アミノメチル-フェニルイミダゾールの調製
発明の分野
本発明は、脳のドーパミンレセプターサブタイプでの非常に選択的な部分アゴ
ニストまたはアンタゴニストとしてまたはそれらのプロドラッグとして、有用な
特性を有するアミノメチル-フェニルイミダゾールの調製の新規方法、特に、5-
フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1-ピペラジニル
]ピリミジンおよびその薬学的に受容可能な酸付加塩の調製に関する。本発明は
また、この方法で有用な新規な中間体に関する。
発明の背景
米国特許明細書第5,159,083号(Thurkaufら、Assignors to Neurogen Corporat
ion)は、ドーパミンレセプターサブタイプに特異的なリガンドの1種として、ア
ミノメチル-フェニルイミダゾール誘導体を記述し、請求している。類似の化合
物は、PCT公報 WO 92/12134(これは、大部分は、USP 5,159,083に対応している)
、WO 96/10018およびWO 96/16040に記述されている。このような化合物は、精神
分裂症のような精神病の処置、また、パーキンソン症候群のような精神運動の障
害の処置において、有望であることが明らかとなっている。
USP 5,159,083の式Iに含まれる化合物のサブグループには、フェニルイミダ
ゾリルメチルピペラジンが包含され、USP 5,159,083の式Iに含まれるが具体的
には開示されていないこのタイプの特に好ましい化合物には、5-フルオロ-2-[4-
[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1-ピペラジニル]ピリミジン、
すなわち、式Aを有する化合物がある:
この化合物は、37ページにて、化合物58として、および55ページ、22〜23行目
の実施例9にて(その二塩酸塩として)、WO 96/16040で開示されており、そこで
は、別名、2-フェニル-4(5)-[(4-(5-フルオロ-2-ピリミジニル)-ピペラジン-1-
イル)-メチル]-イミダゾールが与えられている。他の好ましい化合物には、対応
するデス-フルオロ誘導体である2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メ
チル]-1-ピペラジニル]ピリミジンがある。
一般的な化学方法は、アミノメチル-フェニルイミダゾール誘導体の調製につ
いて、上で挙げた米国特許およびPCT公報;USP 5,159,083(それは、11欄および1
2欄にわたって、スキームIで示されている)に記述されている。(本発明の方法
に関連して)重要な工程には、スキームIの最後の矢印にあるもの、すなわち、
対応する5-クロロメチル化合物を生じるためのチオニルクロライドによる5-ヒド
ロキシ-メチル-2-(必要に応じて、置換フェニル)-1H-イミダゾールの塩素化があ
り、これは、次いで、アミンと反応されて、所望のアミノメチル-フェニルイミ
ダゾール、例えば、フェニルイミダゾリルメチルピペラジンを生じる。式Aの化
合物の調製については、これは、以下のようにして、スキームAで示すことがで

しかしながら、その方法は、特に、式Aの化合物に適用されるとき、多数の欠
点がある:
1.このクロロメチル中間体は、不安定であり、反応性である;
2.従って、それを再結晶によって充分に精製するのは、困難である;
3.結果的に、その調製、さらに転化のプロセスを拡大することが、困難であ
る;
4.5-フルオロ-2-ピペラジニル-ピリミジンとのその縮合は、特に、大規模調
製では、比較的に低い収率で進行する;
5.所望生成物である5-フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-イ
ル)メチル]-1-ピペラジニル]ピリミジンは、その工程から精製するのが容易では
ない。
このタイプの化合物の他の可能な調製方法は、上記スキームAにおいて、式B
のヒドロキシ化合物および式Cのアミノ化合物と類似の化合物の融合を包含する
。この方法は、2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1-ピペラ
ジニル]ピリミジンについて、WO 96/16040の45ページのスキーム3、および実施
例39(B部)に例示されている。しかしながら、この方法では、僅かに約80%の収
率の2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1-ピペラジニル]ピリ
ミジンしか得られず、多量(2当量以上)の式Cの化合物を消費する。さらに、実
用的な収率の式Aの化合物それ自体は得られない。
これらの欠点の一部は、この系列の他の化合物の調製においても、同様にあて
はまる傾向にある。
明らかに、これらの欠点を回避するかまたは少なくする方法であって、より高
い収率および純度で最終生成物を生じるアミノメチル-フェニルイミダゾールの
調製工程を提供し、取扱い(特に、精製)のより容易な中間体から進行し、商業規
模での調製にさらに適当な方法が必要とされている。
発明の要旨
本発明は、このような方法およびそこで有用な中間体を提供する。
本発明は、次式の化合物およびそれらの薬学的に受容可能な酸付加塩を調製す

