(1991)に記載されるよ
うに調製した]の攪拌懸濁液に入れ、窒素雰囲気下で維持し、直射光を遮った。
0℃で1時間後に、1mlのジクロロメタン中0.48gの2,N-ジメチル-N-(3,3-ジフェニ
ルプロピル)-1-アミノ-2-プロパノール溶液(米国特許第4,772,621号に記載され
るように調製した)を-5℃で加えた。3時間0℃で攪拌し、20-25℃で終夜静置し
た後、溶媒を真空で留去し、残渣を20mlの酢酸エチルに溶解した。有機相を、ブ
ライン(4ml)、10%炭酸ナトリウム水溶液(5x4ml)、
ブライン(4ml)、1N塩酸(5x5ml)、ブライン(4ml)、10%炭酸ナトリウム水溶液(2
x5ml)および最後にブライン(4ml)で、次々に洗浄した。有機相を、無水硫酸ナ
トリウムで乾燥し、真空で蒸発乾固した。残渣を、石油エーテル:アセトン85:1
5で溶出するシリカゲル・カラム上でのフラッシュ・クロマトグラフィーにより

イドロクロリド
N-メチル-3,3-ジフェニルプロピルアミン・ハイドロクロリド(2.61g)、無水酢
酸(1ml)およびホルムアルデヒド(水中37%、0.9ml)を含む混合物を、30分間還流

薬理試験
実施例1の化合物のラセミ化合物と比較するために、実施例1および3の(S)-
エナンチオマーを、それらのインビボでの薬理学的活性についてイヌで試験した

うに調製した]の攪拌懸濁液に入れ、窒素雰囲気下で維持し、直射光を遮った。
0℃で1時間後に、1mlのジクロロメタン中0.48gの2,N-ジメチル-N-(3,3-ジフェニ
ルプロピル)-1-アミノ-2-プロパノール溶液(米国特許第4,772,621号に記載され
