Abstract

A pharmaceutical tablet, which may be substantially flat-faced and cylindrical in form, has an exposed immediate release portion to provide an immediate therapeutic level of medicament and a sustained release portion of surface area 2.5 to 25 times that of its side surface to provide a substantially uniform release of medicament for a period greater than four hours. The medicament in the immediate release portion may be dispersed in a filler e.g. talc, terra alba, stearic acid, and milk of sugar, and in the sustained release portion, the medicament may be dispersed in a time delay material e.g. starch, sugar, alginic acid, colloidal magnesium aluminium silicate. The tablet is formed by mixing the medicament with the filler or time delay material, granulating e.g. with water, acacia solution, starch paste or gelatin solution, comprising the granules of the immediate release material to form layers which are then tabletted with granules of the sustained release material to form a laminated tablet. Medicaments specified are aminophylline, dextro-amphetamine sulphate, phenobarbital, amobarbital, chlorprophenpyridamine maleate, thenylpyramine hydrochloride, homatropine methyl bromide, atropine sulphate, hyoscyamine sulphate, scopolamine hydrobromide. Time delay materials are fatty alcohols having from 12 to 31 carbon atoms e.g. lauryl, cetyl, stearyl, myristyl, myricyl, arachyl, carnaubye, ceryl alcohol; esters of fatty acids having 12 to 31 carbon atoms with these alcohols e.g. myricyl, p cetyl, ceryl, stearyl palmitate, myricyl and ceryl cerotate, myricyl melissate, cetyl or stearyl myristate and lauryl laurate; mono-, di-or tri-glyceryl esters of fatty acids having 10 to 22 carbon atoms e.g. glyceryl mono-, di-, tri-palmitate, mono, di-, trilaurate, mono-, di-, tri-docosanoate, mono-, di-, tri-caprate, mono-, di-, tri-myristate, mono-, di-, tri-decenoate; hydrogenated castor, peanut and coconut oil; spermaceti, carnauba and beeswax.
