La presente invenzione riguarda un procedimento per la preparazione di derivati dell’idrossitirosolo di formula generale (III), dove R=OCH3lo CF3o una catena alchilica satura o insatura, lineare o ciclica, ramificata o no, da 1 a 31 atomi di carbonio, e per la preparazione di idrossitirosolo (IV). The present invention relates to a process for the preparation of hydroxytyrosol derivatives of general formula (III), where R = OCH3l or CF3o a saturated or unsaturated alkyl chain, linear or cyclic, branched or not, from 1 to 31 carbon atoms, and for the preparation of hydroxytyrosol (IV).
L’idrossitirosolo (3,4-diidrossifenil etanolo) è un fenolo naturale contenuto in un prodotto tipico dell’area del Mediterraneo, l’olio di oliva (Ragazzi e Veronese, Riv. Ital. Sostanze Grasse 1973, 50, 443-452; Vazquez Roncero et al., Grasas Aceites 1976, 27, 341-345; Cortese et al., Riv. Ital. Sostanze Grasse 1983, 60, 341-351; Montedoro et al., J. Agric. Food Chem. 1992, 40, 1571-1576; Angerosa et al., J. Agric. Food Chem. 1995, 43, 1802-1807) ed è presente anche nelle acque di vegetazione che ne derivano dalla sua produzione (Vaszquez Roncero et al., Grasas Aceites 1974, 25, 341-345; Capasso et al., Phytochemistry 1992, 12, 4125-4128). Recentemente è stato identificato e determinato quantitativamente anche in alcuni vini rossi e bianchi (Di Tommaso et al., J. High Resol. Chromatogr. 1998, 21, 549-553). Hydroxytyrosol (3,4-dihydroxyphenyl ethanol) is a natural phenol contained in a typical product of the Mediterranean area, olive oil (Ragazzi and Veronese, Riv. Ital. In letteratura sono ampiamente descritti gli effetti benefici che derivano dal consumo quotidiano di olio di oliva nei confronti di alcuni disturbi cardiovascolari, neurovegetativi e di alcune forme tumorali (Assmann et al., Europ. J. Cancer Prev. 1997, 6, 418-421; Saenz at al., Farmaco, 1998, 53, 448-449). Molte di queste attività biologiche, prime tra tutte quella antiossidante, sono attribuite all’idrossitirosolo (Visioli e Galli, Nutr. Metabol. Cardiovasc. Dis. 1995, 5, 306-314; Aruoma et al. J. Agric. Food Chem. 1998, 46, 5181-5187; Manna et al., J. Nutr. Biochem. 1999, 10, 159-165; Della Ragione et al., Biochem. Biophys. Res. Comm. 2000, 278, 733-739; Visioli et al., Circulation 2000, 102, 2169-2170). In the literature, the beneficial effects deriving from the daily consumption of olive oil against some cardiovascular and autonomic disorders and some cancers are widely described (Assmann et al., Europ. L’idrossitirosolo viene utilizzato come principio attivo o additivo: 1) nel settore farmaceutico, per la prevenzione e/o il trattamento di malattie cardiovascolari, disturbi neurovegetativi, ischemia cerebrale, diabete di tipo II; 2) nel settore cosmetico, per la preparazione di creme protettive per la pelle e di rossetti per labbra; 3) nel settore alimentare, per stabilizzare o migliorare la qualità di alimenti e bevande, in particolare gli oli vegetali; 4) nel settore agrochimico, per il controllo della crescita di funghi e batteri patogeni. L’idrossitirosolo non è commercializzato dalle più comuni ditte fornitrici di prodotti chimici (Sigma, Aldrich, Fluka, Acros); mentre è disponibile presso Extrasynthèse (Genay, Francia) e Cayman Chemical Europe (Estonia) ad un prezzo elevato, pari a circa 1500 Euro/grammo. E’ presente come componente, seppure in piccole percentuali, in molti estratti vegetali commercializzati dalla Società CreAgri, INC (Olivenol, Hidrox) e da Indena (Oleaselect). Tali procedure sono spesso laboriose, richiedono l’uso di notevoli quantità di solventi organici nocivi e infiammabili e adeguate apparecchiature di estrazione e di purificazione. Inoltre, in alcuni casi, l’idrossitirosolo non è recuperato in resa elevata e viene estratto insieme ad un altro componente di minor pregio, il tirosolo (4-idrossifenil etanolo). 1999, 47, 1745-17489; Visioli and Galli, J. These procedures are often laborious, requiring the use of large quantities of harmful and flammable organic solvents and adequate extraction and purification equipment. Furthermore, in some cases, hydroxytyrosol is not recovered in high yield and is extracted together with another component of lesser value, tyrosol (4-hydroxyphenyl ethanol).
