ル)プロピオニル}−3−ピペリジルカルボニル]−2(S)−アセチルアミノ
−β−アラニン
技術分野
この発明はβ−アラニン誘導体に関する。より詳しくは、この発明は、糖蛋白
IIb/III拮抗剤、血小板凝集阻害剤およびフィブリノーゲンの血小板への結
合阻害剤であるβ−アラニン誘導体に関する。
背景技術
ヨーロッパ特許出願No.512,831 A1には、フィブリノーゲン受容
体拮抗剤が開示されている。
ヨーロッパ特許出願No.445,796 A2には、血小板凝集阻害剤が開
示されている。
発明の開示
この発明はβ−アラニン誘導体に関する。より詳しくは、この発明は、糖蛋白
IIb/IIIa拮抗剤および血小板凝集阻害剤であるβ−アラニン誘導体に関し
、
動脈血栓などの血栓形成;動脈硬化;虚血性心疾患[たとえば狭心症(たとえ
ば安定狭心症、切迫梗塞を含む不安定狭心症など)、心筋梗塞(たとえば急性心
筋梗塞など)、冠動脈血栓など];虚血性脳疾患[たとえば脳梗塞{たとえば脳
血栓(たとえば急性脳血栓など)、脳塞栓症など}、一過性脳虚血(たとえば一
過性虚血発作など)、脳出血後の脳血管攣縮(たとえばクモ膜下出血後の脳血管
攣縮など)など];肺血管疾患(たとえば肺血栓、肺塞栓など);末梢循環障害
[たとえば閉塞性動脈硬化、閉塞性血栓血管炎(すなわちバージャー病)、レイ
ノ病、糖尿病の合併症(糖尿病性血管症、糖尿病性ニューロパシーなど)、静脈
血栓(たとえば深部静脈血栓など)など]などを要因とする疾患の予防および/
または治療用の薬剤;
再狭窄および/または再閉塞、たとえば経皮経管冠動脈形成術(PTCA)後
の再狭窄および/または再閉塞、血栓溶解剤(たとえば組織プラスミノーゲン活
性剤(TPA)など)の投与後の再狭窄および/または再閉塞などの予防および
/または治療用の薬剤;
血栓溶解剤(たとえばTPAなど)または抗凝血剤(たとえばヘパリンなど)
を用いる合併療法用の薬剤;
血管手術、弁置換、体外循環[たとえば外科手術(たとえば開心術、人工心肺
など)、血液透析など]、移植などの場合における血栓形成の予防および/また
は治療用の薬剤;
汎発性血管内凝固(DIC)、血栓性血小板減少、本態性血小板増加、炎症(
たとえば腎炎など)、免疫病などの予防および/または治療用の薬剤;
転移阻害用の薬剤;などとして有用である。
この発明のβ−アラニン誘導体は、細胞接着の阻害剤として有用であると予想
され、さらに汎発性血管内凝固(DIC)、血栓性血小板減少、本態性血小板増
加、炎症(たとえば腎炎など)、免疫病などの予防および/または治療用の薬剤
;
転移阻害用の薬剤;
などとして有用である。
この発明の目的β−アラニン誘導体は下記の式(I)
で表すことができる。
目的化合物(I)は下記の諸方法によって製造することができる。製造法1 製造法2 (上記各式中、R1はアミノ保護基である。)
化合物(II)および(III)の好適な塩は、慣用の無毒の塩などの医薬と
して許容される塩であって、金属塩たとえばアルカリ金属塩[たとえばナトリウ
ム塩、カリウム塩など]およびアルカリ土類金属塩[たとえばカルシウム塩、マ
グネシウム塩など];アンモニウム塩;有機塩基塩[たとえばトリメチルアミン
塩、トリエチルアミン塩、ピリジン塩、ピコリン塩、ジシクロヘキシルアミン塩
、N,N’−ジベンジルエチレンジアミン塩など];有機酸付加塩[たとえば蟻
酸塩、酢酸塩、トリフルオロ酢酸塩、マレイン酸塩、酒石酸塩、メタンスルホン
酸塩、ベンゼンスルホン酸塩、トルエンスルホン酸塩など];無機酸付加塩[た
とえば塩酸塩、臭化水素酸塩、ヨウ化水素酸塩、硫酸塩、燐酸塩など];アミノ
酸との塩[たとえばアルギニン塩、アスパラギン酸塩、グルタミン酸塩など];
などを挙げることができる。
この明細書の以上および以下の記述において、種々の定義の好適な例を次に詳
細に説明する。
