電気泳動ゲル、ファイバースペーサーを有するゲルホルダーおよびその製造方
法
発明の背景
DNA配列決定は、実験室レベルで設計された自動化システムを用いて行うこ
とができる。DNA配列決定のための方法および装置は、米国特許第4,811,218
号明細書;同第4,823,007号明細書;同第5,062,942号明細書;同第5,091,652号
明細書;同第5,119,316号明細書および同第5,122,345号明細書に記載され、これ
らをここに参照として引用する。
これらのシステムに使用される一般的な方法は、制限エンドヌクレアーゼを用
いてサンプルDNAを分断し;対象となる制限フラグメントを増幅し(例えばP
CRを用いる);増幅されたDNAと配列決定プライマー(これは増幅プライマ

ゲルを用いて核酸配列の分析を行うにあたり、ゲルのサイズや厚さのようなゲ
ルの特徴は、分析を行うのに必要な時間およびコストに影響を及ぼす。診断用の
製品として配列決定の利用性を改善するために、配列決定の分析の時間およびコ
ストを減少させるのが望ましいので、短時間でオリゴヌクレオチドフラグメント
の極少量を分析できるゲルを提供するのが有利である。また、ゲルが満たされた
ときにこれらの所望される特性を提供する、大規模に製造され得る一回限りで使
い捨てのゲルホルダーが有利である。
分析時間が短く、配列分析に求められる高い生産性を達成するために、顕著に
薄いゲルを製造することが試みられてきた。とくに多く見られるのは、ゲルを囲
む基体間に極めて均一な空間を形成しかつ維持する組み立て技術を展開し、ゲル
自身に均一な厚さをもたせるというものである。米国特許第4,929,329号明細書
および同第5,164,066号明細書には、例えば前面および後面プレート間のスペー
サーとして、マイラーまたはナイロンモノフィラメントからなる薄いフィルム(
約0.10〜0.02インチ厚)を用いる電気泳動ゲルの形成が開示され、ここ
に参照として引用する。スペーサーはプレートに接着されるのではなく、単に2
つのプレート間におかれ、2つのプレート間のスペースがゲル形成溶液で満たさ
れるときに、クランプを用いて正確な場所に固定される。重合後、重合したゲル
は、正確な場所に配置された2つのプレートに保持される。
これらの特許に開示された電気泳動ゲルの製造方法は、幾つかの欠点を有する
。第1に、ゲル充填操作のときにスペーサーを正確な位置におき、これを保持す
る必要があり、得られるゲルは大規模生産に適合しない。また、ゲルは一旦製造
されると保存期間が短く、またゲルはプレートおよびスペーサに保持されるので

発明の要旨
本発明の上記の目的およびその他の目的は、接着剤を用いてゲルホルダーを形
成する基体に固定されたファイバー、とくにグラスファイバーを用いる電気泳動
ゲルのためのゲルホルダーを形成することにより達成される。したがって、本発
明の一つの見地は、次に示す方法である:
(a)平面の第1基体および平面の第2基体間に接着剤をコーティングした複数
のファイバーを配置し;
(b)前記ファイバーが前記第1基体および第2基体に接着するように基体の外
側に圧力を施し、これにより前記第1基体および第2基体間にゲルチャンバーを形
成し、前記ゲルチャンバーはファイバーの内部コアにより決定された厚さを有す
るものである。
これとは別に、コーティングしていないファイバーを2つ1組にしておき、この
1組の各ファイバー間のラインに接着剤を施すこともできる。接着剤が硬化した