発明の詳細な説明
式IIIの化合物(ここで、Rは、上で定義したアシル基である)は、安定であり、
標準的なアシル化法により、対応するアルコール(式IIIの化合物であって、ここ
で、Rは、Hである)から、良好な収率で、容易に調製される。それらは、例えば
、再結晶により容易に精製される。それらは、式IVの化合物と円滑に反応して、
良好な収率および純度で、式IIの化合物を生じる。
式IIIまたはIIIaの化合物(ここで、R1は、水素である)は、このような水素原
子が、イミダゾール環のいずれかの窒素原子と特異的に結合していないが、一方
から他方に移動し得るので、互変異性体である。このような互変異性体の全ては
、式IIIまたはIIIaに入る。同様に、式IIの化合物(ここで、R1は、水素である)
は、互変異性体である。式Aの化合物もまた、互変異性体形熊で存在し得る。
本明細書および請求の範囲では、以下で挙げた種々の用語は、他に指示がなけ
れば、以下の意味を有する:
「ハロゲン」は、フッ素、塩素、臭素またはヨウ素を表わす。
「アルキル」は、1個〜6個の炭素原子を含有する飽和の直鎖または分枝鎖炭
化水素基(例えば、メチル、エチル、プロピル、2-プロピル、ブチル、2-ブチル

「アルカノイル」は、ホルミル基またはアルキルカルボニル基を表わし、ここ
で、このアルキル基は、上で定義したものと同じである(例えば、プロピオニル
、ブチリル、ペンタノイルまたはヘキサノイル)が、好ましくは、アセチルであ
る。
「芳香族」(芳香族カルボン酸または芳香族スルホン酸に関連して)は、ハロゲ
ン原子およびアルキル基およびアルコキシ基からなる群から選択した1個または
2個の置換基を有し得るベンゼン核またはナフタレン核(好ましくは、非置換フ
ェニル、4-クロロフェニルまたは4-トルイル)を表わす。
「アラルキル」は、アリール-アルキル基を表わし、ここで、このアリール基
は、上で定義したアルキル基(好ましくは、メチルまたはエチル)で置換されてい
る、上で定義した芳香族基(好ましくは、フェニルまたは置換フェニル)である。
「アラルカン(aralkanoic)」は、アリール-アルキル-カルボニル基を表わし、
ここで、このアリール基は、上で定義したアルキル基(好ましくは、メチルまた
はエチル)で置換されている、上で定義した芳香族基(好ましくは、フェニルまた
は置換フェニル)である。
DMSO、DMFおよびTHFは、それぞれ、ジメチルスルホキシド、N,N-ジメチル-ホ
ルムアミドおよびテトラヒドロフランを表わす。
好ましいアルキル基およびアルコキシ基には、メチル基、メトキシ基、エチル
基およびエトキシ基が含まれる。
R(式IIIの化合物において)は、例えば、アルカン-または芳香族-スルホニル基
(例えば、メタンスルホニル、エタンスルホニル、ベンゼンスルホニル、4−クロ
ロベンゼンスルホニルまたは4-トルエンスルホニル)、または芳香族カルボン酸
またはアラルカン酸のアシル基(例えば、ベンゾイル、4-クロロベンゾイル、4-
メチルベンゾイルまたはフェニルアセチル)であり得るが、好ましくは、低級ア
ルカノイル基(例えば、ホルミル、プロピオニルまたはブチリル、または特に、
アセチル)である。
本発明の化合物の特に好ましい群は、式IIを有し、ここで:
XおよびYは、同一または異なり得、水素またはハロゲン原子、あるいは低級ア
ルキル基または低級アルコキシ基である;
R1は、水素原子またはメチル基であり、イミダゾール環の窒素原子に結合して
いる;
R2は、水素原子または低級アルキル基である;
Wは、CHまたはNである;
R3は、水素原子である;そして
R5およびR6は、隣接窒素原子と一緒になって、式

り、ここで、R7は、水素またはフェニル基、ピリジニル基またはピリミジニル基

Wは、最も好ましくは、NR7である。R7中のピリジニル基またはピリミジニル基
は、好ましくは、2-ピリジニル基または2-ピリミジニル基であり、これは、ハロ

式IIIの化合物と式IVの化合物との間の反応は、好ましくは、過剰の有機塩基
または無機塩基を用いて行われる。有機塩基は、好ましくは、第三級有機塩基、
特に、トリアルキルアミン、例えば、トリ(低級アルキル)アミン(例えば、ジメ
チルエチルアミン、トリエチルアミン、トリプロピルアミン、トリ(2-プロピル)
アミン、N,N-ジ(2-プロピル)エチルアミンまたはトリブチルアミン)である。ト
リエチルアミンは効果的であり、その有用性および低価格を考慮すると、特に好
ましい。望ましいなら、第三級有機強塩基、例えば、N,N-ジメチル-アミノピリ
ジンもまた、好ましくは、少量(例えば、トリアルキルアミンの2〜10%)で、添
加し得る。無機塩基は、例えば、ナトリウムまたはカリウムの水酸化物、炭酸塩
または重炭酸塩であり得る。不活性有機溶媒、例えば、芳香族炭化水素(例えば

