核酸センサー
発明の分野
本発明は、サンプル中の選択された核酸配列の存在の検出に使用するためのバ
イオセンサーおよびサンプル中の選択された核酸(NA)配列の存在を検出する方
法に関する。
発明の背景
サンプル中の特有なDNAまたはRNA配列の存在のためのアッセイは多くの
分野における適用を有する。例えば、特異的な微生物を持った患者の感染は、そ
の微生物に特有なNA配列の存在のために患者からの生物学的サンプルの分析に
より評価することができる。別の適用は、食品または環境のサンプルの不純物を
検出するための分析を含む。突然変異によって生じる遺伝病の検出もまた、これ
らの技法によって達成することができる。
これまで、このより高い感受性の方法の普及した使用は、特異的な核酸配列の
量の顕著な増殖化の要求のために及び/又は増殖したDNAを分析するための特
殊化された技法を用いることの要求のため、制限されていた。この増殖化は一般
に、ポリメラーゼ連鎖又はリガーゼ連鎖反応を用いて達成され、さらに増殖され
たDNAはゲル上で分離され、そのゲルがDNAの特異的なバンドの存在により
分析される。
別な方法は、核酸検出の方法を教示する米国特許第4,840,893号に含まれる。
該方法は接合されたリガンドを持ったプローブ配列、第2のリガンドに結合し且
つ該酵素が触媒的に活性である時に電極表面に電荷を転移することができる酵素
メディエーター系(enzyme mediator system)、及び該第1と第2のリガンドと結
合を競うアンチリガンドを用いる。該アッセイは競合系(competition system)で
あり、サンプル中の標的配列の結合は第1のリガンドの能力に影響を及ぼし且つ
電極への電荷転移の割合を変える。本発明は、酵素触媒によって発生した信号に
頼らない、直接ゲート化メカニズム(direct gating mechanism)を提供すること
の効
果を有する。
米国特許第4,868,104号は、重合化することができる第1の及び検出系を有す
る第2の、標的分析物上の異なる配列に結合する、2つのポリヌクレオチド試薬
の使用によってヌクレオチド配列を検出する方法を記載する。核酸検出方法を記
載する幾つかの他の特許は、大部分がプローブの標識化を含む。これらは:米国
特許第4,968,602号;米国特許第5,116,733号;米国特許第4,868,105号;米国特
許第4,716,106号;米国特許第4,883,750号;米国特許第5,242,794号;来国特許
第4,996,142号;米国特許第5,348,855号及び米国特許第5,053,326号を含む。

発明の詳細な説明
本発明者は、サンプル中の特異的なNA配列の存在を検出するために使用する
ことができる装置を提案する。その装置は、それが脂質膜の使用を含むバイオセ
ンサーとして最もよく記載される。バイオセンサーにおいて用いるための膜は、
国際特許出願 Nos PCT/AU88/00273,PCT/AU/89/00352,PCT/AU90/00025及びPCT/
AU92/00132中に開示されている。これらの出願の開示は参考によってここに包含
される。
これらの出願中に開示された通り、好適に修正された脂質分子は、電極/イオ
ンレザバー/隔離二層の、すなわちイオンチャンネルとイオノフォアの合併のた
めに好適である組合せの中に構築することによるであろう。それはまた、これら
の膜のコンダクタンスが分析物(analyte)の存在又は不存に基づいていることも
開示される。二層膜におけるイオンチャンネルを含む各層において、その膜のコ
ンダクタンスは、膜にのびる連続的なイオンチャンネルを形成するためのそれぞ
れの層中のモノマーの整列に基づく。これらの連続的なイオンチャンネルが恒常
的
に形成され及び破壊されるように、その膜のコンダクタンスはこれらの連続的な
イオンチャンネルの寿命に依存される。
第1の態様において、本発明は、サンプル中の選択された核酸配列の存在を検
出することにおいて用いるためのバイオセンサーであり、該バイオセンサーは電
極と、上層及び下層を有する二層膜を備え、該下層は該膜と該電極の間に空間を
存在させるような手法において該電極に近接され且つ結合され、該膜のコンダク
タンス又はインピーダンスは該選択された核酸配列の存在又は不存に基づいてお
