Abstract

The invention comprises suphonamides of the formula <FORM:0985610/C2/1> wherein R1 and R2 are each a chlorine atom or an alkoxy radical of 1-5 carbon atoms, and their preparation by either (a) reacting a salt of a benzenesulphonamide p-substituted by an amino group or a nitrogen containing group convertible to amino, such as acetylamino, benzylideneamino, carbalkoxy, phenylazo or nitro, with an appropriate pyrimidine compound bearing a halogen, e.g. Cl, or an alkylmercapto or alkylsulphonyl radical in the 4-position and, in any order, converting the p-substituent to an amino group by reduction or hydrolysis, if necessary, and, if desired, replacing a chlorine atom R1 and/or R2 by an alkoxy radical of 1-5 carbon atoms, or (b) reacting a functional derivative of a benzenesulphonic acid p-substituted by a group convertible to amino, e.g. a p-nitro- or p-acetylamino-benzenesulphonyl chloride, with an appropriate 4-aminopyrimidine and, in any order, converting the p-substituent to an amino group by hydrolysis or reduction and, if desired, replacing a chlorine atom R1 and/or R2 by an alkoxy radical. 4 - Amino - 2,6 - dimethoxy - 5 - chloropyrimidine is made by reaction of 4-amino-2,5,6-trichloro-pyrimidine with sodium methoxide. 4 - Methylsulphonyl - 2,6 - dimethoxy - 5-chloropyrimidine is made by treating 4-methylthio - 2,5,6 - trichloro - pyrimidine with sodium methoxide and oxidizing the resulting 4-methylthio - 2,6 - dimethoxy - 5 - chloro - pyrimidine with peracetic acid. 2 - Methoxy - 4,5,6 - trichloro - pyrimidine is made by the action of phosphorus oxychloride on 5 - chloro - 4,6 - dihydroxy - 2 - methoxy-pyrimidine. Pharmaceutical preparations, having chemotherapeutic activity, comprise the above sulphanilamides, or their salts with pharmaceutically acceptable bases, e.g. alkali metals, calcium, magnesium, ethylamine, dimethylamine, diethylamine morpholine, ethylenediamine, aminoethanol, diethylaminoethanol or di- or triethanolamine, together with a carrier. The preparations may suitably take forms suitable for internal or external use such as tablets, dragees, suppositories, syrups, powders or capsules or aqueous solutions for injection.
