Abstract

The invention comprises a compound having the formula: <FORM:0852647/IV(b)/1> wherein X is a fluorine, chlorine or bromine atom and R is b -hydroxyl or keto, or a 21-ester thereof with a mono- or di-carboxylic acid, said mono- or di-carboxylic acids containing only carbon, hydrogen and oxygen up to a total of ten carbon atoms or an alkali or alkaline earth metal salt of said dicarboxylic acid esters, and a process for the preparation of a compound having the formula I which comprises contacting a compound having the formula: <FORM:0852647/IV(b)/2> wherein R and X are as defined above and R1 is hydrogen or an acyl hydrocarbon containing up to 10 carbon atoms, to the oxidising enzymes of a micro-organism known in the literature to introduce a double bond in the 1-position of steroids. As micro-organisms there may be employed species of the genus Mycobacterium, e.g. Mycobacterium species 607, M. berolinese, M. lacticola, M. thamnopheos, M. smegmatis and M. phlei, or of the genus Protaminobacter. Illustrative products of the invention are D 1,4-pregnadiene-9a -fluoro-11b ,14a ,17a ,21-tetrol-3, 20-dione, D 1,4-pregnadiene-9a -fluoro-14a ,17a ,21-triol-3,11, 20-trione, D 1,4-pregnadiene-9a -fluoro-11b ,14a ,17a ,21-tetrol - 3,20 - dione21 - acetate,D 1,4 - pregnadiene-9a -fluoro-14a ,17a ,21 - triol - 3,11,20-trione 21-acetate and D 1,4-9a -fluoro-11b ,14a ,17a ,21-tetrol-3,20-dione 21-propionate. Specification 800,644 is referred to.ALSO:A pharmaceutical composition comprises a compound having the formula: <FORM:0852647/VI/1> wherein X is a fluorine, chlorine or bromine atom and R is b -hydroxyl or keto or a 21-ester thereof with a mono- or di-carboxylic acid containing only carbon hydrogen and oxygen up to a total of 10 carbon atoms or an alkali or alkaline earth metal salt of said dicarboxylic acid esters, together with a pharmaceutically acceptable carrier. Such compositions for oral administration are in the forms of tablets, suspensions and elixirs; for topical application in the forms of ointments and salves, and for intra-articular injection aqueous suspensions may be employed.
