Abstract

A polypeptide, enzymal in nature and useful in the treatment of arthritis (Group VI), has a molecular weight of 10,000 to 11,000 and approximately two glutamic acid residues per aspartic acid residue and also contains a minor proportion of arginine, lysine and glycine residues, it contains 60,5% glutamic acid, 30,5% aspartic acid, 3,3% arginine, 3,3% lysine and 2,4% glycine and is obtained from beef liver. The beef liver has the connective tissue removed manually and is then treated for the removal of proteins, albuminoids, histones, basic amino acids, proteoses and peptones, a water-miscible alcohol or ketone is added to the liquid residue in up to 60% concentration, the material allowed to stand at a low temperature for some hours until it forms a top liquid layer of an orange-red colour separated by a solid water of a grey or olive green colour from a lower liquid layer of a greenish-yellow colour, separating the upper liquid layer, adding thereto a water-miscible solvent which is an alcohol or ketone and cooling to a low temperature for some hours to form a flucculent, slightly pinkish precipitate, separating the solid from the liquid, macerating the solid with an aqueous liquid having a pH of 7 to 9, separating the liquid obtained and subjecting it to dialysis, and thereafter heating the dialysed liquid to 43 to 49 DEG C. until a heavy flocculent precipitate forms, separating the liquid, subjecting it to distillation at a reduced pressure and a temperature not exceeding 43 DEG C. until a solid residue is formed.ALSO:A polypeptide useful in the treatment of arthritis and having a molecular weight of 10,000 to 11,000 and approximately two glutamic acid residues per aspartic acid residue and also containing a minor proportion of arginine, lysine and glycine residues, contains 60.5% glutamic acid, 30.5% aspartic acid, 3.3% arginine, 3.3% lysine and 2.4% glycine and is isolated from beef liver. The beef liver has its connective tissue removed manually and is then treated for the removal of proteins, albuminoids, histones, basic amino acids, proteoses and peptones, a water-miscible alcohol or ketone is iadded to the liquid residue in a concentration not exceeding 60% by volume, the material is allowed to stand at a low temperature, e.g. 3 DEG C. for some hours, e.g. 12 hours until it forms a top liquid layer of an orange-red colour separated by a solid wafer of a grey or olive-green colour from a lower liquid layer of a greenish-yellow colour, separating the upper liquid layer, adding thereto a water-miscible alcohol or ketone, cooling to a low temperature, e.g. 3 DEG C. to form a flocculent slightly pinkish precipitate, separating the solid from the liquid, macerating the solid with an aqueous liquid having a pH of 7 to 9, separating the liquid obtained and subjecting it to dialysis, and thereafter heating the dialysed liquid to 43 DEG to 49 DEG C. until a heavy flocculent precipitate forms, separating the liquid, subjecting it to distillation at a reduced pressure and temperature not exceeding 43 DEG C. until a solid residue is formed.
