Descrizione dell'invenzione industriale dal titolo: Description of the industrial invention entitled:
METODO DI PREPARAZIONE DI TIANEPTINA SALE SODICO METHOD OF PREPARATION OF TIANEPTINA SODIUM SALT
Campo dell’invenzione Field of invention
Stato della tecnica The invention relates to a method of preparing tianeptine sodium salt. State of the art
Tianeptine (7 - [(3-chloro-6,1 1-dihydro-6-methylbenzo [c, /] [1, 2] thiazepin-1 1-yl) amino] heptanoic acid S, S-dioxide) (represented from formula I) is a tricyclic compound with psychostimulating, anti-ulcer and antiemetic properties.
It is widely used in the therapy of depression, as it acts on the uptake of serotonin without significant activity on other receptors or on other transport systems of other neurotransmitters involved in depression, in particular monoamines. In the preparation of pharmaceutical compositions usable for the therapeutic treatment of depression it is usually used as the sodium salt.
Being hygroscopic, it is a product that is difficult to handle in the formulation of compositions useful for therapeutic purposes and from this point of view the pharmaceutical grade compound must not contain more than 5% of water. Furthermore, the pharmaceutical product must have an impurity content individually lower than 0.1%.
With reference to these aspects, in WO201 0/070667 a synthesis method of tianeptine in acid form is described specifically aimed at the preparation of a non-hygroscopic product, with a high degree of purity and with high yields by means of a new intermediate ( 7 - [(3-chloro-6,11-dihydro-6methylbenzo [c, /] [1,2] thiazepin-1 1-yl) amino] heptanoylamide S, S-dioxide). The same patent application describes the preparation of the sodium salt of tianeptine by treatment with a strong base chosen from sodium hydroxide and sodium alkoxides (preferred methoxide or ethyl oxide) in an alcoholic solvent selected from methanol, ethanol, propanol , isopropanol and butanol (preferred methanol and ethanol). The expected equivalent ratios between tianeptine in free acid form and bases are between 0.85 and 1.1 eq. and in particular two examples are described in which the tianeptine in the form of free acid and the NaOH or CH3ONa bases are in a molar ratio of 1: 1.1 in both cases, in which therefore the base is added in excess. It should be noted in this regard that, contrary to what is claimed, such a high strong base molar ratio: tianeptine in acid form is particularly disadvantageous in terms of product purity, as such a ratio leads to high values of the two imine isomers c / s and trans of Formula VII, reported below as Formulas Vll-a and Vll-b (note that these are two well separable impurities in HPLC):
Formula Vll-a Formula Vll-b Formula Vll-a Formula Vll-b
The problem of limiting the formation of impurities determines the need to identify new processes for the preparation of this active principle such as to guarantee its high yield and purity and therefore allow its pharmaceutical use.
Sommario Summary
The inventors of the present patent application have now identified a new method of preparation of tianeptine sodium salt based on the use of weak sodium bases.
Therefore, the present invention provides a method of preparing the tianeptine sodium salt for treatment in a solvent of tianeptine in acid form with a base selected between sodium carbonate and bicarbonate.
The advantages achievable with the present invention will become more evident to those skilled in the art from the following detailed description of particular embodiments of the method and of the compounds obtainable with the same given for illustrative and non-limiting purposes.
Descrizione dettagliata dell’invenzione Detailed description of the invention
The method of preparation of the tianeptine sodium salt object of the invention exploits the use of weak bases such as sodium carbonate and bicarbonate, for the salification of tianeptine in the form of free acid prepared according to the synthesis processes previously described .
The use of these bases, instead of the strong ones usually employed, ie sodium hydroxide or alkoxides, is based on the fact that the inventors have found that strong bases more easily generate unwanted by-products for a pharmaceutical grade product. The salification reaction is optimized through the use of solvents selected from alcohols and linear or branched ketones of short-chain alkyls, preferably with a number of carbon atoms between 1 and 4 and water. The preferred solvents among the alcohols are methanol, isopropanol and among the ketones are acetone and methylisobutylketone. These organic solvents can be mixed with each other. The preferred solvent for the salification step is methanol.
To obviate the formation of reaction by-products, in particular imines, it is also preferable to keep the molar ratio between tianeptine in acid form and sodium carbonate close to 1: 0.5, preferably between 1: 0.49 and 1: 0.51. In the same way, it is also preferable to keep the molar ratio between tianeptine in acid form and sodium bicarbonate close to 1: 1, preferably between 1: 0.98 and 1: 1.02. The weight / volume ratio between acid tianeptine and solvent is between 1 (acid tianeptine) and 2 (solvent) and 1 (acid tianeptine) and 10 (solvent). In order to limit the use of solvents, the weight / volume ratio between tianeptine acid and preferred solvent is 1: 2 - 3.
The salification reaction is preferably carried out at the reflux temperature of the solvent and maintained at this temperature until the completion of the salification reaction.
The preferred conditions for this salification step are: base = sodium carbonate, solvent = methanol, molar ratio 1: 0.5 between tianeptine in acid form and sodium carbonate.
After the salification step, the sodium tianeptine is separated and, in this way, purified according to techniques known to a person skilled in the art, preferably by precipitation in isopropanol, filtration and vacuum drying.
As a whole, therefore, the method of preparation of the tianeptine sodium salt according to the invention comprises the steps of:
salificazione della tianeptina in forma acida per trattamento con una base scelta tra carbonato e bicarbonato di sodio in un solvente alla temperatura di riflusso del solvente; salification of tianeptine in acid form by treatment with a base selected from sodium carbonate and bicarbonate in a solvent at the reflux temperature of the solvent;
opzionalmente, concentrazione della miscela di reazione o eliminazione del solvente di reazione e sua sostituzione con altro solvente; optionally, concentration of the reaction mixture or elimination of the reaction solvent and its replacement with another solvent;
precipitazione della tianeptina sodica in un antisolvente e suo isolamento. precipitation of sodium tianeptine in an antisolvent and its isolation.
With the method object of the invention, a product is obtained with a purity profile such as to make it usable as an active ingredient of pharmaceutical grade. The present invention will now be described with reference to the following examples which are provided for illustrative purposes only and are not to be construed as limiting the present invention.
