Abstract

The invention comprises arylsulphonamido thiadiazoles of the formula <FORM:0839316/IV (b)/1> wherein Ar is a phenyl radical substituted by at least one alkyl radical of 1 or 2 carbon atoms and R is a C1-4 alkyl or C2-4 alkoxyalkyl group or a methylthio-, phenyl or benzyl group; and their preparation by treating a 5-halo-1 : 2 : 4-thiadiazole with an alkyl-substituted benzene-sulphonamide in the form of its salt or in the presence of an acid acceptor, or by reacting a 5-amino-thiadiazole with a reactive functional derivative of an alkylbenzenesulphonic acid such as the chloride, bromide, fluoride or azide in the presence of an acid acceptor. Suitable acid acceptors include trialkylamines, pyridine, potassium carbonate and sodium carbonate. The compounds are used pharmaceutically in the treatment of liver diseases (see Group VI). Specified products include 5-o-, m- and p-methyl- and 5-o- and p-ethyl-benzenesulphon-amide derivatives of 3-methyl-, ethyl-, methyl-mercapto-, benzyl-, b -ethoxyethyl-, b -methoxy-ethyl- and phenyl-1 : 2 : 4-thiadiazoles. 5 - Chloro - 3 - b - alkoxyethyl - 1 : 2 : 4 - thiadiazole is made by reacting b -ethoxypropamidine hydrochloride, obtained from b -ethoxypropionitrile, with perchloromethyl mercaptan. The corresponding b -methoxyethyl compound is similarly prepared.ALSO:Therapeutic compositions for treatment of diseases of the liver comprise compounds of the formula <FORM:0839316/VI/1> wherein Ar is a phenyl radical substituted by at least one alkyl radical of 1 or 2 carbon atoms and R is an alkyl radical of 1-4 carbon atoms, an alkoxyalkyl radical of 2-4 carbon atoms or a methylthio-, phenyl or benzyl group (see Group IV (b)), and an inert solid or liquid pharmaceutical carrier. The preparations may suitably be injected intravenously in 10% solution or more preferably are administered orally in a dosage unit, e.g. tablets, containing 0.1-0.25 g. of active material and an inert solid pharmaceutical carrier.
