scrizione del brevetto per invenzione industriale avente per titolo: registration of the patent for industrial invention entitled:
“Procedimento per la preparazione di un composto benzimidazolico in forma amorfa” "Process for the preparation of a benzimidazole compound in amorphous form"
CAMPO DELL’INVENZIONE FIELD OF THE INVENTION
The present invention relates to a process for the preparation of sodium rabeprazole in stable amorphous form, free from traces of crystallinity.
STATO DELLA TECNICA STATE OF THE TECHNIQUE
Rabeprazole sodium is the common name of the compound 2 - [[[4- (3-methoxypropoxy) -3-methyl-2-pyridinyl] -methyl] sulfinyl] - 1H -benzylimidazole sodium salt, having the following formula (I).
Rabeprazole sodium is biologically active as an inhibitor of gastric secretion, and used for the treatment of peptic ulcer.
The preparation of rabeprazole sodium is described in US 5,045,552. Japanese patent application JP 2001/039975 reports that the product obtained in example 33 of US 5,045,552 with m.p. 140-141 ° C (d) corresponds to sodium rabeprazole in amorphous form. US patent 6,180,652 describes the preparation of rabeprazole sodium in amorphous form by freeze-drying of an aqueous solution of a complex of acetone and rabeprazole sodium salt. Application WO 2008/146297 describes the preparation of rabeprazole sodium in amorphous form by means of a solvent / antisolvent system, where the solvent is an aromatic ether and the solvent is for example a C5-C7 cycloalkane. According to application WO 2007/023393 amorphous rabeprazole sodium is obtained with a multiphase process, consisting in contacting the rabeprazole sodium / acetone complex with a first solvent, for example a hydrocarbon, an ether, an alcohol, or a mixture thereof; partially or totally removing said first solvent; contacting the remaining residue with a second solvent, for example a hydrocarbon or an ether, to obtain a wet solid, which is then recovered and dried. Application WO 2008/035192 claims a method for obtaining amorphous sodium rabeprazole comprising the reaction of rabeprazole with a solution of sodium tert-butoxide in tert-butyl alcohol; the removal of tert-butyl alcohol to obtain a residue; addition of diisopropyl ether, followed by stirring and separation of the solid.
Therefore, one of its possible impurities that can be formed, according to the known methods for the preparation of amorphous sodium rabeprazole, is a sulfone having the following formula (II)
Due to the similar physicochemical characteristics between rabeprazole and the sulfone of formula (II), the purification of the final product is difficult and extremely laborious.
Furthermore, as known, different forms of biologically active compounds, such as in particular the polymorphic forms and the amorphous form, can be useful both in therapy, thanks to the different bioavailability, release times and solubility, and in pharmaceutical technique in the preparation of formulations, as physical characteristics which often accompany the different physical form of the drug, such as for example hygroscopicity, flow and / or compacting of the powders, can be advantageously used. Obtaining these forms, and in particular the amorphous form of rabeprazole sodium, however, is not particularly easy from an industrial point of view.
There is therefore the need to have a process, which can be applied industrially, and allows to obtain rabeprazole sodium in amorphous form, with high yields and purity, and free from any trace of crystallinity. Furthermore, the preparation method should not require the use of special and dedicated equipment.

An ethereal antisolvent, according to the present invention, is an ethereal solvent in which rabeprazole sodium is poorly soluble or insoluble. Such ethereal solvent, which can be a cyclic or acyclic ether, is for example diethyl ether, diisopropyl ether, medium t-butyl ether (MTBE), dibutyl ether, cyclopentylmethylether, tetrahydrofuran, 1,4 dioxane or a mixture of two or more, preferably two or three, of said antisolvents. The solution of rabeprazole sodium can be prepared using from about 1 to about 50 volumes of solvent, preferably from about 3 to about 10 volumes, more preferably from about 4 to about 8 volumes; where the term "volume" refers to the number of milliliters (ml) of solvent used for each gram (g) of dissolved hydrated sodium rabeprazole.