発明の詳細な説明
図1は、本発明のゲルホルダーの第1の態様を示すものである。ゲルホルダー
は、上部の基体10、底部の基体11および複数のファイバー12から形成され
る。ファイバーの断面は、図示したような円形や、卵型や矩形であることができ
る。ファイバー12は、互いに平行にかつ基体10、11の対向端部に平行に配
置され、基体の一つの末端から互いに伸びている。底部の基体11は、電気泳動
のサンプルの充填を容易にするために、示したようにゲルホルダーの末端で、上
部基体10を超えて伸び、電気泳動のサンブルの充填を容易にしている。このよ
うに、上部の基体と末端を同じくするか、あるいは図1Aに示したように、底部
の基体が広がって伸びている。これとは別に、開口部13が、この目的のために
上部の基体のひとつの末端付近に設けられている(図1B)。
図2Aは、図1に示す4つのファイバーではなく、2つのファイバーを有する
ゲルホルダーを詳細に示している。ファイバー12は、上部の基体10および底
部の基体11と接触し、接着剤21により基体に接着している。ファイバー12
、上部の基体10および底部の基体11は、電気泳動ゲルで満たされるチャンバ
ー20を共に形成している。
図2Bは、本発明の別の態様を示している。この場合、ファイバー12は、対
をなして基体間におかれ、2つのファイバー間の空間を満たすように接着剤が線
上におかれている。この空間は、隣接する対をなすファイバー間と比較して非常
に小さく、好ましくは図2Bに示したように、2つのファイバーが互いにほぼ接触