「低級」とは、特記ない限り、炭素原子数1ないし6を意味する。
「高級」とは、特記ない限り、炭素原子7ないし20個を有する基を意味する
。
好適な「低級アルキル基」としては、直鎖または分岐状のもの、たとえばメチ
ル、エチル、イソプロピル、プロピル、ブチル、イソブチル、第二級ブチル、第
三級ブチル、ペンチル、イソペンチル、ヘキシル、イソヘキシルなどを挙げるこ
とができる。
好適な「アミノ保護基」としては、下記のアシル基、適当な置換基を1ないし
3個有していてもよいアル(低級)アルキル(たとえばベンジル、フェネチル、
1−フェニルエチル、ベンズヒドリル、トリチルなど)などの慣用の保護基、[
5−(低級)アルキル−2−オキソ−1,3−ジオキソール−4−イル](低級
)アルキル[たとえば(5−メチル−2−オキソ−1,3−ジオキソール−4−
イル)メチルなど]などを挙げることができる。
好適な「アシル基」および「アシル」としては、脂肪族アシル;芳香族アシ
ル;アリール脂肪族アシル;およびカルボン酸、炭酸、カルバミン酸から誘導さ
れた複素環脂肪族アシルなどを挙げることができる。
前記「アシル基」の好適な例としては、下記のものを挙げることができる。
脂肪族アシルとしては、たとえば
低級または高級アルカノイル(たとえばホルミル、アセチル、プロパノイル、
ブタノイル、2−メチルプロパノイル、ペンタノイル、2,2−ジメチルプロパ

よび「複素環基」としては、酸素原子、硫黄原子、窒素原子などのヘテロ原子を
少なくとも1個有する、飽和または不飽和の単環または多環複素環基を挙げるこ
とができ、より好ましい複素環基としては、下記の複素環基
窒素原子1ないし4個を有する3ないし8員(好ましくは5または6員の)不
飽和複素単環基、たとえばピロリル、ピロリニル、イミダゾリル、ピラゾリル、
ピリジル、ジヒドロピリジル、ピリミジル、ピラジニル、ピリダジニル、トリア

前記のアシル部分としては、1ないし10個の同種または異種の適当な置換基
、たとえば
低級アルキル(たとえばメチル、エチル、プロピルなど);
低級アルコキシ(たとえばメトキシ、エトキシ、プロポキシなど):
低級アルキルチオ(たとえばメチルチオ、エチルチオなど);
低級アルキルアミノ(たとえばメチルアミノ、エチルアミノ、プロピルアミノ
など);
シクロ(低級)アルキル[たとえばシクロ(C3−C6)アルキル(たとえばシ
クロペンチル、シクロヘキシルなど)];
シクロ(低級)アルケニル[たとえばシクロ(C3−C6)アルケニル(たとえ
ばシクロヘキセニル、シクロヘキサジエニルなど);
ハロゲン(たとえばフッ素、塩素、臭素、ヨウ素);アミノ;前記のアミノ保
護基;ヒドロキシ;後記の保護されたヒドロキシ;シアノ;ニトロ;カルボキシ
;保護されたカルボキシ;スルホ;スルファモイル;イミノ;オキソ;
アミノ(低級)アルキル(たとえばアミノメチル、アミノエチルなど);カル
バモイルオキシ;ヒドロキシ(低級)アルキル(たとえばヒドロキシメチル、1
または2−ヒドロキシエチル、1または2または3−ヒドロキシプロピルなど)
などを有していてもよい。
好適な「保護されたヒドロキシ基」としては、前記のアシル、適当な置換基を
1個以上有していてもよいフェニル(低級)アルキル(たとえばベンジル、4−
メトキシベンジル、トリチルなど)、三置換されたシリル[たとえばトリ(低級
)アルキルシリル(たとえばトリメチルシリル、第三級ブチルジメチルシリルな
ど)など]、テトラヒドロピラニルなどを挙げることができる。
「アミノ保護基」のさらに好ましい例としては、低級アルコキシカルボニルま
たはアル(低級)アルコキシカルボニルを、最も好ましいものとしては、第三級
ブトキシカルボニルまたはベンジルオキシカルボニルを挙げることができる。製造法1
目的化合物(I)は、化合物(II)またはその塩をアミノ保護基の脱離反応
に付すことによって製造することができる。
