Abstract

1516338 Electro-chemical glucose determination OWENS-ILLINOIS Inc 25 Oct 1975 43955/75 Heading G1N [Also in Division C3] Glucose in an aqueous specimen is determined by passing a diluted specimen through a bed of immobilized glucose oxidase and electro-chemically determining the hydrogen peroxide formed. The glucose specimen, which may comprise blood serum, is injected into a continuously flowing aqueous buffer solution of pH 4-8 (preferably 5-7) and preferably containing sodium chloride for the reference electrode. The diluted specimen is passed through a bed of the glucose oxidase immobilized on particulate refractory oxide, cross-linked polydextron or an anion exchange resin and the resulting solution is passed to a polarographic cell where the hydrogen peroxide is determined and indicated as glucose concentration. The polarographic cell is tubular and may comprise a silver/silver chloride electrode or calomel electrode and a working electrode of platinum, palladium, gold, graphite, other carbon or other inert conductive material. Preferably a platinum counter electrode is also present. A voltage of 0.6V D.C. is applied between counter and reference electrode. The peak current through the measuring electrode is determined.
