Abstract

1499846 Testing for pyrogens BAXTER TRAVENOL LABORATORIES Inc 22 April 1975 [1 May 1974] 16509/75 Heading G1B A material is tested for an unknown pyrogen by (A) calibrating a purified solution of portions of Limulus lysate, which portions precipitate limulus protein in the presence of a pyrogen by:- (i) Making serially diluted dispersions of pyrogen (a plurality containing 0À39 ng/ml or less) (ii) Adding a given amount of limulus lysate solution to each dispersion. (iii) Measuring, photometrically, the amount of limulus protein precipitated, an abrupt increase in the amount of said protein indicating pyrogen. (B) Adding to said material said given amount of fresh limulus lysate solution, under conditions similar to those for A (ii) and measuring photometrically the precipitated protein, an amount equal to or greater than that of the abrupt increase in A (iii) indicating the presence of pyrogen. The precipitated pyrogen may be removed from its supernate for quantitative analysis. In step A (ii) the reaction mixture may be incubated, e.g. at 37‹C for 60 minutes.