り、該膜は両親媒性分子の密に充填されたアレイと、該上層中に拡散された第1
の半膜スパンニングモノマーと該下層中に拡散された第2の半膜スパンニングモ
ノマーとを含む多数のイオンチャンネルを含み、該第1の半膜スパンニングモノ
マーは該上層内に横方向に拡散でき且つ該第2の半膜スパンニングモノマーは該
下層内の横方向に拡散するのを防ぎ、該選択された核酸配列と特異的に反応する
第1のリガンドが該膜の表面に近接した第1の半膜スパンニングモノマーの部分
の先端に付着し、且つ該選択された核酸と反応する第2のリガンド又はそれに付
着したマーカーが該膜の表面に近接した第1の半膜スパンニングモノマーの残部
の先端に付着している。
本発明の好ましい実施態様において、該膜は、その膜内の横方向の拡散を妨げ
る膜スパンニング両親媒体(membrane spanning amphiphiles)を含む。
第2の態様において、本発明は、サンプル中の選択された核酸配列の存在を検
出することにおいて用いるためのバイオセンサーであり、該バイオセンサーは電
極と、上層及び下層を有する二層膜を備え、該下層は該膜と該電極の間に空間を
存在させるような手法において該電極に近接され且つ結合され、該膜のコンダク
タンス又はインピーダンスは該選択された核酸配列の存在又は不存に基づいてお
り、該膜は両親媒性分子の密に充填されたアレイ、該膜内の横方向に拡散するの
を防ぐ膜スパンニング両親媒体及び該上層中に拡散された第1の半膜スパンニン
グモノマーと該下層中に拡散された第2の半膜スパンニングモノマーとを含む多
数のイオンチャンネルを含み、該第1の半膜スパンニングモノマーは該上層内に
横方向に拡散でき且つ該第2の半膜スパンニングモノマーは該下層内の横方向に
拡散するのを防ぎ、該選択された核酸配列と特異的に反応する第1のリガンドが
該膜の表面に近接した第1の半膜スパンニングモノマーの少なくとも一部の先端

第3の態様において本発明は、サンプル内の選択された核酸配列の存在を検出
する方法であり、該方法は上記第1の又は第2の態様のバイオセンサーに該サン
プルを加えることを備える。
当業者によって理解されるであろうように、本発明のバイオセンサーと方法は
、イオンチャンネルを経て膜を横切るためのイオンの能力の変更を生じさせる第
1及び第2のリガンドへの選択された核酸配列の結合によって特徴付けられる。
その検出の特異性は、選択された核酸配列のための第1のリガンドの特異性によ
って提供される。この特異性は多くの手法により提供されてよいが、しかしなが
ら、それは、選択された核酸の中の第1の配列に相補的な配列を含む核酸分子又
はPNAを備える第1のリガンドの使用によりこれが達成されることが目下のと
ころ好適である。理解されるであろうように、第1のリガンド内の配列は、感受
性の要求されるレベルを提供するために十分な何れかの長さ、典型的には少なく
とも10残基である。その核酸分子は修正された塩基を含んでよいこともまた、
理解されるであろう。
その特異性は第1のリガンドによって最初に提供されるように、第2のリガン
ドは選択された核酸配列のために特異的であることは必須でなく、その第2のリ
ガンドが核酸又はそれに付着したマーカーと結合することが単に必要とされる。
従って、該第2のリガンドはDNAに対して指示する抗体、またはそれの結合フ
ラグメントであってよい。第2のリガンドはまた、挿入剤、主溝又は小溝バイン
ダー、三重螺旋形成可能な作因(agent)又はDNAに共有結合できる作因または
それの組合せであってもよい。第2のリガンドは、それぞれの選択されたすなわ
ち検出されるべき核酸配列のために作製される必要のあるただ一つの特異的なリ
ガンドと同じくこれが該バイオセンサーの簡単な構築の結果になる選択された核
酸配列のために特異的でないことが好適である。
該バイオセンサーはまた、該バイオセンサーを越えて変わる第2のリガンドの
多様性を含んでよい。例えば、膜スパンニング両親媒物に付着する第2のリガン
ドは、別の膜スパンニング両親媒物に付着する第2のリガンドの異なる部分であ
ってよい。その必須の特徴は、核酸に又はそれに付着したマーカーに結合した第
2のリガンドである。従って、用語「第2のリガンド」は、核酸又はそれに付着
したマーカーに結合する部分の単独の又は複数の種におよぶものと解される。