In letteratura sono riportate alcune procedure sintetiche dell’idrossitirosolo che utilizzano come materiali di partenza il 3,4-dimetossifenil etanolo (Schopf et al., Liebigs Ann. Chem. 1949, 563, 86-93); l’acido 3,4-dimetossifenil acetico (Baraldi et al., Liebigs Ann. Chem. The literature reports some synthetic hydroxytyrosol procedures that use 3,4-dimethoxyphenyl ethanol as starting materials (Schopf et al., Liebigs Ann. Chem. 1949, 563, 86-93); 3,4-dimethoxyphenyl acetic acid (Baraldi et al., Liebigs Ann. Chem.

Tali sintesi richiedono quattro-cinque passaggi e le rese finali di idrossitirosolo non sono elevate. Più convenientemente, l’idrossitirosolo può essere ottenuto per riduzione dell’acido 3,4-diidrossifenil acetico. In tal caso, però è richiesto l’uso di un reagente pericoloso, quale il litio alluminio idruro e di un solvente anidro, il tetraidrofurano (resa: 79%, Capasso et al., J. Agric. These syntheses require four to five steps and the final yields of hydroxytyrosol are not high. More conveniently, hydroxytyrosol can be obtained by reducing 3,4-dihydroxyphenyl acetic acid. Recentemente, sono stati messi a punto alcuni procedimenti sintetici a partire da materiali di partenza contenuti essi stessi nelle foglie di olivo, nell’olio di oliva e nelle acque di vegetazione, quali l’oleuropeina (una forma di idrossitirosolo esterificato) e il tirosolo. Tale procedura consente di ottenere una soluzione 0.1 M di idrossitirosolo in acido cloridrico dopo idrolisi basica dell’oleuropeina sotto flusso di azoto e successiva acidificazione dell’idrolizzato. Questo processo ha lo svantaggio di utilizzare un materiale di partenza che, se non ricavato da fonti vegetali, non è disponibile presso le più comuni ditte fornitrici di prodotti chimici (Sigma, Aldrich, Fluka, Acros). This process has the disadvantage of using a starting material which, if not obtained from vegetable sources, is not available from the most common chemical product suppliers (Sigma, Aldrich, Fluka, Acros).