この反応が完結するまで(典型的には、50〜60℃で3〜5時間)、室温またはやや
高温(例えば、40〜70℃、好ましくは、50〜60℃)で行われる。式IIの生成物は、
次いで、標準的な手法により、例えば、水の添加および濾過、または水非混和性
有機溶媒での抽出により、単離され得る。それは、遊離塩基または薬学的に受容
可能な酸付加塩として、単離され得る。この遊離塩基は、望ましいなら、薬学的
に受容可能な酸付加塩に転化され得る。好ましい塩には、二塩酸塩およびジメタ
ンスルホネートが挙げられる。
式IIIのイミダゾール出発物質は、対応するヒドロキシメチル化合物(ここで、
Rは、Hである)を適当なアシル化剤でアシル化することにより、調製され得る。
アシル化剤は、例えば、適当なエステル形成試薬(例えば、カルボジイミド)の存
在下にて、酸ハロゲン化物または酸それ自体でさえあり得るが、好ましくは、酸
無水物R2O、またはさらに好ましくは、Ra 2O(ここで、RおよびRaは、それぞれ、
式IIIおよびIIIaについて、上で定義したものと同じである)であり、これは、僅
かに過剰、例えば、5〜20%過剰、好ましくは、8〜12%過剰で使用され得る。
アシル化剤が、単に、酸無水物のとき、塩基は必要ではない。それ以外では、塩
基として、第三級アルキルアミン(例えば、トリエチルアミン)が使用され得る。
式IIIの生成物は、次いで、標準的な方法により、例えば、もし沈殿するなら、
冷却し、濾過することにより、またはもしこの溶媒が水混和性なら、水を添加し
、そして沈殿物を濾過するか、または水非混和性有機溶媒での抽出により、単離
され得る。
5-ヒドロキシメチル-2-フェニル-1H-イミダゾールおよび5-フルオロ-2-ピペラ
ジニル-ピリミジンは、市販されている。
5-アセトキシメチル-2-フェニル-1H-イミダゾールは、非常に安定であって、
再結晶のような簡単な方法により容易に精製されるので、5-クロロ-メチル-2-フ
ェニル-1H-イミダゾールよりも優れた試薬である。さらに、その5-フルオロ-2-
ピペラジニルピリミジンとの縮合は、円滑できれいに進行して、高収率かつ高純
度で、5-フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1-ピペ
ラジニル]ピリミジンが生じる。これらの理由のために、5-アセトキシメチル-2-
フェニル-1H-イミダゾールは、5-クロロメチル-2-フェニル-1H-イミダゾールよ

5-フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1-ピペラジ
ニル]ピリミジンは、薬学的組成物中で、その二塩酸塩として、非常に有利に投

実施例
本発明を例示するために、以下の実施例を提示する。工程I: 5-アセトキシメチル-2-フェニル-1H-イミダゾールの調製 5-ヒドロキシメチル-2-フェニル-1H-イミダゾール(1.3 Kg)およびトルエン(10

ニル-1H-イミダゾール(1.17 Kg、1.1当量)、DMAP(N,N-ジメチルアミノピリジン
、30g)、トルエン(7.2 L)、エタノール(1.8 L)およびトリエチルアミン(1.095
L)を、20〜25℃で混合し、そしてこの混合物を、反応が完結するまで(典型的に
は、約20〜25時間)、撹拌しながら、50〜55℃で加熱する。次いで、水(3.75 L)
を添加し、そして得られた懸濁液を0〜5℃まで冷却する。生成物を濾過により
除き、それを水(6×1.4 L)および冷トルエン(3×700 ml)で洗浄し、そして真
空下にて、一定重量まで、50〜55℃で乾燥する。
収量:約1.55 Kg(92%)、HPLCにより、99%以上の純度;m.p.146.4℃(DSC、
分解)、MS:339(MH+)。工程III: 5-フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1 -ピペラジニル]ピリミジン二塩酸塩の調製
還流状態(75〜80℃)で、5-フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-
イル)メチル]-1-ピペラジニル]ピリミジン(25g)を乾燥エタノール(250 ml)に溶
解する。脱色炭(1.25g)を添加し、黒色懸濁液を20〜30分間撹拌し、そして炭素
を濾過により除く。
55〜60℃で、濾液に、濃HCl(水性)(13.5 ml)を添加し、溶液を、30〜40分間に
わたって、20〜25℃まで冷却し、次いで、0〜5℃までさらに冷却し、そしてそ
の温度で2時間保存する。固形物を吸引濾過により除き、そしてエタノール(25
ml)で洗浄する。
収量;28g(93%)、少なくとも99%の純度;190℃以上で、分解し始める(この
分解温度は、加熱速度に依存する)。
この塩は、エタノールから再結晶し得る。工程IV: 5-フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル]-1- ピペラジニル]ピリミジンジメタンスルホネートの調製
55〜60℃で、5-フルオロ-2-[4-[(2-フェニル-1H-イミダゾール-5-イル)メチル
]-1-ピペラジニル]ピリミジン(0.5g)をエタノール(6ml)に懸濁させる。メタン
スルホン酸(0.21 ml)を添加し、混合物を30分間撹拌する。それを0〜5℃まで
冷却し、そして濾過して、ジメシレート塩(m.p.228〜230℃、分解)0.7g(90%)
を得る。