多数の適用におけるその分野の当業者によって予期されるであろうように、検
出されるべき核酸配列はマーカー、例えばビオチニル化PCR生成物で標識され
るであろう。
それは、しかしながら、第1及び第2のリガンドの両方が、選択された核酸配
列のために特異的であることが本発明の範囲内であることも明らかである。その
ような実施態様において、第1と第2のリガンドは選択された核酸配列の中の異
なる配列によって特異的である。
第1及び第2のリガンドは、多数の手法のいずれかで付着させてよい。例えば
ストレプトアビジンは上記した出願中に記載された通り付着されてよい。これは
ビオチニル化核酸を結合させるために使用することもできる。第1及び第2のリ
ガンドは、リンカーを経てイオンチャンネル又は膜スパンニング脂質に付着させ
ることが好適である。リンカーを使用する場合、該リンカーはホスホラミダイト
(phosphoramidite)基であることがさらに好適である。
そのリンカー長の変更は、バイオセンサーにおいて検出されたゲート化応答に
影響を及ぼしてよいことも、予期される。
第1及び第2のリガンドが核酸配列である場合、これは、中央におけるRNA
配列とそれぞれの端でリガンドとして作用するものである2つのDNA配列を有
する分子の生成によって有利には達成されてよい。その分子の2つの端は、バイ
オセンサーの表面に付着され、さらにその主要なRNA部分は付着した2つの分
離したDNA配列をバイオセンサーから離すためにRNaseHで消化される。
好適な実施態様において、第1のリガンドは、選択された核酸の中の第1の配
列に相補の配列を含む核酸分子を備える。第2のリガンドは、選択された核酸配
列内の第2の配列に相補の配列を含む核酸分子を備えることがさらに好適である
。
そのような相補の配列の存在のため、第1と第2のリガンドは、ハイブリッド形
成によって選択された核酸に結合する。
第1と第2のリガンドは、サンプル中の選択された核酸配列の異なる領域に結
合するために選択したオリゴヌクレオチド配列であることが目下の処好適である
。国際特許出願 Nos PCT/AU88/00273,PCT/AU88/00273,PCT/AU89/00352,PCT/A
U90/00025及びPCT/AU92/00132に記載されたインピーダンスブリッジを用いて特
異的なNA配列を検出する又は感知するための方法は、例えばビオチニル化膜ス
パンニング脂質(MSL)とビオチニル化グラミシジンの両方を持った膜を作製す
ることによって達成されてよい。ビオチンに強力に結合するストレプトアビジン
は、これらのビオチニル化化合物との反応によって固定化される。
2つのビオチニル化オリゴヌクレオチド配列の混合物(第1と第2のリガンド)
は次いでストレプトアビジンに付着される。これらの配列は、相補でないことが
好適である。関係のある配列のために適当なプローブであろう1つの配列、例え
ば感染性生物プローブ、及びその他の配列は、標的DNAの別の部分に相補とさ
れるだろう。
検出すべきDNAの添加に関して、2つのビオチニル化配列の架橋が、もしそ
のDNAが2つの付着した、しかし異なっていない配列にハイブリダイズする配
列を含むならば生じる。
第4の態様において本発明は、サンプル内の選択された核酸の存在の検出方法
であり、該方法は、以下の工程:
(i)電極と上層及び下層を有する二層膜を備え、該下層は該膜と該電極の間に
空間を存在させるような手法において該電極に近接され且つ結合され、該
膜のコンダクタンス又はインピーダンスは該選択された核酸配列の存在又
は不存に基づいており、該膜は両親媒性分子の密に充填されたアレイと、
該上層中に拡散された第1の半膜スパンニングモノマーと該下層中に拡散
された第2の半膜スパンニングモノマーとを含む多数のイオンチャンネル
を含み、該第1の半膜スパンニングモノマーは該上層内に横方向に拡散で
き且つ該第2の半膜スパンニングモノマーは該下層内の横方向に拡散する
のを防ぎ、第1の核酸配列と特異的に反応する第1のリガンドが該膜の表
面に近接した第1の半膜スパンニングモノマーの部分の先端に付着し、且
つ第2の核酸配列と特異的に反応する第2のリガンドが該膜の表面に近接
した第1の半膜スパンニングモノマーの残部の先端に付着し、該第1又は
