Description
CLAIMS: 1.
A method for producing a composition, comprising the steps of: dry milling a solid biologically active material and a millable grinding matrix in a mill comprising a plurality of milling bodies, for a time period sufficient to produce particles of the biologically active material dispersed in an at least partially milled grinding material wherein the biologically active material is metaxalone.
The method of Claim 1 where the composition produced by said method comprises particles of the biologically active compound at or above a volume fraction of 25 v/v%.
The method of claims 1 or 2, wherein the average particle size, determined on a particle number basis, is equal to or less than a size selected from the group consisting of: 2000 nm, 1900 nm, 1800nm, 1700nm, 1600nm, 1500nm, 1400nm, 1300nm, 1200 nm, 1100nm, 1000nm, 900nm, 800nm, 700nm, 600nm, 500nm, 400 nm, 300nm, 200nm and 100 nm.
The method of claims 1 or 2, wherein the particles have a median particle size, determined on a particle volume basis, equal or less than a size selected from the group consisting of: 20000nm, 15000nm, 10000 nm, 7500nm, 5000nm, 2000 nm, 1900 nm, 1800nm, 1700nm, 1600nm, 1500nm, 1400nm, 1300nm, 1200 nm, 1100nm, 1000nm, 900nm, 800nm, 700nm, 600nm, 500nm, 400 nm, 300nm, 200nm and 100 nm.
The method of claim 4, wherein the percentage of particles, on a particle volume basis, is selected from the group consisting of: 50 %, 60%, 70%, 80%, 90%, 95% and 100 % less than: a. 2000nm (% < 2000 nm); or b. 1000nm (% < 1000 nm); or is selected from the group 0%, 10%, 20%, 30%, 40%, 50 %, 60%, 70%, 80%, 90%, 95% and 100 % less than: c. 500nm (% < 500 nm); d. 300nm (% < 300 nm); or e. 200nm (% < 200 nm).
The method of claim 4, wherein the Dx of the particle distribution, as measured on a particle volume basis, is selected from the group consisting of less than or equal to ,000nm, 5000nm, 3000nm, 2000nm, 1900 nm, 1800nm, 1700nm, 1600nm, 1500nm, 1400nm, 1300nm, 1200 nm, 1100nm, 1000nm, 900nm, 800nm, 700nm, 600nm, 500nm, 400 nm, 300nm, 200nm, and 100 nm; wherein x is greater than or equal to 90.
The method of any preceding claim, wherein the milling time period is a range selected from the group consisting of: between 10 minutes and 2 hours, between 10 minutes and 90 minutes, between 10 minutes and 1 hour, between 10 minutes and 45 minutes, 93 . between 10 minutes and 30 minutes, between 5 minutes and 30 minutes, between 5 minutes and 20 minutes, between 2 minutes and 10 minutes, between 2 minutes and 5 minutes, between 1 minutes and 20 minutes, between 1 minute and 10 minutes, and between 1 minute and 5 minutes.

The method of any preceding claim, wherein the dry milling is undertaken in a mechanically agitated attritor mill (horizontal or vertical), vibratory mill or nutating mill, wherein the milling medium is steel balls having a diameter selected from the group consisting of: between 1 and 20 mm, between 2 and 15 mm and between 3 and 10 mm.
The method of any preceding claim, wherein the total combined amount of biologically active material and grinding matrix in the mill at any given time is equal to or greater than a mass selected from the group consisting of: 200 grams, 500 grams, 1 kg, 2kg, 5 kg, 10 kg, 20 kg, 30 kg, 50 kg, 75 kg, 100kg, 150 kg, 200 kg.

The method of claim 10, wherein the concentration of the single material or the major component in a mixture of two or more materials is selected from the group consisting of: 5 - 99 % w/w, 10 - 95 % w/w, 15 - 85 % w/w, of 20 — 80% w/w, 25 - 75 % w/w, 30 - 60% w/w, 40 -50% w/w and the concentration of the second or subsequent material is selected from the group consisting of: 5 - 50 % w/w, 5 - 40 % w/w, 5 - 30 % w/w, of 5 — % w/w, 10 - 40 % w/w, 10 -30% w/w, 10 -20% w/w, 20 — 40% w/w, or 20 - 30% w/w or if the second or subsequent material is a surfactant or water soluble polymer the concentration is selected from 0.1 -10 % w/w, 0.1 -5 % w/w, 0.1 -2.5 % w/w, of 0.1 — 2% w/w, 0.1 -1 %, 0.5 —5% w/w, 0.5 -3% w/w, 0.5 -2% w/w, 0.5 - 1.5%, 0.5 -1 % w/w, of 0.75 — 1.25 % w/w, 0.75 -1% and 1% w/w.

