Abstract
1478575 Apparatus for enumerating microorganisms CANADIAN PATENTS & DEVELOPMENT Ltd 25 July 1975 [14 Aug 1974] 31176/75 Heading C6F An apparatus for enumerating micro-organisms consists essentially of a membrane filter element 1, Figs.1 and 2, for retaining on a surface thereof the micro-organisms present in a fluid sample filtered thereby, which surface carries an imprinted pattern 2 of a barrier material defining a plurality of ordered, microbial colony-isolating cells 3. The cells are of an area smaller than the area normally expected for the microbial colonies 4 so that these tend to grow perpendicular to the surface of the membrane, increasing in thickness and thus enhancing the optical contrast between colony and filter. Overlapping and confluent growth of the colonies is prevented. In general the area of each cell is not greater than 25 mm<SP>2</SP> and the width of the barrier material is greater than 0À025 mm. Barrier materials: paraffin wax, petrolatum, silicone wax or oil, epoxy resin, polytetrafluoroethylene or polystyrene solutions, rubber, acrylic resin and beeswax. The gridded membrane 21, Figs.5 and 6, may be mounted in a stackable container 25 upon an absorbent pad 29 for retaining nutrient solution. In other embodiments the cells are arranged on the membrane in the form of a spiral and the barrier material defines circular cells, Figs.3 and 4 respectively (not shown). An Example is directed to enumerating colonies of Serratia marcescens using cellulose ester membranes imprinted with a square grid pattern of paraffin wax with lines 0À125 mm wide and 0À5 mm apart.
