Abstract

1505400 Immunological reagent AMERICAN CYANAMID CO 3 July 1975 [1 Aug 1974] 28140/75 Heading G1B [Also in Division C3] A diagnostic reagent comprises chemically and serologically inert polystyrene latex particles of particle size in the range 0À05 to 1 microns having adsorbed on the surface a cross-linked proteinaceous substance which is immunologically active, for example tetanus toxoid or human chorionic gonadotropin cross-linked with N-ethyl-N'-(3- dimethyl aminopropyl)carbodiimide hydrochloride. The reagent may be prepared by mixing the antigen or antibody with the cross-linking reagent in a buffered aqueous medium, heating or allowing the mixture to stand to effect cross-linking, separating unreacted cross-linking agent, for example by dialysis, then mixing the cross-linked antigen or antibody with the polystyrene particles, or alternatively mixing the antigen or antibody, cross-linking agent and polystyrene particles together in the first step.