この反応は加水分解、還元などの慣用の方法で実施される。
加水分解は、塩基、またはルイス酸などの酸の存在下で実施されることが好ま
しい。
好適な塩基としては、無機塩基および有機塩基、たとえばアルカリ金属[たと
えばナトリウム、カリウムなど]、アルカリ土類金属[たとえばマグネシウム、
カルシウムなど]、それらの水酸化物または炭酸塩または重炭酸塩、トリアルキ
ルアミン[たとえばトリメチルアミン、トリエチルアミンなど]、ピコリン、1
,5−ジアザビシクロ[4.3.0]ノン−5−エン、1,4−ジアザビシクロ
[2.2.2]オクタン、1,8−ジアザビシクロ[5.4.0]ウンデク−7
−エンなどを挙げることができる。
好適な酸としては、有機酸[たとえば蟻酸、酢酸、プロピオン酸、トリクロロ
酢酸、トリフルオロ酢酸など]および無機酸[たとえば塩酸、臭化水素酸、硫酸
、塩化水素、臭化水素など]を挙げることができる。
三ハロゲン化酢酸[たとえばトリクロロ酢酸、トリフルオロ酢酸など]などの
ルイス酸を用いる脱離は、カチオン捕捉剤[たとえばアニソール、フェノールな
ど]の存在下で行うのが好ましい。
この反応は、通常、反応に悪影響を及ぼさない溶媒、たとえば水、アルコール
[たとえばメタノール、エタノールなど]、塩化メチレン、テトラヒドロフラン
、それらの混合物、または他の溶媒中で行われる。液体の塩基または酸もまた溶
媒として使用できる。反応温度は特に限定されず、通常、冷却ないし加温下で行
われる。
脱離反応に適用される還元方法としては、化学還元および触媒還元を挙げるこ
とができ、化学還元に用いられる好適な還元剤としては、金属[たとえば錫、亜
鉛、鉄など]または金属化合物[たとえば塩化クロム、酢酸クロムなど]と有機
酸または無機酸[たとえば蟻酸、酢酸、プロピオン酸、トリフルオロ酢酸、p−
トルエンスルホン酸、塩酸、臭化水素酸など]との組合せを挙げることができる
。
触媒還元に用いられる好適な触媒としては、白金触媒[たとえば白金板、白金
海綿、白金黒、コロイド白金、酸化白金、白金線など]、パラジウム触媒[たと
えばパラジウム海綿、パラジウム黒、酸化パラジウム、パラジウム炭、コロイド
パラジウム、パラジウム−硫酸バリウム、パラジウム−炭酸バリウムなど]、ニ
ッケル触媒[たとえば還元ニッケル、酸化ニッケル、ラネーニッケルなど]、コ
バルト触媒[たとえば還元コバルト、ラネーコバルトなど]、鉄触媒[たとえば
還元鉄、ラネー鉄など]、銅触媒[たとえば還元銅、ラネー銅、ウルマン銅など
]などの慣用のものを挙げることができる。
還元は、通常、反応に悪影響を及ぼさない慣用の溶媒、たとえば水、メタノー
ル、エタノール、プロパノール、N,N−ジメチルホルムアミドなど、またはそ
れらの混合物中で行われる。さらに、化学還元で用いられる前記の酸が液体であ
る場合、それらもまた溶媒として使用できる。また、触媒還元で用いられる好適
な溶媒は、前記の溶媒、および他の慣用の溶媒、たとえばジエチルエーテル、ジ
オキサン、テトラヒドロフランなど、またはそれらの混合物であってもよい。
この還元の反応温度は特に限定されず、通常、冷却ないし加温下で行われる。
上記のようにして得られた目的化合物(I)が塩の形態である場合、それは慣
用の方法で遊離形態に変換できる。製造法2
目的化合物(I)は、化合物(III)を脱塩反応に付すことによって製造す
ることができる。
この反応は、中和、再結晶、脱塩樹脂カラムクロマトグラフィーなどの慣用の
方法によって実施される。
前記の製造法1および2によって得られた化合物は、粉砕、再結晶、カラムク
ロマトグラフィー、再沈殿などの慣用の方法によって分離し、精製することがで
きる。
目的化合物(I)は、溶媒和化合物[たとえば包接化合物(たとえば水和物な
ど)であってもよい。
目的化合物(I)は結晶質であるので、安定しており、取り扱いやすい。