接着剤は、基体およびファイバーの材料を結合できるものであれば特に制限さ
れない。好適な態様において、これはガラス−ガラス結合であるが、幾つかの用
途によってはガラス−プラスチック結合であることもできる。好適な接着剤は、
紫外線硬化性アクリレート接着剤、例えばM36,427タイプ68(Edmund Scientif
ic)または熱硬化性アクリル接着剤、例えばEPO-TEK 353 ND(Epoxy Technology
Inc.)等が挙げられる。多の適切な接着剤は、Deco Cementまたは5-minute Ep
oxy(Thorlabs)である。
もし接着剤が、図2に示した態様のように、ゲルがチャンバーを満たしたとき
にこれと接触するかもしれないならば、幾つかの接着剤が、ゲルの重合に悪影響
を及ぼすことに注意するべきである。すなわち、紫外線硬化性のゲルのためのホ
ルダーにおける紫外線硬化性接着剤の使用は、接着剤およびゲルの硬化への干渉
により、理想的ではない。この問題は、接着剤が、ファイバーの対によりゲルチ
ャンバーから分離している図2Bの態様では生じない。
本発明のゲルホルダーは、図3に示したように、接着剤でコーティングされた
ファイバーを用いることにより製造される。このようなファイバーは、基体間の
空間を有効に定めることのできる材料から製造した内部コア31を有する。好適
な材料はガラスであり、また実質上圧縮されない重合性材料、例えばナイロンで
ある。ファイバーは、接着剤の層32でコーティングされる。この層は、基体へ
のファイバーの接着を確実にし、しかし接着剤がゲルチャンバーの多くの部分を
ブロックするほど多くない程度に、十分な接着剤を提供する厚さを有するべきで
ある。好適な厚さは、5〜20μmである。ファイバー材料および接着剤は、チ
ャンバーの内部でのゲルの重合に悪影響を及ぼさず、また電気泳動の最中または
後の分離した材料の検出に使用される波長で強く蛍光を発しないようなものを選
択するべきである。
本発明のゲルホルダーを形成するために使用される基体は、電気泳動ゲルおよ
び使用される検出方法と調和する、平らな、平面の任意の材料であることができ
る。幾つかの場合において、電気泳動ゲルの重合または電気泳動ゲル上の分離し
たフラグメントの観察に干渉しないプラスチック材料を使用することができる。
しかしながら、好ましくは、基体はガラス製であるのがよく、最適には、蛍光を
あまり発しないガラスから形成されるのがよい。紫外線透過性の高い、1mmのボ
ロシリケートガラスが他の好適な材料である。
本発明のゲルホルダーを形成するために使用される基体の少なくとも1つは、
電気泳動ゲル内の材料の観察が可能な材料で形成されていなければならない。し
かしながら、幾つかの装置において、検出光線源、例えばゲル43中の蛍光分子
を励起するための光線42を生成するレーザー41および発した光線45を検出
するための検出器44が、図4に示したように、電気泳動ゲルの同じ側面におか
れることがある。この場合、底部の基体11は、サンプルの観察を可能する機能
よりも、むしろ背面からの照射が最小になるように選択される。例えば、蛍光検
出システムを備えた配列決定装置に使用するためのゲルホルダーの場合、底部の
基体11は、そこに到達する励起光をすべて吸収する、すなわち実質上非蛍光性
の着色ガラスから製造することができる。背面の蛍光は、基体の蛍光性不純物か
らそのほとんどが発せられるので、これによって背面の蛍光の量を顕著に減少さ
せ、観察の感度を改善することができる。
背面の蛍光を減少する別の方法は、透明の薄い基体を用いることである。例え
ば、図5に示すように、一方の基体50を厚い(かつ好ましくは吸収性で非蛍光
性の)材料から形成し、他方の基体51を非常に薄い(すなわち0.1mm以下)
透明の低蛍光性材料から製造する。好適な薄い材料は、“カバースリップ”ガラ
スを含む。また、薄い領域61は、図6Aに示すようにファイバーに対して鉛直
方向か、図6Bに示すようにゲルの一つの端部を覆うように励起/検出領域に配
置され得る。このような基体は、所望の形状になるように連続的な基体を成形す
ることにより(図7A)、あるいは連続的な薄い基体72上に厚い材料のブロッ
ク71を固定することにより形成することができる。後者の場合、厚い材料のブ
ロック71は、吸収する、非蛍光性の材料から形成し、背面の蛍光を減少させる
ことができる。
ゲルホルダーを形成するために、ファイバーは基体の1つの表面上にまずおか
れる。基体は、互いに平行にかつ基体の2つの対向する端部と平行にするべきで
ある。ファイバーは、互いに平行にこれらを維持するフレームにおかれ、また互
いに一定の距離のあるように配置され得る。もし2つのファイバーを使用するな
らば、これらは基体の端部付近に置くのが有利であり、内部に1つの大きなゲル
チャンバーを形成することができる。ゲルチャンバーが複数の部分に分割される
ように、2つ以上のファイバーを使用することがきる。この場合、ファイバーは
チャンバーの各部が4つのサンプルを受け入れるのに十分に大きい(すなわち配
列決定プロセスに使用される各鎖末端オリゴヌクレオチド混合物毎に1つのサン
プルレーン)ように、一定の間隔でおかれるのが好ましい。このように、16レ
ーン(4つの完全配列)を有するゲルにおいて、合計5つのファイバーが使用さ
れる。2つを超えるファイバーの使用は、幅広いゲルにとくに好適である。なぜ
ならば、内部ファイバーは、ゲルチャンバーの中央部の基体の沈下の防止に役立
つからであり、このようにして一層調和した幅のゲルチャンバーを形成すること
ができる。
本発明の方法の1つの態様において、次のステップはファイバーが2つの基体
間に配置されるサンドイッチ構造を形成するように、ファイバー上に第2基体を
おくことである。基体は、ファイバーの並びのどちらの側にも接触する。十分な
圧力を施して、ファイバーがサンドイッチ構造において基体に固定される。次に

本発明の方法の第2の態様において、ファイバーを第1基体に接着させ、その

図2Bで示される形態のようなゲルホルダーを形成するために、ファイバーの
対が所望の整列状態でおかれ、外部のフレームにより空気中に吊り下げられる。
ファイバーは、0.5〜2.5mmの狭い間隙をなして対になっている。この間隙
は、基体同士を固定するのに必要な接着剤の幅により定められる。整列したファ
イバーは、一方の基体上におかれる。ファイバーが適切な位置にあるとき、ファ
イバー間の空間は、例えばEPO-TEK 353 NDのようなエポキシ接着剤で満たされる
。第2基体は、整列したファイバー上におかれ、サンドイッチ構造を形成する。