A composition comprising particles of metaxalone dispersed in at least two partially milled grinding materials, wherein the particles have at least one of a. a median particle size as measured on a particle volume basis is less than or equal to a size selected from the group consisting of 2000nm, 1900 nm, 1800nm, 1700nm, 1600nm, 1500nm, 1400nm, 1300nm, 1200 nm, 1100nm, 1000nm, 900nm, 800nm, 700nm, 600nm, 500nm, 400 nm. 300nm, 200nm and 100 nm; and b. an average particle size as measured on a particle number basis is less than or equal to a size selected from the group consisting of 2000nm, 1900 nm, 1800nm, 1700nm, 1600nm, 1500nm, 1400nm, 1300nm, 1200 nm, 1100nm, 1000nm, 900nm, 800nm, 700nm, 600nm, 500nm, 400 nm, 300nm, 200nm and 100 nm.
The composition of claim 25, wherein the Dx of the particle distribution, as measured on a particle volume basis, is selected from the group consisting of less than or equal to 3000nm, 2000nm, 1900 nm, 1800nm, 1700nm, 1600nm, 1500nm, 1400nm, 1300nm, 1200 nm, 1100nm, 1000nm, 900nm, 800nm, 700nm, 600nm, 500nm, 400 nm, 300nm, 200nm, and 100 nm; wherein x is greater than or equal to 90.

The composition of claim 28, wherein the composition comprises, w/w, metaxalone 20- 45%, poloxamer 407 0-3%, sodium lauryl sulfate O-3%, and lactose monohydrate 50- 80%.
The composition of claim 29, wherein the composition comprises, w/w, metaxalone %, poloxamer 407 1.5%, sodium lauryl sulfate 1.5%, and lactose monohydrate 67%.
The composition of claim 25 which, upon administration to a subject, provides improved pharmacokinetic and/or pharmacodynamic properties compared with a standard reference metaxalone composition as measured by at least one of speed of absorption, dosage potency, efficacy, and safety.

Claims (27)
1. A method for producing a composition, comprising the steps of: dry milling a solid biologically active material, at least one milling aid and a millable 5 grinding matrix in a mill for a time period sufficient to produce milled particles of the biologically active material dispersed in an at least partially milled grinding matrix, wherein the biologically active material is metaxalone, wherein the milled particles of metaxalone have a median particle size determined on a particle volume basis, or an average particle size determined on a 10 particle number basis, equal to or less than 5000nm, and wherein the total combined amount of metaxalone, milling aid and grinding matrix in the mill at a given time is equal to or greater than 200 grams.
2. The method of claim 1, wherein the Dx of the particle size distribution, as measured on a particle volume basis, is selected from the group consisting of less than or equal to 15 10,000nm, wherein x is greater than or equal to 90.
3. The method of any one of the preceding claims, wherein the milling time period is a range selected from between 1 minute and 2 hours.
4. The method of any one of the preceding claims, wherein the dry milling is undertaken in a jet mill, rod mill, roller mill, crusher mill, mechanically agitated attritor mill (horizontal or 20 vertical), vibratory mill or nutating mill.
5. The method of any one of the preceding claims, wherein the milling bodies are steel balls.
6. The method of any one of the preceding claims, wherein the total combined amount of biologically active material, milling aid and grinding matrix in the mill at a given time is 25 equal to or greater than a mass selected from the group consisting of: 500 grams.
7. The method of any one of the preceding claims, wherein the grinding matrix comprises a single material or a combination of two or more materials in any proportion.
8. The method of any one of the preceding claims, wherein the grinding matrix is selected from the group consisting of: polyols, sugar alcohols, monosaccharides, disaccharides, 30 trisaccharides, polysaccharides, carbohydrates, enteric polymer coatings, milk products, organic acids and conjugate salts of organic acids, hydrogen carbonates of pharmaceutically-acceptable alkali metals, ammonium salts and inorganic substances.
9. 10. The method of any one of the preceding claims, wherein the at least one milling aid is selected from the group consisting of: colloidal silica, a surfactant, a polymer, a stearic acid and derivatives thereof.