目的化合物(I)の有用性を示すために、化合物(I)の薬理試験データを以
下に示す。
試験1:血小板へのフィブリノゲン結合 試験化合物 実施例1の化合物試験方法
洗浄ヒト血小板をゲル濾過により血小板多血漿から調製した。洗浄血小板を、
20μM ADPで10分間活性化し、0.8%パラホルムアルデヒドで30分
間固定した。次に血小板を、遠心分離により洗浄し、2mM塩化カルシウムおよ
び1mM塩化マグネシウムを含むHEPESタイロード緩衝液に懸濁した。
10μg/ml FITCフィブリノーゲンおよび試験化合物を用いて、35
0μlの血小板を最終濃度2x108/mlでインキュベートした。反応物を室
温で30分間放置した。FACScanフローサイトメーターを用いて、結合フ
ィブリノーゲンのFITC蛍光を測定した。
50倍過剰無標識フィブリノーゲンの存在下で、結合を差し引いて、特異結合
を計算した。結果をIC50値、すなわち結合を50%阻害するに要する用量で
表した。試験結果

この発明の医薬組成物は、目的化合物(I)を有効成分として、直腸内、肺内
(鼻または口腔吸入)、鼻内、眼内、外用(局所塗布)、経口または非経口(皮
下、静脈内および筋肉内を含む)投与または吸入に適した有機または無機の担体
または賦形剤と共に含有する医薬製剤の形態、たとえば固体、半固体または液体
の形態で用いることができる。
有効成分を、たとえば通常の無毒の医薬として許容される担体と共に配合し、
錠剤、ペレット剤、トローチ、カプセル剤、坐剤、クリーム、軟膏、エアロゾル
剤、吸入用散剤、液剤、乳剤、懸濁剤および用途に適した他の形態としてもよい
。さらに、必要に応じて、補助剤、安定化剤、増粘剤、着色剤および香料を使用
してもよい。
目的化合物(I)は、疾患の経過または症状に対して所望の効果をもたらすに
十分な量が医薬組成物に含有される。
この発明の医薬組成物は、この技術分野における慣用の方法によって製造でき
る。薬剤の生物学的利用能を改善するために、この技術分野で一般的に使用され
る技術を、必要に応じて、この発明の医薬組成物に適用してもよい。
医薬組成物をヒトまたは動物に投与する場合、静脈内投与(静脈内注射を含む
);筋肉内投与;肺内投与または経口投与;または計量式用量吸入器、噴霧器ま
たは乾燥粉末吸入器からのエアロゾル剤の吸入により投与するのが好ましい。
目的化合物(I)の治療有効用量は、治療を受ける個々の患者の年齢および症
状により変動し、左右されるが、一般的には、静脈内投与の場合、ヒトまたは動
物の体重1kgに対して1日当たり0.001ないし100mgの目的化合物(
I)を、筋肉内投与の場合、ヒトまたは動物の体重1kgに対して1日当たり0
.001ないし100mgの目的化合物(I)を、経口投与の場合、ヒトまたは
動物の体重1kgに対して1日当たり0.001ないし200mgの目的化合物
(I)を、ヒトまたは動物における前記の疾患の予防および/または治療のため
に投与すればよい。
下記の実施例は、この発明をさらに詳しく説明するために示したものである。実施例1
N−[(R)−1−{3−(1−ベンジルオキシカルボニル−4−ピペリジル
)プロピオニル}−3−ピペリジルカルボニル]−2(S)−アセチルアミノ−
β−アラニン(0.5g)、1N塩酸(0.94ml)と10%Pd−C(0.
1g)のテトラヒドロフラン(5ml)中の混合物を、大気圧下で2時間水素化
した。触媒を濾過により除去し、濾液を真空中で濃縮した。残留物を水に溶解し
、炭酸水素ナトリウム飽和水溶液で中和し、イソプロパノール:水=(1:1)
を溶離溶媒とするHP−20樹脂を用いて脱塩後、凍結乾燥し、エタノールから再
結晶して、N−[(R)−1−{3−(4−ピペリジル)プロピオニル}−3−ピ
ペリジルカルボニル]−2(S)−アセチルアミノ−β−アラニン(0.34g
、91.0%)を白色結晶として得た。
