ウィルス粒子を細胞にターゲッティングするアダプター分子
本発明は、ウィルス粒子を細胞表面分子に結合し易くするアダプター分子に関
する。本発明はまた、アダプター分子をコードする核酸、アダプター分子を発現
する細胞系統、および遺伝子治療におけるアダプター分子の使用に関する。
序論
遺伝子治療の共通した目標は、現在もっともよく用いられている厄介なex viv
o法ではなく、in vivoで遺伝子を送達可能にすることである。in vivoで送達す
る技術の開発において、送達系を特定の細胞型にターゲッティングすることによ
って遺伝子を選択的に送達することが可能であればかなり有利となる。本発明は
、レトロウィルスベクターを用いて、ターゲッティングされる遺伝子の送達を達
成するための新規の方法を述べる。
近年、レトロウィルスベクターのターゲッティングにおいていくらかの好結果
が得られた。ヒトを対象とした用途に使用される現在のレトロウィルスベクター
は、多様な種の広範囲の細胞型を感染させる両種指向性(amphotropic)エンベロ
ープタンパク質を担持する点で無差別である。これは、実験室において使用され

向させるために該SU成分がなんらかの方法で改変されていることである。エンベ
ロープタンパク質の構造および機能が変化に対して非常に敏感であるために、こ
の種のエンベロープ改変が有効なターゲッティング遺伝子導入に導き得ないこと
は明らかである。
従って、レトロウィルスベクターから標的細胞まで遺伝物質を送達する改良し
た方法が必要となる。本発明はそのようなニーズに応えることを目的とする。
本発明
本発明に従って、同種指向性受容体を特定の細胞型にターゲッティングし、非
改変の同種指向性ベクターを使用して遺伝子をこれらの細胞に送達する、代替的
かつ新規のアプローチを述べる。これは、エンベロープタンパク質が、マウス細
胞に感染する場合と同様の結合および融合事象を生じるという相当な利点を有す
る。
本発明は1つの態様において、細胞表面分子に対して結合親和性を有する第一
のアミノ酸配列と、ウィルス表面分子に対して結合親和性を有する第二のアミノ

本発明のアダプター分子中の第一のアミノ酸配列は、細胞表面分子に対して特
異的な親和性を有するいかなるアミノ酸配列でもよい。例えば、抗体または抗体
誘導体、天然リガンドまたは天然リガンドの誘導体、あるいはファージ展示(pha
ge display)等の種々のあらゆる公知の親和性選択システムによって選択された
ペプチドであり得る。アダプター分子への使用に適した抗体誘導体の例として、
一本鎖抗体(scFv)が挙げられる。scFvは、あらゆる特定の細胞表面分子をターゲ
ッティングするために使用され得る。特に有用なscFvは、腫瘍特異的細胞表面抗
原および組織特異的細胞表面タンパク質に対して結合親和性を有するものである
。
本明細書において記載する第一および第二のアミノ酸配列は、グリコシル化等
の、タンパク質およびペプチドに関連した通常の修飾を伴い得ることが理解され
るであろう。
また、本発明のアダプター分子においては、第一のアミノ酸配列がカル
ボキシ末端に、第二のアミノ酸配列がアミノ末端に、あるいは第一のアミノ酸配
列がアミノ末端に、第二のアミノ酸配列がカルボキシ末端に位置し得ることが理
解されるであろう。
本発明のアダプター分子は、記載のようにウィルス粒子による細胞の感染を仲
介し得る。本明細書において使用する「感染」という用語は、ウィルス粒子が細
胞表面に付着し、遺伝物質がウィルス粒子から細胞に導入されることを意味する
。このように、アダプター分子は、形質導入の過程、すなわちウィルス仲介遺伝
子導入を助けることができる。
第一および第二のアミノ酸配列は、アダプター分子において共有結合的または
非共有結合的に結合されている。共有結合的に結合されている場合、アダプター
分子は、単一の融合遺伝子から、または化学的架橋によって生産され得る。会合
が非共有結合的である場合、両成分はストレプトアビジン−ビオチン架橋、また