12. The method of any one of claims 10 or 11, wherein the surfactant is sodium lauryl 25 sulfate, or Poloxamer 407, or both.
13. The method of any one of the preceding claims, wherein the grinding matrix comprises at least one of lactose anhydrous, lactose monohydrate or mannitol.
14. The method of any one of the preceding claims, wherein the milling aid has a concentration selected from the group consisting of: 0.1 -10 % w/w. 30

The method of any one of the preceding claims, wherein at least one facilitating agent is used, wherein the at least one facilitating agent is selected from the group consisting of: surfactants, polymers, binding agents, filling agents, lubricating agents, sweeteners, flavouring agents, preservatives, buffers, wetting agents, disintegrants, effervescent agents, agents that may form part of a medicament, including a solid dosage form, 35 povidone (PVP), crosslinked PVP, cross linked carmellose, sodium starch glycolate, povidone K12, povidone K17, povidone K25, povidone K29/32, and povidone K30.

17. The method of any one of the preceding claims, wherein the volume fraction of the biologically active material in the mill during milling is at or above a volume fraction of 15 v/v%.
18. A composition comprising metaxalone, produced by the method of any one of claims 1- 5 17.
19. A pharmaceutical composition comprising the composition according to claim 18.
20. A dosage form comprising milled particles of metaxalone produced according to the method of any one of claims 1-17.
21. The dosage form of claim 20, formulated as a single or a multi-dose dosage form. 10
22. The dosage form of claim 20 or claim 21, comprising 100mg, 200mg, 225mg, 400mg or 450mg of milled metaxalone particles.
23. The dosage form according to any one of claims 20 to 22 formulated as an oral suspension, a capsule or a tablet.
24. The dosage form according to claim 23, wherein: 15 (i) administration of a single dose of the dosage form to a human subject provides a C greater than that of a standard conventional metaxalone composition when max normalised to the same dosage as the standard conventional metaxalone composition; (ii) administration of a single dose of the dosage form to a human subject in oral suspension, capsule or tablet form exhibits in comparative pharmacokinetic testing with a 20 standard conventional metaxalone composition, a C of at least 5% greater to at least max 150% greater than the C exhibited by the standard conventional metaxalone composition max when normalised to the same dosage as the standard conventional metaxalone composition; (iii) administration of a single dose of the dosage form or the pharmaceutical 25 composition to a human subject provides an AUC greater than that of a standard conventional metaxalone composition when normalised to the same dosage as the standard conventional metaxalone composition; (iv) administration of a single dose of the dosage form to a human subject exhibits in comparative pharmacokinetic testing with a standard conventional metaxalone 30 composition, an AUC of at least 5% to at least 150% greater than the AUC exhibited by the standard conventional metaxalone composition when normalised to the same dosage as the standard conventional metaxalone composition; (v) administration of a single dose of the dosage form to a human subject provides a T less than that of a standard conventional metaxalone composition; max 35 (vi) administration of a single dose of the dosage form to a human subject exhibits in comparative pharmacokinetic testing with a standard conventional metaxalone composition, a T less than the T exhibited by the standard conventional max max metaxalone composition; and/or 96 256841/3 (vii) administration of a single dose of the dosage form to a human subject provides a median T of less than 5 hours. max
25. The dosage form of any one of claims 20 to 24, wherein administration of a single dose of the dosage form or the pharmaceutical composition to a human subject suffering from 5 acute pain provides perceptible pain relief to the human subject.
26. The pharmaceutical composition of claim 19 for use for the treatment of a human.
27. A method for manufacturing a pharmaceutically acceptable dosage form comprising the step of combining a therapeutically effective amount of metaxalone prepared by a method according to any one of the claims 1 to 17 together with a pharmaceutically 10 acceptable carrier to produce a pharmaceutically acceptable dosage form. 97
