"Composizione farmaceutica per la somministrazione orale di acidi grassi omega polienoici ed uno o più principi attivi con essi incompatibili, e processo per la sua preparazione" "Pharmaceutical composition for the oral administration of polyenoic omega fatty acids and one or more active ingredients incompatible with them, and process for its preparation"
CAMPO DELL'INVENZIONE FIELD OF THE INVENTION
The present invention relates to the field of pharmaceutical compositions, and in particular to a new pharmaceutical composition for the oral administration of polyenoic omega fatty acids and one or more active ingredients incompatible with them.
STATO DELLA TECNICA STATE OF THE TECHNIQUE
Omega polyenoic fatty acids are long-chain polyunsaturated fatty acids, comprised between 18 and 22 carbon atoms; among these, essential are the omega-3-polyenoic acids, in which the first unsaturated bond is found between the third and fourth carbon atom, starting from the terminal methyl group, and the omega-6-polyenoic acids, in which the first unsaturated bond is found between the sixth and seventh carbon atom.
Among the omega-3-polyenoic fatty acids, in which fish oil is rich, the most common are eicosapentaenoic acid, generally indicated with the acronym EPA, and docosahexaenoic acid, indicated by the acronym DHA. Another important omega-3-polyenoic fatty acid, this of non-marine but vegetable origin, is alpha-linolenic acid, indicated by the acronym ALA.
It has now been widely demonstrated that omega polyene fatty acids compete with arachidonic acid for binding with the cyclooxygenase and lipooxygenase enzymes, generating a lowering of blood triglyceride levels; they also possess an antiplatelet and antithrombotic action due to their effect of reducing the synthesis of thromboxane A2; they promote vasodilation and increase bleeding time.
Thanks to their precious biological effects, omega polyenoic fatty acids are indicated in the prophylaxis of relapses after angioplasty and to reduce angina attacks, as well as in the treatment of hypertriglyceridemia, in association with a modification of the dietary regimen or even where the response to diets and other non-pharmacological measures alone, has proved inadequate.
In the vast majority of cases the dietary sources of omega polyenoic fatty acids are in fact not sufficient to obtain an efficient therapeutic response, and these acids must be taken in the form of pharmaceutical compositions.
The intake of omega polygenic fatty acids can allow cholesterol-lowering drugs, such as statins, such as simvastatin, to work more effectively, just as the fatty acids themselves can enhance the effects of drugs taken to thin the blood. such as platelet antiplatelet agents, for example acetylsalicylic acid.
Up to now, the hypothesis of the physical association in the same formulation of polyenoic omega fatty acids with one or more other active ingredients incompatible with them, such as simvastatin or acetylsalicylic acid, has been considered impracticable, due to incompatibility of the components that would lead to the formation of degradation substances.
As far as the Applicant is aware, no products have been made up to now that include these active ingredients in a single composition, but rather, to obtain the beneficial effects of their combination, it was necessary to resort to the administration of different formulations, each containing only one active ingredient.
On the contrary, the advantages of having a single composition for the simultaneous oral administration of omega polyenoic fatty acids with active ingredients incompatible with them, especially in terms of patient compliance, are evident, and therefore the need is felt to point a composition of this type.

The two active ingredients present in the pharmaceutical composition of the invention - fatty acids on one side and one or more active ingredients incompatible with them on the other - are kept isolated from each other within the same dosage unit; in particular, the active ingredient present in a higher dosage, i.e. the fatty acid, is encapsulated, for example in a soft or hard gelatin capsule, while the second active ingredient, possibly in mixture with one or more other active ingredients, is evenly distributed around the capsule, by one of the two preparation processes described below.
According to the invention, the present compositions typically contain within the capsule from 100 to 1,500 mg of fatty acids, preferably about 1,000 mg. Such fatty acids are preferably selected from the group consisting of omega-3-polyenoic fatty acids, omega-6-polyenoic fatty acids, and their mixtures, more preferably they are mixtures of omega-3-polyenoic fatty acids.
