Abstract
Catalase is recovered from the cells of Micrococcus lysodeikticus by lysing a suspension of the cells in a 0.5% to 2.0% solution of sodium chloride containing a lysing enzyme, precipitating the lysed cells and proteinaceous material by adding ethanol to the lysed suspension in such a proportion as to bring the concentration of ethanol to between 40 and 52% by volume (calculated at 95% by volume ethanol), separating the supernatant liquid, and adding ethanol to the separated liquid in such a proportion as to bring the concentration of ethanol to between 70 and 85% by volume (calculated as 95% by volume ethanol) to precipitate catalase and recovering the catalase. The pH of the sodium chloride solution is preferably maintained between 6.0 and 9.0, the preferred enzyme is lysozyme and the lysis is preferably carried out at 30 DEG to 45 DEG C.
