Abstract

1497316 Pituitary polypeptide compositions RESEARCH CORP 11 Nov 1975 [14 Nov 1974 3 Nov 1975] 46577/75 Heading A5B [Also in Division C3] A pharmaceutical composition, which may be used for the control of intestinal mobility and which also has vasoconstrictor activity comprises a pharmaceutically acceptable carrier and a compound which is selected from (1) a peptide designated coherin A having the following characteristics (i) contains glycine (ii) is soluble in water, slightly soluble in absolute ethanol, insoluble in acetone and diethyl ether (iii) has the following ultra-violet spectrum; # max, 274 nm (pH 1), 240 nm and 290 nm (pH 11); # min, 248 nm (pH 1), 233 nm and 272 nm (pH 11) (iv) has a molecular weight of 300 to 600 (v) has a melting point of 200 to 205‹C (vi) has an isoelectric point at pH 6.0 (vii) has an Rf value of 0.70 (n-butanol/pyridene/acetic acid/ water, 60:40:12:48 by volume); an Rf value of 0.33 (n-butanol/acetic acid/water, 4:1:5 by volume) by thin-layer chromatography on silica gel on glass; (2) a peptide designated coherin B having the following characteristics (i) contains glycine, serine and glutamic acid (ii) is soluble in water, slightly soluble in absolute ethanol, insoluble in acetone and diethyl ether (iii) has the following ultra - violet spectrum; # max 276 nm (pH 1), 239 nm and 290 nm (pH 11); # min 236 nm (pH 1), 253 nm and 276 nm (pH 11) (iv) has an isoelectric point at pH 8.5 (v) has an Rf value of 0.58 (nbutanol/pyridene/acetic acid/water, 60:40:12:48 by volume), an Rf value of 0.03 (n-butanol/ acetic acid/water, 4:1:5 by volume) by thinlayer chromatograph on silica gel on glass and pharmaceutically-acceptable salts of (I) and (2). The peptides (I) and (2) may be obtained from bovine posterior pituary.
