Abstract
The present disclosure relates to a novel analyte detection device and methods for detecting analytes of interest, such as allergens. In particular, the present disclosure provides a portable analyte detection device that is intended for on-site use by consumers. A fused split peroxidase-analyte binder complex is disclosed having a split peroxidase in at least two fragments, each fused to an analyte binder fragment capable of binding an analyte of interest.
Claims (19)
1. A fused split peroxidase-analyte binder complex comprising a split peroxidase, wherein the split peroxidase is at least two fragments, where each fragment is fused, optionally through a linker, to an analyte binder fragment consisting of a peptide or protein of interest which is capable of binding an analyte of interest, wherein the analyte is an allergen.
2. The fused split peroxidase-analyte binder complex of claim 1, wherein the split peroxidase is an ascorbic peroxidase.
3. The fused split peroxidase-analyte binder complex of claim 1, wherein the allergen of interest is a food allergen.
4. The fused split peroxidase-analyte binder complex of claim 3, wherein the food allergen is gluten and the analyte binder is a peptide that is capable of binding gluten.
5. An analyte detection device for detecting an analyte of interest in a sample which comprises: a fused split peroxidase-analyte binder complex comprising a split peroxidase, wherein the split peroxidase is at least two fragments, where each fragment is fused, optionally through a linker, to an analyte binder fragment consisting of a peptide or protein of interest, which is capable of binding an analyte of interest; and a substrate that is capable of undergoing catalysis by the fused split peroxidase- analyte binder complex once reconstituted in the presence of an analyte of interest, wherein the catalysis of the substrate is detectable.
6. The analyte detection device of claim 5, further comprising one or more buffers, and optional reagents.
7. The analyte detection device of claim 5, wherein the split peroxidase is an ascorbic peroxidase and the substrate is ascorbate.
8. The analyte detection device of claim 5, wherein the analyte of interest is an allergen.
9. The analyte detection device of claim 8, wherein the allergen is a food allergen.
10. The analyte detection device of claim 9, wherein the food allergen is gluten and the analyte binder is a peptide that is capable of binding gluten.
11. The analyte detection device of claim 5, wherein the catalysis of the substrate results in a colorimetric or fluorogenic signal which is detectable.
12. The analyte detection device of claim 5, wherein the device comprises: a sample collection apparatus; and a reaction vessel comprising the fused split peroxidase-analyte binder complex and the substrate.
13. The analyte detection device of claim 12, wherein the reaction vessel comprises: a casing; one or more compartments; and a container.
14. The analyte detection device of claim 13, wherein the reaction vessel contains a reaction viewing window.
15. The analyte detection device of claim 13, wherein the reaction vessel further comprises an absorbent material.
16. Use of the fused split peroxidase-analyte binder complex of any one of claims 1 - 4 for detection of an allergen in a sample.
17. The use of claim 16, wherein the sample is a food sample.
18. Use of the analyte detection device of any one of claims 5 - 14 for detection of an analyte of interest in a sample.
19. The use of claim 18, wherein the sample is a food sample.
