Abstract

A method for stimulating a therapeutic immune response in a subject comprising introducing to the subject or to a target gp. of cells from the subject, an expressible polynucleotide encoding a eukaryotic heat shock protein (HSP) is claimed. Also claimed are: (1) a method for stimulating a prophylactic immune response against recurrence of a disease from which a subject is suffering,comprising administering to the subject or to a target gp. of cells from the subject harbouring an antigen associated with the disease, an expressible polynucleotide encoding a eukaryotic HSP; (2) a recombinant vector comprising: (a) a promoter system effective to promote expression of the vector in mammalian cells; and (b) a region encoding a cytosolic HSP to which an amino terminal signal sequence effective to promote uptake of the expressed cytosolic HSP by the endoplasmic reticulum has been added; (3) a method for stimulating an immune response to an antigen present in a target gp. of mammalian cells, comprising administering to the target gp. an expressible polynucleotide, where expression of the polynucleotide is effective to increase the amt. of antigen-associated HSP present in the cells.
