Abstract
A mutein of basic fibroblast growth factor (bFGF) is claimed Also claimed is a recombinant DNA having a base sequence which codes for a mutein of bFGF and a transformant harbouring a vector contg. a recombinant DNA having a base sequence which codes for a mutein of bFGF. - The prodn. of the structural gene which encodes the mutein is carried out by e.g. (a) hybridising with a mutagenic digonucleotide primer a single-stranded DNA comprising 1 strand of the structural gene of bFGF, (b) elongating the primer using DNA polymerase to form a mutational heteroduplex and (c) replicating this mutation heteroduplex. The resulting mutational heteroduplex is used to transform an infectable host organism or cell. In the replication of the heteroduplex by the host, progenies are produced from both chains. Following the replication, the mutant gene is isolated from a progeny of the mutant's chain and is inserted in an approp. vector which is used to transform an approp. host organism or cell.