第2の核酸配列のいずれか一方は選択された核酸の配列内にある、バイオ
センサーを用意すること;
(ii)該バイオセンサーに、選択された核酸を含むことが推測されるサンプルを
加えること、及び該膜のコンダクタンス及び/又はインピーダンスを測定
すること;及び
(iii)(ii)からバイオセンサーに対抗核酸を加えること、及び該膜のコンダ
クタンス及び/又はインピーダンスを測定すること、該対抗配列はそれが
第1及び第2の核酸配列の両方を含むように選択される、
を備える。
もしその標的核酸がサンプル中に存在するならば、その対抗配列は、第1及び
第2のリガンドの両方に結合することかできず、かくて架橋する。(ii)と(iii)
において、コンダクタンス/インピーダンス測定における相違の不存は、サンプ
ル中の選択された核酸の存在を示している。
第5の態様において本発明は、サンプル内の選択された核酸の存在の検出方法
であり、該方法は以下の工程:
(i)電極と上層及び下層を有する二層膜を備え、該下層は該膜と該電極の間に
空間を存在させるような手法において該電極に近接され且つ結合され、該
膜のコンダクタンス又はインピーダンスは該選択された核酸配列の存在又
は不存に基づいており、該膜は両親媒性分子の密に充填されたアレイ、該
膜内の横方向に拡散するのを防ぐ膜スパンニング両親媒体及び該上層中に
拡散された第1の半膜スパンニングモノマーと該下層中に拡散された第2
の半膜スパンニングモノマーとを含む多数のイオンチャンネルを含み、該
第1の半膜スパンニングモノマーは該上層内に横方向に拡散でき且つ該第
2の半膜スパンニングモノマーは該下層内の横方向に拡散するのを防ぎ、
該選択された核酸配列と特異的に反応する第1のリガンドが該膜の表面に
近接した第1の半膜スパンニングモノマーの少なくとも一部の先端又は該
膜の表面に近接した該膜スパンニング両親媒体の先端に付着し、且つ該選
択された核酸と反応する第2のリガンド又はそれに付着したマーカーが該
膜の表面に近接した第1の半膜スパンニングモノマーの該少なくとも一部
以外又は該膜の表面に近接した該膜スパンニング両親媒体の先端に付着し
た、バイオセンサーを用意すること;
(ii)該バイオセンサーに、選択された核酸を含むことが推測されるサンプルを
加えること、及び該膜のコンダクタンス及び/又はインピーダンスを測定
すること;及び
(iii)(ii)からバイオセンサーに対抗核酸を加えること、及び該膜のコンダ
クタンス及び/又はインピーダンスを測定すること、該対抗配列はそれが
第1及び第2の核酸配列の両方を含むように選択される、
を備える。
もしその標的核酸がサンプル中に存在するならば、その対抗配列は、第1及び
第2のリガンドの両方に結合することができず、かくて架橋する。(ii)と(iii)
において、コンダクタンス/インピーダンス測定における相違の不存は、サンプ
ル中の選択された核酸の存在を示している。
第6の態様において本発明は、サンプル中の選択された核酸配列の存在を検出
することにおいて用いるためのバイオセンサーであり、該バイオセンサーは電極
と上層及び下層を有する二層膜を備え、該下層は該膜と該電極の間に空間を存在
させるような手法において該電極に近接され且つ結合され、該膜のコンダクタン
ス又はインピーダンスは該選択された核酸配列の存在又は不存に基づいており、
該膜は両親媒性分子の密に充填されたアレイと、該上層中に拡散された第1の半
膜スパンニングモノマーと該下層中に拡散された第2の半膜スパンニングモノマ
ーとを含む多数のイオンチャンネルを含み、該第1の半膜スパンニングモノマー
は該上層内に横方向に拡散でき且つ該第2の半膜スパンニングモノマーは該下層
内の横方向に拡散するのを防ぎ、該選択された核酸配列と特異的に反応する第1
のリガンドが該膜の表面に近接した第1の半膜スパンニングモノマーの部分の先
端に付着し且つ第2のリガンドが該膜の表面に近接した該第1の半膜スパンニン
グモノマーの残部の先端に付着し、該第2のリガンドは該第1のリガンドが該第
2のリガンドに結合するように該選択された核酸配列のそれに結合され或いはそ
の中に含まれている。
第7の態様において本発明は、サンプル中の選択された核酸配列の存在を検出
することにおいて用いるためのバイオセンサーであり、該バイオセンサーは電極
