1482785 Lyophilised products E I DU PONT DE NEMOURS & CO 30 Oct 1974 47058/74 Addition to 1316522 Heading A5B An homogeneous lyophilised biologically active substance is prepared by a method which comprises (a) spraying a solution or colloidal suspension of the substance in a liquid, which will both freeze and vaporise from the solid state at a temperature at which the biological activity of the substance is not destroyed, in the form of droplets into a stirred boiling fluorocarbon refrigerant at below -20‹C to freeze the droplets, (b) subjecting the frozen droplets to a vacuum at a temperature such that substantially all the frozen liquid sublimes from the droplets; and (c) allowing the particles to come to room temperature and pressure. The liquid may be organio or water. The active substance may be a vitamin, hormone, tranquilizer, antibiotic, an enzyme, gelatine or other protein. The products of the Examples are bacitracin, polymyxin B, tetracycline, chlorpromazine testosterone, cholesterol and gelatine. The refrigerant may be dichlorofluoromethane. The product is in the form of spheres of diameter not greater than 1 mm and a "standard dissolution time" (defined) not exceeding 2 minutes, at least 90% of the particles being pores.
