Abstract

The present invention provides novel polynucleotide sequences encoding recombinant human paraoxonase 1 enzymes, having enhanced catalytic activities towards a wide range of substrates including pro-atherogenic and pro-inflammatory molecules, quorum sensors and neurotoxic organophosphorus compounds, for high level expression in Escherichia coli. The invention also provides a novel method to produce the recombinant enzymes in highly pure and active form in high yield by refolding the recombinant enzymes from the inclusion. The molecules disclosed in the present invention can be used as therapeutic agents against PON1-associated disease or conditions. The molecules can also be used as detoxifying and decontaminating agents in numerous terrorist, drug, and environmental scenarios involving military and civilian welfare.
