Abstract

2,4 - Diamino - 6 - hydroxy - 7,8 - dihydropteridine is made by partial reduction of 2,4-diamino - 6 - hydroxypteridine or 2,4 - diamino-6,7-dihydroxypteridine by means of catalytic hydrogenation or nascent hydrogen. The 2,4-diamino-6-hydroxypteridine starting material may be produced by reacting 2,4,5,6-tetramino-pyrimidine with free glyoxylic acid in the presence of dilute mineral acid (below 6N). Pharmaceutical preparations active as inhibitors of the biochemical synthesis of folic acid comprise 2,4-diamino-6-hydroxy-7,8-dihydropteridine and an excipient, suitably in the form of tablets, capsules, cachets, ampoules, suppositories, powder, granules, solutions, suspensions or ointments, or as sterile formulations for parenteral use.
