1488619 Thieno-s-triazolodiazepines TAKEDA YAKUHIN KOGYO KK 25 March 1975 [5 April 1974] 12478/75 Heading C2C Novel thieno - s - triazolodiazepines of the general Formula I wherein each of K<SP>1</SP> and R<SP>2</SP> is a hydrogen atom or an alkyl group, optionally substituted by an alkyl-substituted amino, hydroxy or alkoxy group, or NR<SP>1</SP>R<SP>2</SP> is a heterocyclio group, each of R<SP>3</SP> and R<SP>4</SP> is a hydrogen atom or C 1-4 alkyl group or R<SP>3</SP> and R<SP>4</SP> together form an alkylene group, R<SP>6</SP> is a hydrogen atom or C 1-4 alkyl group, R is a phenyl group optionally having halogen, C 1-4 alkyl, trifluoromethyl or C 1-4 alkoxy substituents, Y is O, S or NH and A is an ethylene or trimethylene group optionally having C 1-4 alkyl substituents, are prepared (a) by reacting an N-(2-thienyl)-s-triazole of the general Formula IV wherein X is a halogen atom or a non-halogen active ester residue of a hydroxy group with H 2 N-A-YH, or (b) by reacting a reactive derivative of a carboxylic acid of the general Formula II with R<SP>1</SP>R<SP>2</SP>NH and H 2 N-A-YH. N - (2 - Thienyl) - s - triazoles (IV) are prepared by reacting a carboxylic acid (II) or a reactive derivative thereof with R<SP>1</SP>R<SP>2</SP>NH. Carboxylic acids (II) are prepared by diazotizing a 2-aminothiophene of the general Formula VII followed by treatment with wherein R<SP>6</SP> is a lower alkyl group and X<SP>1</SP> is a halogen atom, reacting the resulting hydrazone of the general Formula IX with ammonia, reacting the resulting amine of the general Formula X with X-CHR<SP>5</SP>-COX<SP>1</SP>, cyclizing the resulting amide of the general Formula XII with an acid or a base and hydrolysing the resulting ester of the general Formula II<SP>1</SP> Pharmaceutical compositions having pharmacological effects on the central nervous system comprise, as active ingredient, a thieno-striazolodiazepine (I), together with a pharmaceutically acceptable carrier or diluent therefor, and may be administered orally or parenterally.
