Abstract

Η παρούσα εφεύρεση αναφέρεται σε μια μέθοδο πολλαπλού ποσοτικού προσδιορισμού αλληλουχιών νουκλεϊκών οξέων (αλληλουχίες-στόχοι) με εκθετική ενίσχυση τους, παρουσία συναγωνιστικών αλληλουχιών. Η μέθοδος αποτελείται από τα εξής στάδια: Ανάμιξη των αλληλουχιών-στόχων με κατάλληλα σχεδιασμένους συναγωνιστές, πολλαπλή εκθετική ενίσχυση των αλληλουχιών-στόχων και των συναγωνιστών και πολλαπλή ποσοτικήφθορισμομετρική δοκιμασία υβριδοποίησης χρησιμοποιώντας μίγμα διακριτών πληθυσμών νανο- ή μικροσωματιδίων συζευγμένων με ολιγονουκλεοτίδια-ανιχνευτές. Τα νανο- ή μικροσωματίδια διαφοροποιούνται είτε φασματικά ή κατά μέγεθος ή κατά το σχήμα και η ταξινόμηση τους πραγματοποιείται με κυτταρομετρία ροής. Οι συναγωνιστές έχουν τις ίδιες θέσεις πρόσδεσης εκκινητών με τις αντίστοιχες αλληλουχίες-στόχους. Τα προϊόντα εκθετικής ενίσχυσης για κάθε αλληλουχία-στόχο και τον αντίστοιχο συναγωνιστή έχουν (κατά προτίμηση) το ίδιο μέγεθος, αλλά διαφέρουν ως προς ένα τμήμα της αλληλουχίας, ώστε να είναι εφικτόςο προσδιορισμός των προϊόντων με δοκιμασία υβριδοποίησης. Ο λόγος των εντάσεων φθορισμού που λαμβάνονται από την αλληλουχία-στόχο και το συναγωνιστή είναι συνάρτηση της αρχικής ποσότητας της αλληλουχίας-στόχου στο μίγμα πριν από την εκθετική ενίσχυση.
