Abstract

An antibiotic "Viomycin" (Registered Trade Mark), is prepared from the fermentation broth obtained by cultivating a Viomycin-producing strain of Streptomyces puniceus, or a mutant thereof, in an aqueous nutrient medium containing carbohydrate, preferably adjusted to pH7, with submerged aerobic conditions, a temperature of 24 DEG to 30 DEG C. and 2 to 7 days fermentation. The culture medium contains carbohydrate, e.g. sugars, starch and glycerol; organic nitrogen, e.g. soybean meal and wheat gluten; growth substances, e.g. distillers' solubles; sodium chloride and buffering agents, e.g. calcium carbonate. The antibiotic may be isolated by (a) precipitation with acid fast dyes or (b) adsorption on a cationexchange resin. For (a) the preferred dye is Eriochrome Violet, others which may be used are: Erio Fast Cyanine, Pontamine Fast Orange, Alizarin Red S, Synthracine Blue, Pontamine Steel Blue, Polar Yellow, Methyl Orange and Pontamine Green. The dye salt formed may be treated, e.g. triethylamine sulphate in the presence of a solvent, e.g. methanol or methanol-acetone and the Viomycin sulphate filtered off. Alternatively a solution of the dye salt may be treated with an inorganic salt, e.g. Bacl or bacl2, and the precipitated metal dye salt removed. The antibiotic may be obtained from the resin eluate by freeze drying, or by the addition of solvents such as methanol and butanol. Two antibiotics are present in the fermentation broth; the major component Viomycin is isolated by the above methods. The preparation of the reineckate, picrate, b -naphthelene sulphonate, sulphate and hydrochloride of Viomycin is described. Viomycin is a basic antibiotic containing C,H,O and N and forming salts with organic and inorganic acids. The sulphate and hydrochloride (mpt. 265-8 DEG C.) are soluble in water, sparingly soluble in methanol, acetone and b -methoxyethanol. Viomycin sulphate [a ]25D = -31.3 DEG (H2O 1 per cent), E1 per cent1 cm. at 267.5 mm = 285. The micro organism S. puniceus is described.
