Abstract

1484031 Insolubilized proteins; modified synthetic polypeptides AJINOMOTO CO Inc 15 Oct 1974 44563/74 Headings C3H and C3R An immobilized biologically active protein (e.g. enzyme, antigen, antibody, peptide hormone, natural protein of animal or plant origin) comprises a polypeptide of repeating units of dicarboxylic acidic amino acids and, optionally, repeating units of neutral and/or basic α-amino acids, wherein at least one α- or #-carboxyl group of said acidic amino acid is converted to an acid azide group through which "the biologically-active protein" is covalently bonded. Biologically active protein excludes l-asparagine. The composite may also comprise an 'adhesive' (polyurethane, epoxy resin, polyester resin, polyvinyl chloride, polyvinyl acetate, polyvinylidene chloride, polyacrylate, polyamide). The polypeptide azide is preferably prepared by reaction of the polypeptide comprising units of the alkyl ester of the acidic amino acid with hydrazine, and treatment of the hydrazide with aqueous nitrous acid and/or nitrite solution. In the examples, the azides are prepared from poly - r - methyl - D - glutamate, copoly - (L- glutamic acid α-methyl ester/DL-methionine); the enzymes are urease, acylase, uricase; the adhesives are polyurethane diisocyanate/epichlorohydrin, 3,9 - bis - (3 - aminopropyl)- 2,4,8,10 - tetraoxospiro - [5,5] - undecane/epichlorhydrin, polyvinyl chloride. The product may be as filaments, or coated on to a substrate (glass beads, alumina particles). In comparative examples, the activities of the composites from azides of copoly-(L-methionine/L-glutamic acid r-methyl ester), carboxymethylcellulose and polymethacrylic acid with uricase are given.