と、上層及び下層を有する二層膜を備え、該下層は該膜と該電極の間に空間を存
在させるような手法において該電極に近接され且つ結合され、該膜のコンダクタ
ンス又はインピーダンスは該選択された核酸配列の存在又は不存に基づいており
、該膜は両親媒性分子の密に充填されたアレイ、該膜内の横方向に拡散するのを
防ぐ膜スパンニング両親媒体及び該上層中に拡散された第1の半膜スパンニング
モノマーと該下層中に拡散された第2の半膜スパンニングモノマーとを含む多数
のイオンチャンネルを含み、該第1の半膜スパンニングモノマーは該上層内に横
方向に拡散でき且つ該第2の半膜スパンニングモノマーは該下層内の横方向に拡
散するのを防ぎ、該選択された核酸配列と特異的に反応する第1のリガンドが該
膜の表面に近接した第1の半膜スパンニングモノマーの少なくとも一部の先端又
は該膜の表面に近接した該膜スパンニング両親媒体の先端に付着し、且つ第2の
リガンドが該膜の表面に近接した第1の半膜スパンニングモノマーの該少なくと
も一部以外又は該膜の表面に近接した該膜スパンニング両親媒体の先端に付着し
、該第2のリガンドは該第1のリガンドが該第2のリガンドに結合するように該
選択された核酸配列のそれに結合され或いはその中に含まれている。
第6と第7の態様において特異化された配置において、該バイオセンサーは競
合によって選択された核酸配列の存在を検出する。その選択された核酸配列は、
もし試験するべきサンプル中に在るならば、第1のリガンドに結合するための第
2のリガンドと競合する。
第8の態様において本発明は、サンプル中の選択された核酸配列の存在を検出
することにおいて用いるためのバイオセンサーであり、該バイオセンサーは、電
極と、上層及び下層を有する二層膜を備え、該下層は該膜と該電極の間に空間を
存在させるような手法において該電極に近接され且つ結合され、該膜のコンダク
タンス又はインピーダンスは該選択された核酸配列の存在又は不存に基づいてお
り、該膜は両親媒性分子の密に充填されたアレイと、該上層中に拡散された第1
の半膜スパンニングモノマーと該下層中に拡散された第2の半膜スパンニングモ
ノマーとを含む多数のイオンチャンネルを含み、該第1の半膜スパンニングモノ
マーは該上層内に横方向に拡散でき且つ該第2の半膜スパンニングモノマーは該
下層内の横方向に拡散するのを防ぎ且つ該選択された核酸配列と特異的に反応す
る第1のリガンドが該膜の表面に近接した第1の半膜スパンニングモノマーの部
分の先端に付着している。
選択された核酸配列を含むサンプルの付加に関するこの構成において、第1の
リガンドは選択されたDNA配列に結合するであろうし、且つ第1の半膜モノマ
ーの移動性を変化させ又は該チャンネルを通るイオン流を制限するであろう。こ
れは、異なるコンダクタンスを持った膜の結果を与えるであろう。
本発明の他の好適な実施態様において、第1及び第2の半膜スパンニングモノ
マーはグラミシジン又はその誘導体の一つである。本発明のさらに好適な実施態
様において、二層膜は、膜と電極の間に空間が存在するようにリンカー分子(lin
king molecules)を経て電極に付着される。好ましいリンカー分子は、出願PCT/A
U92/00132中に開示されたそれらである。本発明のよりさらに好適な実施態様に
おいて、第2の半膜スパンニングモノマーはリンカー基を経て電極に付着される
。
本発明のさらに別の好適な実施態様において、二層膜は、膜スパンニング脂質
、アルカエバクテリア(archaebacteria)中に見出されるそれらと同じ、を含む。
「架橋」は、第2の半膜スパンニングモノマーによる整列の外に第1の半膜ス
パンニングモノマーを運ぶ架橋配列によって特徴付けされる。これは、第1のコ
ンダクタンスとは異なる第2のコンダクタンスを有する膜を生じる。コンダクタ
ンスにおけるこの相違は測定が可能であり且つサンプル中の選択されたDNA配
列の存在の証拠となるであろう。
第1と第2のリガンドは同じであっても又は異なっていてもよい。特異的なN
A配列に結合するオリゴヌクレオチドリガンドは、当該技術分野で周知である。
本発明の代替の実施態様において、該リガンドはペプチド核酸(PNA)である
。PNAに関する情報は、Science,1991,254,1497 1500;J.Am.Chem.Soc.
