Abstract
The invention comprises retrocortin compounds of the general formula <FORM:0826362/IV (b)/1> in which R is a keto or b -hydroxy radical, and Y is a hydrogen or halogen atom, and a process for-their preparation by reacting tertiary butyl-acetyl chloride with the corresponding retrocortin derivative possessing a free 21-hydroxy group, the reaction being effected in the presence of a base and preferably at from 0-5 DEG C. Specified products include the 21-tertiary butyl-acetates of retrocortin, hydro-retrocortin, and their 9a -fluoro- and 9a -chloro derivatives. In examples: (1) retrocortin in pyridine is treated with t-butylacetyl chloride in chloroform, the mixture poured into water and retrocortin 21-t-butylacetate isolated with chloroform and crystallized from ethanol; similarly (2) to (4) are prepared the t-butylacetates of hydro-retrocortin, its 9a -fluoro derivative, and of 9a -fluororetrocortin. Specifications 823,940, 826,361 and 826,364 are referred to.ALSO:Pharmaceutical compositions useful in the treatment of rheumatic conditions comprise suspensions of the 21-t-butylacetates of retrocortin, hydroretrocortin, or their 9a -fluoroderivatives in a suitable vehicle, particularly an aqueous carrier, which may also contain surface active agents or suspending agents and which may further contain other esters of retrocortin and related compounds in addition to the t-butylacetate ester. A specified composition contains the 21-t-butylacetates of hydroretrocortin and 9-fluoro-hydroretrocortin in an aqueous medium. Specifications 823,940, 826,361 and 826,364, [all in Group IV (b)], are referred to.
