552,808. Pyridine derivatives. HOFFMANNLA ROCHE & CO. AKT.-GES., F. July 29, 1941, Nos. 9627, 9628, 9629 and 9630. Convention dates, Sept. 25, 1940, Oct. 4, 1940, Oct. 31, 1940 and Nov. 1, 1940. [Class 2 (iii)] 2 - Methyl - 4 - ethoxymethyl - 6 - hydroxypyridines having a carbalkoxy, carboxyamide or nitrile group in the 5-position, are prepared by reacting 2-amino-4-oxo-5-ethoxypentene-2 with malonic acid dinitrile, malonic acid alkyl esters, malonic acid diamide or cyanacetamide, preferably in presence of a solvent. The products are intermediates for the synthesis of Vitamin B6. The 2-amino-4-oxo-5-ethoxypentene-2 is prepared by reacting ethoxyacetylacetone with ammonium chloride in aqueous solution or with aqueous ammonia, and may be used without isolation. Examples are given of the interaction of the pentene with (1) malonic acid dinitrile in the cold or cyanoacetamide under heat to give the 5-cyano product, (2) malonic acid diethyl ester in the presence of sodium ethylate to form the 5-carboxylic acid ethyl ester, (3) malonic acid diamide to produce the 5-carboxylic acid amide.