Molto diversa è, invece, la disponibilità del tirosolo. Infatti, è commercializzato anche da tali ditte, tra l’altro, ad un costo molto basso (10-13 Euro/grammo). Una interessante applicazione dell’uso di tale composto per produrre idrossitirosolo è stata sviluppata sfruttando l’ossidrilazione aromatica operata da una polifenolo-ossidasi, la tirosinasi EC 1.14.18.1, cui segue un work-up riducente in presenza di acido ascorbico (Espin et al., J. Agrìc. Food Chem. 2001, 49, 1187-1193; Espin et al., US Patent No. 2003/0180833). La sintesi procede in resa quantitativa in un mezzo acquoso, a pH neutro e a temperatura ambiente. Tuttavia, per ottenere 1 grammo di idrossitirosolo è necessario utilizzare 1 litro di soluzione acquosa tamponata; pertanto l’estensione a livello industriale richiede l’impiego di grandi volumi di solvente. Le possibili applicazioni in campo industriale dell’idrossitirosolo possono essere estese aumentandone la liposolubilità e la stabilità. Queste proprietà possono essere realizzate derivatizzando la funzione alcolica primaria con gruppi protettivi facilmente rimuovibili per idrolisi (acida, basica o enzimatica) e lasciando inalterata la funzione o-difenolica, cui sono attribuite le attività biologiche dell’idrossitirosolo (Rice-Evans at al., Free Rad. Biol. Med. 1996, 7, 933-956). The possible applications in the industrial field of hydroxytyrosol can be extended by increasing its liposolubility and stability. Il più semplice derivato lipofilo dell’idrossitirosolo è l’idrossitirosolo acetato (3,4-diidrossifenil etil acetato), recentemente identificato qualitativamente e determinato quantitativamente nell’olio di oliva (Gordon, et al., J. Agric. Food Chem. 2001, 49, 2480-2485). Di tale composto è stata anche studiata l’attività antiossidante che è risultata essere confrontabile a quello del'idrossitirosolo in oli ed emulsioni. Trova applicazioni come additivo in alimenti (quali burro e latte) e in composizioni farmaceutiche e cosmetiche (Alcudia Gonzalez et al. 2004, WO005237.1-32). J. Agric. Food Chem. 2001, 49, 2480-2485); più recentemente è stata riportata un'esterificazione enzimatica dell’idrossitirosolo utilizzando etil acetato come agente acilante e lipasi. Utilizzando catalizzatori acidi (ad esempio, acido solforico o acido ptoluensolfonico) ed agenti acilanti a catena lunga, a partire daH’idrossitirosolo, sono stati preparati esteri a catena lunga (Alcudia Gonzalez, et al. 2004, W0005237, 1-32; Trujillo et al. J. Agric. Food Chem. 2001, 49, 2480- 2485); more recently, an enzymatic esterification of hydroxytyrosol has been reported using ethyl acetate as an acylating agent and lipase. Using acid catalysts (e.g. sulfuric acid or ptoluenesulfonic acid) and long-chain acylating agents, starting from H'hydroxytyrosol, long-chain esters were prepared (Alcudia Gonzalez, et al. 2004, W0005237, 1-32; Trujillo et al. J. Agric. Food Chem. 2006, 54, 3779-3785).
In conclusione, da quanto finora riportato a discapito delle molteplici applicazioni industriali dell’idrossitirosolo risulta evidente la carenza di una metodologia sintetica che, partendo da un materiale commercialmente disponibile a basso costo o reperibile da fonti naturali, permetta di ottenere, in pochi passaggi e utilizzando reagenti non pericolosi, sia l'idrossitirosolo che i suoi derivati. Dopo 24 ore si osserva la completa scomparsa del substrato. Dopo aver portato la miscela di reazione a temperatura ambiente, si aggiungono 1.2 mmoli di DMP. Trascorsi 40 minuti, si aggiunge Na2S204(2 mmoli) e si lascia sotto agitazione magnetica per 5 minuti. Dopo aver allontanato il solvente per distillazione a pressione ridotta, il residuo viene solubilizzato con etil acetato e trattato con una soluzione satura di NaHC03. Dopo 8 ore si osserva la completa scomparsa del substrato. Dopo aver portato la miscela di reazione a temperatura ambiente, si aggiungono 2 mi di acqua e 1.2 mmoli di DMP. Trascorsi 50 minuti, si aggiunge Na2S204,(2 mmoli) e si lascia sotto agitazione magnetica per 5 minuti. Dopo aver allontanato il solvente per distillazione a pressione ridotta, il residuo viene solubilizzato con etil acetato e trattato con una soluzione satura di NaHC03. La fase acquosa viene estratta con etil acetato. Le fasi organiche riunite sono lavate con una soluzione satura di NaCI fino a neutralità, quindi seccate su Na2S04anidro.