,1992,114,1895 1897,J.Am.Chem.Soc.,1992,114,9677 78;J.Chem.S
oc.Chem.Commun.,1993,800 801.Nature,1993,365,566,68;米国特許第4,
795,700号と国際特許出願No.WO 93/18187中に見出されるものでよく、さらにこ
れらの文献の開示は参考によってここに併合される。これらは、リガンドとして
PNAの使用によって多くの効果が得られる。これらは、二重鎖DNAにおいて
特異的な配列を認めるための及び鎖置換(strand displacement)又は三重螺旋構
造によるいずれか一方のそれに結合するためのPNAの能力を含む。これは、単
鎖又は二重鎖との結合が可能であるが鎖置換によって結合できないオリゴヌクレ
オチドと対照をなす。PNAは単鎖DNAに結合することも可能であることに注
目すべきである。その標的プローブ配列は緊密な結合のためにPNA分子とする
ことができる。
代替の実施態様において、二つのビオチニル化オリゴヌクレオチド配列の混合
物はストレプトアビジン上に固定化される。一つは標的プローブ配列であり第2
は選択のいずれかの適当な架橋配列である。サンプルの添加によって、標的分析
物は捕捉される。その標的分析物は、関係のある配列を有することのみが要求さ
れる。架橋は起こらず、それ故にゲート化(gating)は観測されず、さらにコンダ
クタンスの第1の測定が実行される。第2のオリゴヌクレオチド配列は、そのシ
ステムに対抗するために用いられる。この対抗配列は、標的オリゴヌクレオチド
配列と架橋配列と相補の配列の両方を含む。もし両方のエピトープの結合が生じ
ると、架橋が起こるであろうし、かくしてイオンチャンネルがゲートオフになり
(gating off)、さらに膜のコンダクタンスが変化する。もしエピトープ2つでの
み結合か生じると(標識DNAの、そのプローブ配列への結合のために)、架橋が
起こることがなく、かくしてゲート化が見られることはない。対抗配列の付加に
続いて起こるコンダクタンスにおける変化の不存は、それ故に標的分析物の存在
の証拠である。
NAプローブ配列は特に、いずれかの長さとすることができ且つ検出のための
標的分析物に相補の塩基配列を含むであろう。その標的分析物はDNA又はRN
A配列とすることができる。PNAプローブ配列もまた、いずれかの長さとする
ことができ且つ検出のための標的分析物に相補の塩基配列を含むであろう。その
標的分析物はPNAが両方に結合するようなDNA又はRNA配列とすることが
できる。PNAはDNAよりもより緊密にDNAとRNAと結合し、故に該対抗
配列は既に結合された標的配列と置換できないものとすべきである。
標的配列の幾つかの実施例が下記に示される。

コレラ菌(Vibrio cholerea c)の全てのコレラ毒素生産株は23塩基対オ
リゴヌクレオチド配列とともに特にハイブリッド形成する(水検出、健康)

The membrane comprises a densely packed array of amphiphilic molecules, a first semi-membrane spanning monomer diffused into the upper layer and a second semi-membrane spanning monomer diffused into the lower layer. If binding of both epitopes occurs, crosslinking will occur, thus gating off the ion channel and further altering the conductance of the membrane. If binding occurs only at two epitopes (due to the binding of the labeled DNA to its probe sequence), no cross-linking occurs and thus no gating is seen. The absence of a change in conductance following the addition of the countersequence is therefore evidence of the presence of the target analyte.
