Abstract

1505573 Piperine-derived hydroxyketones CIBA-GEIGY AG 31 July 1975 [31 July 1974 16 Dec 1974] 32051/75 Heading C2C The invention comprises compounds of formula and their salts, wherein R 1 is optionally substituted aryl; alk 1 and alk 2 are C 2-7 alkylene (each of which separates the N and CH which they connect by 2 carbon atoms); R 2 is OH or acyloxy; "Ph" is optionally substituted o-phenylene; n is 0 or 1; and (a) R 3 is H, OH or C 1-7 alkyl, R 5 is H or C 1-7 alkyl (or R 3 -R 5 constitute a further bond, and R 4 and R 6 are each H, or (b) R 3 R 4 C is CO, R 5 is H or C 1-7 alkyl and R 6 is H, or (c) R 5 R 6 C is CO, R 3 is H, OH or C 1-7 alkyl and R 4 is H. In examples, these compounds are prepared by reacting with 2 - (4 - piperidyl)isoindolinone, 2 - (4- piperidyl)isoindolindione or 2 - (4 - piperidyl)- 3,4 - dihydroisoquinolin(2H)- 1 - one, and, if desired, esterifying the product, in which R 2 is OH. Starting materials and intermediates otherwise prepared are 3 - (pyridylamino)isobenzofuranones, 2 - (4 - pyridylaminomethyl)benzoic acid, 2 - (4 - pyridyl)isoindolinone and 2 - (1- benzyl - 4 - piperidyl) - 3,4 - dihydroisoquinolin- (2H)-1-one. Therapeutic compositions having antihypertensive, vasodilatant, blood-pressure lowering, tachycardia inhibiting, noradrenolytic, antiarrhythmic and positive inotropic activity, comprise compounds of the above formula. The Specification lists further (optional) active ingredients.
