Abstract
1482663 Encapsulated medicaments ROUSSEL-UCLAF 29 July 1974 [27 July 1973] 33380/74 Heading A5B [Also in Division B8] A process for the preparation of microbeads which comprise at least one water-insoluble or sparingly soluble pharmacuetically-active ingredient enclosed within a mantle composed of at least one methacrylic polymer, the mean diameter of said microbeads being within the range of from 1-90 microns, in which an organic phase, comprising the methacrylic monomer or mixture of such monomers, a cross-linking agent and the pharmaceutically active ingredient is emulsified in an aqueous phase containing one or more salt(s) under agitation with a rotary stirrer running at a speed of at least 1000 r.p.m., a polymerisation catalyst is then added to the emulsion thus formed, the polymerisation is allowed to proceed, and the microbeads thus formed are recovered. The methacrylic polymer is suitably a copolymer of methacrylic acid and/or methyl acrylate and/or butyl acrylate. The active ingredient is suitably one of the following:- hormones and contraceptive agents, corticosteroids, antibiotics, antiparasitic agents and cytostatic or cytolitic agents. The exemplified active ingredients are testosterone acetate, chlorpromazine and 13#-ethyl-17α ethynyl- 17#-hydroxy-gona-4, 9, 11-triene-3-one. In the preparation, the salt(s) present in the aqueous phase are preferably alkali metal, alkaline earth metal or ammonium sulphate, the crosslinking agent may be divinyl benzene, and the catalyst is suitably 2, 2'-azo bis-isobutyronitrile.
