Abstract
The invention comprises 16-methyl steroids of the general formulae: <FORM:0909614/IV (b)/1> <FORM:0909614/IV (b)/2> and their corresponding D 1,4-analogues (R and R1 are both hydrogen or a hydrocarbon carboxylic acyl group of less than 12 carbon atoms, or R is hydrogen and R1 is a hydrocarbon carboxylic acyl group, X is keto or b -hydrogen, and Y is hydrogen, fluorine, chlorine or bromine) and processes for their preparation from 16a -methyl-D 5-pregnene-3b ,17a -diol-20-one by tribromination to the 5,6,21-tribromo derivative treatment with sodium iodide and then potassium acetate followed by oxidation of the 3b -ol and isomerisation of the D 5-bond to yield 16a -methyl-D 4-pregnene 17a ,21-diol-3,20-dione 21-acetate, which may be dehydrogenated with selenium dioxide or by micro-organisms to the D 1,4-analogue. The 9a -halo compounds are subsequently prepared by introduction of an 11a -hydroxy group with Rhizopus nigricans or Aspergillus niger, dehydration to a D 4,9(11)-compound, treatment with N-bromoacetamide, conversion of the bromohydrin to the 9b ,11b -epoxide and finally cleavage of the oxide with HF or HCl.
