Abstract
1491178 Penicillin derivatives RICHARDSON-MERRELL Inc 25 June 1976 [22 July 1975] 26433/76 Heading C2C Novel compounds I and II (Z is a single bond, -CH 2 - or -CH 2 CH 2 - and R 1 is H, or (1-5 C alkanoyl)-oxymethyl) and salts thereof are prepared by condensation of a compound III or IV or functional derivatives thereof with a compound V (B is a blocking group) or a functional derivative thereof, followed by removal of the blocking group. Intermediates isolated are 2 - (2,3 - methylenedioxyphenyl)acetaldehyde and 3 - (2,3- methylenedioxyphenyl)propionaldehyde prepared by reaction of 2,3-methylenedioxybenzaldehyde and 2 - (2,3 - methylenedioxyphenyl)acetaldehyde respectively with triphenylmethoxymethylphosphonium chloride and phenyl lithium; 2,3-methylenedioxyphenyl glycine, 3-(2,3- methylenedioxyphenyl)alanine, 2-amino-4-(2,3- methylenedioxyphenyl)butyric acid and 2 - amino - 4 - (3,4 - methylenedioxyphenyl)butyric acid prepared by hydrolysis of an appropriate hydantoin derivative; the methyl acetoacetic ester enamine of D - (2,3 or 3,4)-methylenedioxyphenylglycine sodium salt, prepared by reaction of D - (2,3 or 3,4) - methylenedioxyphenylglycine with NaOCH 3 and methylacetoacetate; and 2,3- or 3,4 - methylenedioxyphenylalanyl chloride hydrochloride, prepared by reaction of PCl 5 with the free acid. Pharmaceutical compositions, useful as antibiotics, comprise a compound I or II together with a suitable excipient.
