Abstract

Prepn. of polynucleotides comprises coupling of (a) a nucleotide having a masked 3'-phosphate diester group with (b) a nucleoside or nucleotide having an available, free 5'-hydroxy gp. in the presence of a coupling agent is characterised in that (a) is used in molar excess (pref. 2:1) w.r.t. (b) so that practically all of (b) is consumed in the reaction. Used in production of alignonucleotides partic. oligodesoxyribonucleotides having a defined sequence of four different nucleotides which makes it possible to insert spent DNA segments in plasmide DNA to code it e.g. for stimulating the production of peptidic (peptide) hormones having specific amino acid e.g. TTT polynucleotide sequence stimulates phenylalanine production, GGA sequence stimulates glycine (where T=thymine, A=adenine and G=guanine).
