Abstract

The invention relates to a drug in the form of a homogeneous ultra-fine powder for administration by inhalation at least two-thirds, preferably 80%, of whose weight consists of particles within the range 2-6m , preferably 3m , mean diameter and which contains substantially no particles larger than 10m , preferably 7m , mean diameter and a negligible proportion by weight of particles smaller than 1m mean diameter. Specified drugs are chymotrypsin or isoprenaline sulphate. The drug in homogeneous ultra-fine powder form is prepared by forming a suspension of the drug in finely divided form by precipitating the drug from a solution thereof in a solvent therefor by addition of a solvent miscible with said solvent but in which the drug is insoluble, freezing the suspension, if desired after washing and/or concentration, to a microcrystalline form and subsequently removing the frozen liquid, by distillation, under reduced pressure. Homogeneity is improved by high-speed stirring during mixing of the two liquids, subjecting the mixed liquids to ultrasonic vibrations or seeding with nuclei from a previous run. The powdered drug may also be prepared by forming a medicinally inert carrier, e.g. a low molecular weight dextran fraction as above and absorbing the drug therein from solution and freeze drying as above. Specified solvents employed are water, water-miscible solvents, e.g. methanol, ethanol, isopropanol, tertiary butanol or acetone or waterimmiscible solvents, e.g. ether or benzene. The wet preparation obtained by precipitation or absorbtion on a preformed carrier precipitate may prior to freezing be washed, concentrated by filtration or decantation with another solvent having different physical characteristics to that in which the powder is suspended, i.e. any liquid in which the drug and inert carrier, if present, are insoluble.
