Abstract

A sustained release, orally administrable pharmaceutical tablet is prepared by substantially completely coating by spraying a granulation mixture comprising a "high dosage" medicament and a binding agent therefor with a sustained release lipid material resistant to disintegration and slowly dispersable in the gastro-intestinal tract to form coated granules which are thereafter tableted. The lipid material may be sprayed as a solution in a volatile solvent and may be a wax, a fatty acid, alcohol or ester, many examples of which are given, including animal, vegetable and mineral waxes, esters of C12-31 fatty acids with C12-31 alcohols, C10-22 fatty acids, C10-22 fatty alcohols, mono-, di- and tri-glyceryl esters of C10-22 fatty acids and hydrogenated oils. Specified medicaments are those administered in 0.3 to 1 g. doses such as tranquilizers, e.g. chlorpromazine, diuretics, e.g. aminophylline, sedatives, e.g. secobarbital, antibiotics, e.g. tetracycline, vitamins, hematinics, antacids and analgesics, e.g. aspirin.