The dissolution of rabeprazole sodium in hydrated form in the ketone solvent can be carried out at a temperature between about 40 and 65 ° C. To facilitate the dissolution of rabeprazole sodium, the solution is preferably heated to reflux and maintained at this temperature until it is completely dissolved.
The concentration of the solution can be carried out by removing the ketone solvent, for example by distillation, typically at atmospheric pressure, so as to reduce it to about 1.5-3 volumes, preferably about 2 volumes.
The concentrated solution, thus obtained, is added to the ethereal antisolvent, typically by dripping it into the antisolvent.
The anti-solvent is typically used at a temperature between about 5 and 35 ° C, preferably at room temperature.
The amount of ethereal antisolvent used can be between about 1 and about 200 volumes, preferably between about 5 and about 100 volumes, more preferably between about 10 and about 100 volumes, where the term "volumes" means the number of milliliters ( mi) of antisolvent used for each gram (g) of rabeprazole sodium in hydrated form dissolved in the starting solution.
To facilitate the formation of the amorphous solid, the temperature of the mixture can be cooled for example to a temperature of between about -5 ° C and 5 ° C, preferably between about 0 ° C and 5 ° C.
The time for adding the rabeprazole sodium solution in the antisolvent can vary from a few minutes to a few hours, preferably within an interval of between about 10 minutes and an hour.
The solid of rabeprazole sodium in amorphous form thus obtained is preferably then washed with an ethereal antisolvent as described above, preferably the same antisolvent used in step c).
The recovery of sodium rabeprazole in amorphous solid form can be carried out using techniques known to those skilled in the art, for example by filtration on a Buchner filter, or by centrifugation.
The process of the invention is particularly advantageous because it allows to obtain sodium rabeprazole in amorphous solid form without any trace of crystallinity, as can be seen from the relative XRPD graph in the figure. Furthermore, rabeprazole sodium amorphous can be obtained with a very high yield, normally greater than 90% up to a quantitative yield, i.e. equal to about 100%, and with a chemical purity evaluated by HPLC greater than or equal to 96%, preferably greater than or equal to 98%.
Rabeprazole sodium in amorphous form as obtainable according to the process of the present invention surprisingly proved to be a stable solid from the chemical point of view, since even after a few months no formation of degradation products / impurities was found in the same.
Furthermore, rabeprazole sodium in amorphous solid form as obtainable by means of the process of the present invention, even following long storage times, has not developed any conversion into its crystalline form, resulting therefore also stable from a physical point of view.
Rabeprazole sodium in amorphous solid form thus obtained has a D50 comprised between about 25 and 250 µm. If desired, this value can be reduced by micronization or fine milling.
On the basis of what has been described above, a further object of the present invention is therefore a pharmaceutical composition comprising rabeprazole sodium in amorphous solid form, as obtainable according to the process of the present invention, as active principle and at least one pharmaceutically acceptable excipient and / or carrier.
This pharmaceutical composition can be prepared in a pharmaceutical form according to known methods. The dosage of the active ingredient present in this composition may be that commonly used in the clinic for rabeprazole sodium.
Il seguenti esempi illustrano rinvenzione. The following examples illustrate the invention.
Esempio 1: Preparazione di rabeprazolo sodico in forma solida amorfa Example 1: Preparation of sodium rabeprazole in amorphous solid form
Rabeprazole sodium monohydrate (50.0 g, 0.125 mol), is dissolved in acetone (300 ml) at reflux temperature (about 56 ° C). The solution thus obtained is concentrated at atmospheric pressure up to a residual volume of about 100-125 ml, then cooled to room temperature and dropped in about 30 minutes in tert-butylmethylether (750 ml) at a temperature of 20-25 ° C. After about 1 hour the suspension obtained is cooled to a temperature of 0-5 ° C. After about 1 hour the suspension is filtered on a buckner funnel and the solid washed with tert-butylmethylether. The amorphous solid is dried at about 45 ° C under vacuum to constant weight. 48g of Rabeprazole sodium are obtained in amorphous form with a yield of 96% and having the XRPD diffractogram shown in Figure, which demonstrates that the amorphous sodium Rabrepazole thus obtained is pure and has no trace of crystallinity.
